Abstract:
Fluorescence quantitative PCR method was used to detect the relative expression levels of carboxylesterase gene(CPIJ002786-RA)in sensitive and resistant strains of Culex pipiens pallens of both laboratory population(LC50=0.03 mg/L and 2.00 mg/L by larval bioassay),and sensitive and resistant strains(differentiated by WHO susceptibility tube bioassay)in 4 field populations(Shanghe/Gudao/Huimin of Shandong Province,Jiangxinzhou of Jiangsu Province).The expression level of carboxylesterase(CPIJ002786-RA)was 2.77-fold higher in resistant strains than in susceptible strains of laboratory populations(P<0.01),and were 3.65-,5.67-and 2.75-fold higher in three resistant strains of field populations(Shanghe,Gudao and Huimin)than in susceptible strains(P<0.05).However,the expression level of carboxylesterase was no significant difference between resistant and sensitive strains(P>0.05)in Jiangxinzhou field population.The over expression of carboxylesterase gene(CPIJ002786-RA)in Cx.pipiens pallens may be closely related to the deltamethrin resistance,which means a potential application value as an index gene in field resistance detection.
