Detection and pathogenic characteristics of two new Escherichia coli subtypes producing Shiga toxin 2k and 2l in sheep feces in a district of Beijing

Abstract:
This study was aimed at analyzing the status and pathogenic characteristics of Shiga toxin-producing Escherichia coli(STEC)carried by sheep in a district of Beijing,to support STEC infection risk assessment,traceability investigation,and scientific prevention and control.Fecal samples from sheep in one district of Beijing were collected,and STEC strains were iso-lated.The isolates were tested for 15 antibiotic resistance types,and whole genomes were sequenced.Multi-locus sequence typ-ing,serotyping,and Shiga toxin subtyping were performed on the basis of the whole genome sequences of the isolates.A phylogenetic tree was constructed on the basis of whole genome single nucleotide polymorphisms(cgSNPs).5 STEC strains were isolated from 129 sheep fecal samples,and the i-solation rate was 3.88%(5/129).The 5 strains were classified into four serotypes and four ST types,and the stx subtypes 2k,2l,and 1a were found.All 5 STEC strains were sensitive to the tested antibiotics,including ampicillin.Whole-genome phylogenetic analysis revealed that the stx2k-STEC strains were distantly related to the stx2k-STEC strains isolated from patients with diarrhea in the local area,whereas the stx2l sub-type strain was closely related to the food-derived stx2l-STEC strain isolated in this city.The characteristics of the sheep-de-rived STEC strains in this region were diverse,and the new subtypes stx2k and stx2l were detected.The risk of human infec-tion by these strains requires further monitoring and assessment.
