The effects of miR-210 on osteogenic differentiation of periodontal ligament stem cells via regulating Twist1 in inflammatory microenvironment

Abstract:
Objective:To investigate the effects of microRNA-210(miR-210)on osteogenic differentiation of periodontal ligament stem cells(PDLSCs)via regulating Twist1 in inflammatory microenvironment.Methods:PDLSCs were cultured and divided into control group,inflammation group,miR-negative control(NC)group,inflammation+miR-NC group,inflammation+miR-210 group,miR-NC+NC plasmid group,inflammation+miR-210+NC plasmid group and inflammation+miR-210+Twist1 plasmid group.The in-flammation microenvironment was established by 10 μg/L TNF-α treatment in the inflammation groups,and the miR sequences or plasmids were transfected by liposomes.The expression levels of miR-210 and Twist1 were detected after 24 h treatment.A590 levels of alizarin red staining and expression levels of Runt-related transcription factor 2(Runx2)and osteocalcin(OCN)detection were performed after 2 weeks of osteogenic differentiation induction.Results:The expression level of miR-210 in inflammation group was lower and the expression level of Twist1 was higher than that in control group(P<0.05).The expression level of miR-210 and A590,Runx2 and OCN after osteogenic induction in the inflammation+miR-NC group was lower than that in the miR-NC group,and the expression level of Twist1 was higher than that in the miR-NC group(P<0.05).The expression level of miR-210,A590,Runx2 and OCN after osteogenic induction in the inflammation+miR-210 group was higher than those in inflammation+miR-NC group,and the expression level of Twist1 was lower than that in inflammation+miR-NC group(P<0.05).The expression level of Twist1 and the flu-orescence value of wild-type Twist1 reporter gene in miR-210 group was lower than those in miR-NC group(P<0.05).The expres-sion level of Twist1 in the inflammation+miR-210+Twist1 plasmid group was higher than that in the inflammation+miR-210+NC plas-mid group,and the level of A590,Runx2 and OCN after osteogenesis induction were lower than that in the inflammation+miR-210+NC plasmid group(P<0.05).Conclusion:The miR-210 expression decreace followed by Twist1 expression increace in inflammation microenvironment inhibites the osteogenic differentiation of PDLSCs.
