Abstract:
Objective To analyze the correlation between the expression of fas-activated kinase domain-containing proteins(FASTKDs)and the prognosis of non-small cell lung cancer(NSCLC)and its predictive value.Methods By analyzing the somatic mutation in Catalog of Somatic Mutations in Cancer database,the effect of FASTKDs was analyzed.The mRNA expression level of FASTKDs in different pathological subtypes of lung cancer was analyzed retrospectively.Kaplan-Meier survival analysis was used to evaluate the predictive significance of FASTKDs specific expression in NSCLC samp les.Real-time quantitative polymerase chain reaction(RT-qPCR)and Western blot were used to verify the mRNA and protein expression of FASTKDs in human NSCLC cells in vitro.Results FASTKDs in lung adenocarcinoma(LUAD)were highly expressed(P<0.05),while only FASTKD1,FASTKD3,FASTKD4 and FASTKD5 in lung squamous cell carcinoma were highly expressed(P<0.05).In all patients with NSCLC and LUAD pathological subtypes,only the increased expression levels of FASTKD1 and FASTKD3 mRNA were significantly correlated with the higher overall survival(OS)(P<0.05),but the increased expression levels of FASTKD2 and FASTKD4 mRNA in NSCLC patients were correlated with the decreased OS(P<0.05).Compared with normal human bronchial epithelial cells,the mRNA and protein expression of FASTKD1-5 in human NSCLC cells increased significantly(P<0.05).Conclusion Different expression of FASTKDs is related to the prognosis of NSCLC patients and is a potential tumor marker of NSCLC.
