Effect of Erianin on inflammatory response in diabetes peripheral neuropathy rats by regulating TXNIP/NLRP3 signaling pathway

Abstract:
Objective To investigate the effect of Erianin on inflammation by regulating thioredoxin interaction protein/NOD-like receptor thermal protein domain associated protein 3(TXNIP/NLRP3)signaling pathway in rats with diabetes peripheral neuropathy(DPN).Methods SD rats were divided into normal control(NC)group,DPN group,Erianin 10 mg/kg(Eri 10)group,Erianin 40 mg/kg(Eri 40)group,Eri 40+pLVX-NC group,and Eri 40+TXNIP overexpression vector(Eri 40+pLVX-TXNIP)group,10 in each group.BG,IL-18,IL-1β,IL-10,reactive oxygen species(ROS)and nerve motor and sensory conduction velocity were detected in each group.HE was used to detect the histopathological changes of sciatic nerve.The mRNA and protein expressions of TXNIP,NLRP3 and Caspase-1 were detected by qRT-PCR and Western blot.Results Compared with NC group,BG,IL-18,IL-1β,ROS,mRNA and protein expressions of TXNIP,NLRP3 and Caspase-1 were increased(P<0.05),while IL-10,motor and sensory conduction velocity were decreased in DPN group(P<0.05).BG,IL-18,IL-1β,ROS,mRNA and protein expressions of TXNIP,NLRP3 and Caspase-1 were decreased successively(P<0.05),while IL-10,motor and sensory conduction velocity were increased successively in DPN,Eri 10 and Eri 40 groups(P<0.05).Compared with Eri 40 and Eri 40+pLVX-NC groups,BG,IL-18,IL-1β,ROS,the mRNA and protein expressions of TXNIP,NLRP3 and Caspase-1 were increased(P<0.05),while IL-10,motor and sensory conduction velocity were significantly decreased in Eri 40+pLVX-TXNIP group(P<0.05).Conclusions Erianin can regulate TXNIP/NLRP3 signaling pathway to reduce inflammatory response and protect sciatic nerve in rats with DPN.
