Diagnostic Value of Cytokines Secreted by Neutrophil in Ulcerative Colitis

Abstract:
Background:Immunity plays a crucial role in the development of ulcerative colitis(UC).However,the specific changes in key cytokines and immune cells remain unclear.Aims:To explore the diagnostic value of cytokines secreted by neutrophils in UC.Methods:The GSE231993 dataset was selected.Single-cell sequencing was used to detect the changes in the proportion of immune cells in UC.The relative contributions of corresponding receptor-ligand pairs were calculated,and the most significantly changed cytokines were screened out.A total of 75 patients diagnosed with UC at Beilun District People's Hospital from March 2021 to July 2023 were included.The levels of tumor necrosis factor(TNF)-α,interleukin(IL)-6,IL-8,IL-1β,and IL-13 were detected by ELISA.A Logistic regression diagnostic model was constructed based on these five cytokines,and its diagnostic efficacy for UC was evaluated.Results:In the GSE231993 dataset,the proportion of neutrophils specifically increased in UC tissues.According to the relative contributions of receptor-ligand pairs in neutrophils,the level changes of TNF-α,IL-6,IL-8,IL-1β,and IL-13 were the most significant.The levels of TNF-α,IL-6,IL-8,IL-1β,and IL-13 in UC patients were significantly higher than those in the control group.The area under the curve(AUC)of the diagnostic model constructed based on these five cytokines for diagnosing UC was 0.856,and it was 0.874 in the GSE107499 dataset.Conclusions:The changes in neutrophils are the most significant during the progression of UC.The diagnostic model based on TNF-α,IL-6,IL-8,IL-1β,and IL-13 can effectively determine the occurrence of UC.
