Effect of polydatin on myocardial injury in dilated cardiomyopathy rats

Abstract:
Objective To investigate the effect of polydatin(PD)on myocardial injury in rats with doxorubicin-induced dilated cardiomyopathy(DCM)based on the sonic hedgehog(Shh)/glioma-associated oncogene 1(Gli1)signaling pathway.Methods A total of 60 SD rats were divided into six groups using a randomization method:Con group,DCM group,PD low-dose group,PD medium-dose group,PD high-dose group,and Cyclopamine group,each group contained 10 rats.Except for the Con group,all other groups were induced with doxorubicin to establish DCM rat models.The cardiac function parameters of each group were measured,and pathological changes including myocardial tissue fibrosis and cardiomyocyte apoptosis were observed.Serum inflammatory factors,type Ⅰ and type Ⅲ collagen expression in myocardial tissues,as well as Shh/Gli1 signaling pathway protein expression in myocardial tissues of all groups were analyzed.Results Compared with Con group,rats in DCM group exhibited more severe histopathological damage and fibrosis in myocardial tissues.Significant increases were observed in left ventricular end-systolic diameter(LVESD),left ventricular end-diastolic diameter(LVEDD),myocardial cell apoptosis rate,as well as the average absorbance values of interleukin(IL)-1β,IL-18,type Ⅰ collagen,and type Ⅲ collagen(P<0.05).Left ventricular ejection fraction(LVEF),left ventricular fractional shortening(FS),Shh,and Gli1 expression levels decreased significantly(P<0.05).Compared with the DCM group,histopathological damage and fibrosis reduced in the PD low-dose,medium-dose,and high-dose groups.LVESD,LVEDD,myocardial cell apoptosis rate decreased,and average absorbance values of IL-1β,IL-18,type Ⅰ collagen,and type Ⅲ collagen reduced.LVEF,FS,Shh,and Gli1 expression levels increased significantly(P<0.05).Compared with PD high-dose group,pathological damage and fibrosis of myocardial tissue increased,while LVESD,LVEDD,cardiomyocyte apoptosis rate,IL-1β,IL-18,average absorbance of type Ⅰ collagen and type Ⅲcollagen increased,and expression of LVEF,FS,Shh and Gli1 Decreased in cyclopamine group(P<0.05).Conclusion PD may improve cardiomyopathy in DCM rats by activating Shh/Gli1 signaling pathway.
