Study on ‘Cot toai bo’ (Drynaria bonii Rhizome) product was processed from fresh stem and root of Drynaria bonii Christ., collected at Lang Son from July 2003 to September 2003. Results: chemical analysis showed that Cot toai bo contains flavonoids, steroid saponins, sterols, tannins, deoxidized sugar. Total flavonoid in stem and root accounted for 0.60±0.03% and total saponin was 3.71±0.06%. After processing by traditional method, total flavonoids and saponins in processed materials reduced in comparison with raw material. Compare 2 processing methods: in rice alcohol processed material, whose chemical constituents were reduced very little but in fried material, whose chemical constituents were reduced more than raw material. Rice alcohol processed Cot toai bo had analgesic and anti-inflammatory effects better than raw material and this processing was a more appropriate method than fried method

The acute toxicity and analgesic, sedative effects of Drynaria fortunei - Polypodiaceae were studied on experimental mice and determined LD50 by Litchfield-Wilcoxon method. OBJECTIVETo evaluate the effects of Drynaria fortunei naringin on the total protein content and ultra-structure of human periodontal ligament cells (hPDLCs).

METHODSThrough enzyme digestion combined tissue-culture method, primarily culture and identify the human periodontal ligament cells. Coomassie brilliant blue staining was used to detect the total protein content of hPDLCs with the effects of difference concentration of Drynaria fortunei naringin at difference times. Transmission electron microscope was used to study the ultra-structure of hPDLCs with the effects of Drynaria fortunei naringin.

RESULTSIn vitro, the addition of Drynaria fortunei naringin at dose of 100, 10, 1, 0.1 mg x L(-1) in cultures resulted in an increase of total protein content at the 3rd, 5th, 7th day, but the maximum response was obtained with 1 mg x L(-1) Drynaria fortunei naringin. There were more rough endoplasmic reticulums, mitochodrias and ribosomes in the experimental group than in the control.

CONCLUSIONDrynaria fortunei naringin may stimulate the protein synthesis and metabolism of hPDLCs.

One of the traditional Korean medicine, Drynaria fortunei (D. fortunei) is one of candidates known to be effective for the treatment of inflammation, hyperlipemia, arteriosclerosis, rheumatism, and gynecological diseases such as osteoporosis and bone resorption. The present study investigated the antimicrobial activity of methanol (MeOH) extract and n-butanol (n-BuOH), chloroform (CHCl3), and ethyl acetate (EtOAc) fractions of D. fortunei against oral bacteria. The n-BuOH and CHCl3 fractions (MICs, 0.0078 to 0.3125 mg/ml; MBCs, 0.019 to 0.625 mg/ml) were demonstrated as strong antibacterial activity than the MeOH extract and EtOAc fraction. The combination effects of n-BuOH fraction with ampicillin or gentamicin were synergistic against some oral bacteria. We suggest that D. fortunei could be employed as a natural antibacterial agent in oral care products.

OBJECTIVETo evaluate the biological effects of Drynaria fortunei naringin on the proliferation and alkaline phosphatase (ALP) activity of human periodontal ligament cells (hPDLC).

METHODShPDLC were primarily cultured and identified in vitro through enzyme digestion combined tissue culture method. 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method was used to observe the proliferation of hPDLC with the effects of different concentrations of Drynaria fortunei naringin at difference times. The method recommended by International Federation of Clinical Chemistry and Laboratory Medicine (IFCC) was adopted to investigate the effect of Drynaria fortune naringin on the alkaline phosphatase (ALP) activity of hPDLC. At the same time, we used bright blue method to detect the contents of protein in each sample. Then ALP activity in per milligram protein was accounted.

RESULTSSignificant proliferative promotion to hPDLC by Drynaria fortunei naringin could be observed at the dose of 10, 1, 0.1 mg x L(-1). Significant ALP activity of hPDLC promotion of Drynaria fortune naringin could be observed at the dose of 100, 10, 1, 0.1 mg x L(-1) and the dose of 1 mg x L(-1) Drynaria fortune naringin had greatest promotion on ALP activity of hPDLC.

CONCLUSIONDrynaria fortune naringin might significantly promote the proliferation and increase the ALP activity of hPDLC.

OBJECTIVETo provide practical method for microscopic authentication of traditional Chinese medicine Gusuibu and its adulterants.

METHODBy means of light microscope, scanning electron microscopy and tissue section techniques, the morphology, the size of the rhizome scales and their bearing position in the original plants of Gusuibu and its adulterants, i. e. Drynaria roosii, D. delavayi, D. quercifolia and Pseudodrynaria coronans were analyzed.

RESULTThere were significant differences between scales length of D. roosii, D. delavayi and P. coronans, while there was no significant difference between that of D. roosii and D. quercifolia. The scale teeth of D. delavayi were usually curved, bifid and uneven distributed at the scale fringe, which was different from that of the other three species. The base of the scales sinks in epidermis in D. roosii, D. quercifolia, and P. coronans, while it bore at the raised part of epidermis in D. delavayi.

