Research progress of NLRP3 natural product inhibitors in treatment of nonalcoholic fatty liver disease

- Abstract: Nonalcoholic fatty liver disease(NAFLD)is the most prevalent chronic liver disease characterized by abnormal accu-mulation of fat in liver and an inflammatory response,with a complex pathogenesis.Overactivation of the pyrin-domain-con-taining protein 3(nucleotide-binding oligomerization domain,leucine-rich repeat and pyrin domain-containing 3,NLRP3)in-flammasome triggers the secretion of interleukins,induces pyrop-tosis,and promotes the release of a large number of pro-inflam-matory proteins,all contributing to the development of NAFLD.A large body of evidence suggests that plant-derived NLRP3 in-hibitors are safe and effective in improving the occurrence and progression of NAFLD.The purpose of this article is to summa-rize the molecular structure and mechanism of action of 12 plant-derived NLRP3 natural product inhibitors,which alleviate the de-velopment of NAFLD by acting on NLRP3 inflammasomes in dif-ferent NAFLD models.These studies confirm that inhibition of NLRP3 inflammasome plays a key role in the improvement of NAFLD,providing a reference for further research on the correla-tion between plant-derived compound treatment and NLRP3 in-flammasome and NAFLD.
