1.Acute liver injury and expression of P-selectin in serum of mice induced by agkistrodon halys pallas venom

Objective To explore the relationship between pathological changes of liver,serum ALT,expression of P-selectin in serum of mice and different dose of venom,the poisoning time induced by Agkistrodon Halys Pallas venom.Methods Using two -factor factorial design,72 male Kunming mice of clean grade were randomly ( random number) divided into 9 groups (n =8).As per the different doses of snake venom injected and different lengths of time after venom injected,the 9 groups were 0-dose (0.0 mg/kg) 3 hours after saline given group,0-dose 8 hours after saline given group,0-dose 24 hours after saline given group,low-dose (0.5 mg/kg) 3 hours after venom given group,low-dose 8 hours after venom given group,low-dose 24 hours after venom given,high-dose ( 1.0 mg/kg) 3 hours after venom given group,high-dose 8 hours after venom given group and high-dose 24 hours after venom given group.The pathological change of liver tissue was determined,and ALT and expression of P-selectin in serum were detected.Results In the venom given groups,the liver histopathologic scores,serum ALT value and expression of Pselectin in serum were significantly higher than those in control group ( P < 0.05 ),and those in high-dose group were higher than those in low-dose group (P < 0.05 ),and those in 8h groups were higher than those in 3h groups (P<0.05),and those in 24 h groups were higher than those in 8 h groups (P <0.05).As the dose of venom increased and the length of time after venom given extended,the liver histopathologic scores,serum ALT and expression of P-selectin in serum were higher.A positive correlation between Pselectin expression and liver histopathologic scores was found after correlation analysis ( r =0.98,P <0.05 ).Conclusions Agkistrodon snake venom can cause acute liver injury with elevated ALT and Pselectin in mice.As higher dose of venom given and longer length of time after venom given extended,more serum ALT and P-selectin are produced.Between acute liver injury and high expression of P-selectin exists a correlation,suggesting overexpression of P-selectin is one of the mechanisms of acute liver injury induced by Agkistrodon Halys Pallas venom.
2.Preliminary development of warning scale of suicide ideation of inpatients with somatic disease

Objective To form a concise and useful warning scale of suicide ideation of inpatients with somatic disease.Methods 2 677 inpatients who were more than 15 years old were randomly selected in a comprehensive hospital during the period of 2010.11-2013.02.Self-rating Depression Scale (SDS),Self-rating Anxiety Scale(SAS) and self-designed questionnaires which included general information,suicide risk factors and somatic disease were applied.Chi square test,partial correlation analysis,t test,correlation analysis and ROC curve analysis were used for statistical analysis.Results 1.The warning scale of suicide ideation which contained 18 items was formed.2.The Cronbach's Alpha reliability of the warning scale was 0.84 and its split-half reliability was 0.60.Criterion-related validity studies showed that the scores of the 18 items were positive correlated with SDS and SAS(r SDS =0.50,r SAS =0.54,P<0.01).This scale also had good structure validity.3.The ROC curve concluded that the area under curve was 0.83,the sensitivity was 68.40% and the specificity was 79.20%.Positive and negative predictive values were 21.22% and 96.83% respectively.Conclusion The reliability and validity of the warning scale entirely accorded with psychometric demands.It also has good specificity and negative predictive value,so it is suitable for clinical promotion.
3.Expression and specific cell response of autoantigen Dsg3 in pemphigus vulgaris

Objective:In view of analysis of role of autoantigen Dsg3 in specific T cell response to understand the molecular mechanism of autoimmune diseases.Methods:Based on the sequence analysis of autoantigen Dsg3,the five fractions cDNAs of Dsg3 were cloned and Dsg3-GST fusion proteins induced by IPTG in E.coli.JM109 were purified,then mixed-cultured with T cell from PV patients.The T cell proliferation response were analyzed by MTT.Results:Dsg3 E1,E2 and E4,E5 stimulated T cell from PV patients,not controls.Conclusion:Dsg3 E1,E2 and E4,E5 contained the epitopes relevant to T-B cell interaction,which play an important role in the pathogenesis of pemphigus vulgaris.
4.RECOMBINANT EXPRESSION AND EPITOPE ANALYSIS OF AUTOANTIGEN Dsg3 IN PEMPHIGUS VULGARIS

The present study was to analyze the epitopes of autoantigen Dsg3 in pemphigus vulgaris (PV), and to elucidate the molecular mechanisms underlying autoimmune diseases. Based on the sequence analysis of autoantigen Dsg3 in Genbank five fragments of Dsg 3 cDNA (E1, E2, E3, E4, and E5) were cloned using RT PCR. Then PCR products were inserted into the expression vector pGEX 2T, and transformed into JM109 strain of E. coli. Dsg3 recombinant proteins were purified from the cell lysates. The epitopes of Dsg3 in PV patients were analyzed with five Dsg3 recombinant proteins by Western blot. The results showed that Dsg3 E1, E2 and E4 recombinant proteins reacted with PV patients′ sera, but not with control or normal sera. The dominant epitopes of Dsg3 in PV patients were different. The above data indicated that the epitopes of Dsg3 were located in E1, E2, and E4, which might play an important role in the pathogenesis of pemphigus vulgaris.
5.Diagnosis and treatment of acute peripheral artery injury

