Abstract:
Objective To determine the level of prostaglandin E2 (PGE2 ) and leukotriene B4 (LTB4 ) after treatment with a unique anti-inflammatory herbal preparation (Zyflamend) in 7,12-dimethylbenz(a) anthracene (DMBA)-induced oral precancerous lesions in golden Syrian hamsters.Methods The hamsters were divided into five groups.The Syrian hamsters were applied with 0.5% DMBA solution topically to the left cheek pouch three times per week for three consecutive weeks.The negative control group was not treated.After the last treatment by DMBA,the 0.5% DMBA treated hamsters were divided into four groups at random.The positive group was not treated,the other groups received low,middle and high doses of Zyflamend respectively three times within the following week.The animals were sacrificed 4 weeks after the start of the experiment.They were injected with 5-bromo-2'-deoxyuridine (BrdU) 50 mg/kg before they were killed.A piece of tissue was snap-frozen in liquid nitrogen for liquid chromatographic tandem mass spectrometry analysis.Results The level of PGE2 was increased from (0.377 ±0.290) μg/g to (0.523 ±0.286),(0.488 ±0.147) and (0.774 ±0.314) μg/g.There was no significant difference between positive group and groups with low,middle doses of Zyflamend ( P > 0.05 ).The level of PGE2 was significantly increased in the treatment group of high dose of Zyflamend.Low,middle and high doses of Zyflamend significantly decreased the LTB4 level from (0.135 ±0.046)to (0.094 ±0.066),( 0.096 ± 0.077 ) and ( 0.067 ± 0.012 ) μg/g respectively ( P < 0.05 ).Conclusions Zyflamend have inhibitory effect on oral carcinogenesis.
