Effects of anisodine hydrobromide on myocardial oxidative stress injury in rats with exercise-induced fatigue

Abstract:
Objective To investigate the effect of anisodine hydrobromide on myocardial oxidative stress injury in rats with exercise-induced fatigue.Methods The rats were divided into control group,model group and experimental-L,-M,-H groups.The rats in control group and model group were intragastrically infused with 0.9%NaCl.The rats in experimental-L,-M,-H groups were fed with 0.3,0.6 and 1.2 mg·kg-1·d-1 anisodine hydrobromide solution,respectively.The rats in five groups were all given the drug for four consecutive weeks.Except for control group,the rats in other four groups were subjected to weight-bearing exhaustive swimming exercise to establish the exercise-induced fatigue rat model 2 hours after drug administration,and the model was established for a total of four weeks.The levels of serum lactate dehydrogenase(LDH)was measured by micromethod,the apoptosis of cardiomyocytes was evaluated by TUNEL staining,the activity of superoxide dismutase(SOD)was detected by micromethod,and the expression of nuclear factor E2 related factor 2(Nrf2)protein was detected by Western blot.Results The serum LDH levels of control group,model group and experimental-L,-M,-H groups were(991.79±123.88),(2 983.06±281.28),(2 309.06±271.81),(1 856.17±214.16)and(1 380.29±189.81)U·L-1,respectively;the positive rates of myocardial TUNEL were(5.09±0.42)%,(48.43±5.57)%,(30.97±3.43)%,(21.57±2.31)%and(9.49±1.36)%,respectively;the SOD activities of myocardium were(102.06±8.96),(59.65±7.66),(73.25±7.60),(80.78±6.22)and(84.76±8.94)U·mg prot-1,respectively;the relative expression levels of nuclear Nrf2 protein were 1.00±0.08,0.31±0.03,0.60±0.05,0.89±0.06 and 1.21±0.09,respectively.The above indicators in the model group were compared with control group,and the above indicators in the experimental-L,-M,-H groups were compared with model group,and the differences were statistically significant(all P<0.05).Conclusion Anisodine hydrobromide can reduce the oxidative stress injury and apoptosis of cardiac muscle in rats with exercise-induced fatigue,and its mechanism is related to the activation of Nrf2-mediated antioxidant pathway.
