Effects of MiR-198 on Proliferation and Migration of Hepatocellular Carcinoma Cell Lines

Abstract:
Objective To investigate the effects of miR-198 on proliferation and migration of hepatocellular carcinoma cell lines.Methods RT-qPCR was used to detect the expression of miR-198 in different hepatocellular carcinoma cell lines,and the prolifera-tion,migration and tumorigenic ability of miR-198 on Huh7 cell lines were identified by in cell experiments and in animal experiments.Results RT-qPCR test results showed that compared with Chang liver and LO2 normal liver cell lines,the expression levels of miR-198 in HepG2,Huh7,Hep3B and MHCC97H hepatocellular carcinoma cell lines were decreased,especially in Huh7 cells.Flow cytome-try was used to confirm that miR-198 promoted the apoptosis of Huh7 cells.CCK-8 and soft agar clone formation experiment showed that compared with the control group,the proliferation of Huh7 hepatocellular carcinoma cell line after transfection with miR-198mimics was significantly reduced.The results of scratch test and Transwell test showed that miR-198 could inhibit the migration ability of Huh7 hepatocellular carcinoma cell line.The tumor formation experiment in nude mice proved that the tumor-forming ability of Huh7 hepato-cellular carcinoma cell line transfected with miR-198 was obviously inhibited.Conclusion The expression level of miR-198 in HepG2,Huh7,Hep3B and MHCC97H hepatocellular carcinoma cell lines was significantly decreased,and the lowest expression was found in Huh7 hepatocellular carcinoma cell line.MiR-198 can inhibit the proliferation and migration ability of Huh7 hepatocellular car-cinoma cell line,and reduce its tumor-forming ability.
