Abstract:
Adenovirus(Ad) vector vaccines use the modified Ad as a delivery tool to deliver the gene encoding the target antigen to the host cell, inducing humoral and cellular immune response, which have good characteristics such as stable expression of heterologous genes, strong immunogenicity and low pathogenicity, and can be produced on a large scale. The SARS-CoV-2 vaccine prepared by this platform not only curbs the spread of the epidemic in a short period of time, but also provides a technical platform and valuable experience for dealing with emerging infectious diseases in the future. It has shown significant potential in the prevention and control of infectious diseases and has become an important direction of current vaccine research. In this paper, the latest clinical application progress of Ad vector vaccines, the key challenges in the process of clinical transformation and the molecular mechanism of inducing host immune response are reviewed, in order to provide reference for related research.
