Comparison of two detection methods for the samples from the kindergarten by comparative tests of positive Escherichia coli

Abstract:
Objective To compare the advantages and disadvantages of 2 Escherichia coli culture methods , so as to establish and improve disinfection and control in all the kindergartens of the area .Methods The standard strain of Escherichia coli [ ATCC25922] was collected in October and November of 2012, then, was diluted 10 times and each time with a proper ratio .Finally, 300 samples of solution were obtained with proper dilution levels of 10 -7 , 10 -8 and 10 -9 .Then, 1ml of the above positive control su-pernatant was inoculated in the unblended lactose bile salt fermentation tubes (3 tubes) by using method 1 for later detection .At the same time, detection was made in the remaining supernatant by using method 2, then, the test solution was put in the lactose bile salt fermentation culture solution , and was inoculated at a temperature of ( 36 ±1 )℃ for 24 hours for parallel comparison experiments . Results Positive detection rate of method 1 was 229, while that of method 2 was 249.Byχ2 test, statistical difference could be noted in the 2 Escherichia coli culture methods (P<0.05).Conclusion Method 2 was simpler and more economic and seemed to have a higher detection rate , when compared with that of method 1.It was recommended that test method 2 could replace method 1 in the Esch-erichia coli test standard DB31/8-2004.
