Exploring the mechanism of Tongxinluo treat ischemic stroke based on network pharmacology

Abstract:
Objective To predicting the potential mechanism of action in treating ischemic stroke(IS)via network pharmacology and molecular docking.Methods TCMSP database was used to obtain the drug active components and targets of caxin collaterals,the disease targets corresponding to IS were obtained through DisGeNET database and GeneCards database,and two potential pharmacological targets were obtained by Venny 2.1.Potential pharmacological targets were carried out enrichment analysis of gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)by using the DAVID database.AutoDock tool was used for molecular docking between key targets and active compounds.Results A total of 62 active in gredients,406 targets,148 potential targets for pharmacological effect were obtained.GO enrichment analysis yielded 1556 gene function information.The KEGG enrichment analysis identified 119 pathways in total,including lipid and atherosclerosis pathways,cancer signaling pathway,and phosphoinositide 3-kinase-akt(PI3K-Akt)signaling pathway.Molecular docking results indicated that prostigmasterol and baicalein bind strongly to core targets including interleukin(IL)-6,tumor necrosis factor(TNF),et al.Conclusion Tongxinluo capsule may act on IL-6,TNF,etc.And lipid and atherosclerosis pathway,cancer signaling pathway and PI3K-Akt signaling pathway are regulated to play anti-inflammatory,promote cell regeneration,anti-apoptotic mechanism and other mechanisms to treat IS.