CONCLUSION[corrected] Morphology, size and bearing position of the rhizome scales have significant differences in the several species. Therefore, these characteristics can be applied to the identification of Gusuibu and its adulterants.

Drynariae Rhizoma is warm in nature and bitter in taste, mainly acting on liver and kidney systems. It is a common Chinese herbal medicine for the treatment of fracture and bone injury. The chemical compositions of Drynariae Rhizoma mainly include flavonoids, triterpenoids, phenylpropanoids and lignans. At present, modern pharmacological and clinical studies have shown that Drynariae Rhizoma has the effects of anti osteoporosis, promoting fracture healing, kidney protection, anti-inflammatory, promoting tooth growth, preventing and treating aminoglycoside ototoxicity and lowering blood lipid. In addition, the toxicity evaluation experiment of Drynariae Rhizoma has also shown that it has no obvious toxic and side effects. Naringin is a kind of dihydroflavone in Drynariae Rhizoma. Many studies have shown that naringin and other total flavonoids play an important role in anti-osteoporosis, promoting fracture healing, anti-inflammation, promoting tooth growth and lowering blood lipid. In this study, the research progresses on chemical consti-tuents and pharmacological activities of Drynariae Rhizoma in recent years were reviewed, and some mechanisms of action were summarized, to provide references for the further research and development of Drynariae Rhizoma.

OBJECTIVETo study the chemical components of Drynaria fortunei.

METHODThe herbal material was extracted with 75% ethanol. The chemical components were isolated by silica gel and sephadex LH-20 column chromatography. The structures were identified on the basis of chemical and spectroscopic data.

RESULTFour compounds were isolated, three of which were identified as (-)-epiafzelechin-3-O-beta-D-allopyranoside, (-)-epiafzelechin and beta-sitosterol.

CONCLUSION(-)-epiafzelechin-3-O-beta-D-allopyranoside, (-)-epiafzelechin were obtained from D. fortunei for the first time.

OBJECTIVETo evaluate influence of assemble flavone of rhizome drynaria (AFDR) on the value of the blood serum alkalinity phosphatase (ALP), calcium (Ca), phosphorus (P), creatinine (Cr) and glutamic-pyruvic transaminase (GPT) in rats model with skull defects.

METHODSSixty SD male rats with age of 6-month were feeded for a week and then were randomly divided into control group and AFDR group, with 30 rats in each group. Left and right skull of rats were perforated with electromotive drill and the model of skull defects was made. Injectable bone regeneration vomposite (IBRC) was implanted right skull defects. The rats of control group and AFDR group were respectively lavaged with AFDR and deionized water at the first day after operation. The rats were respectively killed at the 2nd,4th and 8th week and the blood serum ALP, Ca, P, Cr, GPT were detected and analyzed by statistics.

RESULTSAt the 2nd week after operation, blood serum ALP in AFDR group was higher than that of control group. At the 4th week after operation, blood serum Ca, P, and calcium-phosphorus product in AFDR group was higher than that of control group; there was no significant difference in GPT between two groups. At the 8th week after operation, blood serum Cr in AFDR group was lower than that of control group.

CONCLUSIONWhen AFDR is used in the repairing of bone defect for 2-4 weeks, it may affect the level of ALP, Ca, P, and without toxicity to liver and kindey.

OBJECTIVETo study the therapeutic effects of Rhizoma Drynariae and estrogen on osteoporosis in ovariectomized (OVX) rats.

METHODSFifty-five female rats were randomly divided into 5 groups, the normal control (normal),the sham operated (sham), the model, the estrogen, and the Rhizoma drynariae (RD) groups; ovariectomized rats were used as postmenopausal osteoporosis model. The changes of morphology and dynamic parameters in different groups were determined by bone histomorphometry.

RESULTSCompared with the model group, the trabecular volume (TBV/TTV) , trabecular thickness (MTPT) and density (MTPD) in the other four groups were significantly increased, while the trabecular template spacing (MTPS) and the ratio of trabecular surface to trabecular volume (TBS/TBV) significantly decreased (P <0. 05); and the osteoid surface (TOS), single label surface [Sfract (s) ] ,double label surface [Sfract (d) ] and bone formation rate (Svf) also decreased,while osteoid maturation period (OMP) increased in the latter four groups. No significant difference of cortical width (MCW) was found between these 5 groups. Compared with the normal and sham groups, TOS, Sfract ( s) , Sfract ( d) , Svf in the estrogen and RD groups increased significantly, while OMP decreased; no significant difference was found in other parameters.

CONCLUSIONRhizoma Drynariae has the similar effect with estrogen in maintaining normal trabecular structure and connection by inhibiting the increased bone turnover of postmenopausal osteoporosis.