Objectives To study the diagnosis, treatment and prognosis of acute peripheral artery injury, and ways to improve the treatment. Methods Clinical data of 211 cases of acute peripheral arterial injuries admitted between January 1988 and June 2002 in Anzhen hospital were analyzed retrospectively. Of the 211 cases,73 cases were with blunt injuries, 99 penetrating injuries, 37 iatrogenic injuries, 2 gunshot injuries. Management procedures included arterial primary or patch repair in 28 cases, ligation alone in 17, end-to-end anastomosis in 41, interposition reverse saphenous vein graft in 37, interposition prosthetic graft in 29, thrombectomy in 14, excision of pseudoanurysm in 36, interventional thrombolytise in 2, ultrosonic-guided packing in 2, conservative management in 2. 6.Correlation between traditional Chinese medicine syndromes in primary immunoglobulin A nephropathy and A267G in 5'-untranslated region within exonal of megsin gene

OBJECTIVE: To observe the correlation between traditional Chinese medicine (TCM) syndromes ("deficiency of qi and yin" and "deficiency of liver yin and kidney yin") and A267G in 5'-untranslated region within exonal of megsin gene, and to search the substantial genetic basis for micro-differentiation of TCM syndromes in primary immunoglobulin A nephropathy (IgAN). METHODS: A total of 120 IgAN cases meeting the diagnostic criteria were enrolled. The sequence of single nucleotide polymorphism (SNP) of A267G in 5'-untranslated region within exonal of megsin gene was tested. The correlation between SNP and TCM syndromes was observed. RESULTS: There were 83 cases carrying GG genotype, 34 cases carrying GA genotype and 3 cases carrying AA genotype in 120 cases of primary IgAN. There was a high proportion of "deficiency of liver yin and kidney yin" in IgAN cases with AA and GA genotypes, and a high proportion of "deficiency of qi and yin" in IgAN cases with GG genotype (P<0.01). Odds ratio in TCM syndrome distribution between GG genotype and GA plus AA genotype was 9.800, and 95% confidence interval was 3.969-24.199. The discrepancy also resided in IgAN patients with different genders and ages. CONCLUSION: A267G in 5'-untranslated region within exonal of the megsin gene may be one of the substantial genetic basis for differentiating "deficiency of liver yin and kidney yin" syndrome and "deficiency of qi and yin" syndrome in primary IgAN.
7.Experience in the treatment of arterial embolism of the extremities, report of 536 cases

Objective To evaluate the treatment for acute embolism in the extremities and define the primary factors affecting the prognosis. Methods From December 1984 to December 2006,536 patients with acute embolism of extremities were treated. Embolectomy with Forgarty catheters were performed in 507 cases, including salvage surgery in 34 cases and conservative therapy in 29 cases. Results The cure rate, mortality and amputation rate were 76.68%,9.51% and 10.26% respectively. Embolism recurred in 11.7% cases. Cardiopulmonary (27.5% ) and renal failure ( 25.5% ) were main causes of death during perioperative period. No bleeding nor other major complications occurred in 221 patients with atrial fibrillation who routinely received wafarine under proper monitoring. About 25.6% patients underwent heart valves surgery during hospitalization or within one year after discharge. Conclusions Patients suffering from acute embolism of the extremities should receive combination treatment. The main factors affecting the prognosis include time and degree of ischemia, and ischemia-reperfusion injury. Anticoagulant and etiologic treatment should be adopted in those with cardiogenic embolus and atrial fibrillation.
8.miR-146a regulates monocyte hyper-reactivity to LPS and Poly I : C in patients with primary biliary cirrhosis

Objective To investigate whether the hyper-reactivity of monocytes from the patients with primary biliary cirrhosis (PBC) to LPS and Poly I ∶ C was associated with miR-146a.Methods Peripheral blood mononuclear cells from PBC patients and healthy controls were stimulated with 10 μg/ml of LPS or Poly I ∶ C.The levels of IL-1 β,IL-6,TNF-α and IFN-α in the culture medium were detected by ELISA.The relative expressions of miR-146a,miR-21,let-7e,miR-155 and miR-125b were detected by RT-PCR.The regulatory role of miR-146a on the production of inflammatory factors in LPS or Poly I ∶ C stimulated THP-1 cells was studied by gain-and-loss function assay.Results The up-regulation of miR146a,which was induced by both LPS or Poly I ∶ C,was impaired in the monocytes from PBC patients.The production of LPS or Poly I ∶ C induced inflammatory factor,could be enhanced by miR-146a down-regulation.Conclusion The hyper-reactivity of monocytes from PBC patients to LPS and Poly I ∶ C was associated with miR-146a.
9.Clinical value of serum tumor markers SCC and TSGF in cervix carcinoma diagnose

Objective To explore the clinical value of squamous cell carcinoma associated antigen (SCC) and tumor specific growth factor (TSGF) in cervical carcinoma. diagnose. Methods The serum samples of SCC and TSGF were collected from 40 health volunteers, 56 cervical intraepithelial neoplasia (CIN) patients and 101 squamous cell cervical cancer patients. SCC and TSGF amount were measured by MEIA and biochemistry olorimetry method respectively, and were compared. Results The mean level of serum SCC and TSGF in the cervical cancer group was (6.95 ± 1.23) ng/mL and (81.0 ± 16.3)U/mL respectively, significantly higher than that in the CIN group and control group. The sensitivity and accuracy of combining SCC with TSGF in diagnosing cervical carcinoma were 91.1%and 87.4% respectively, both higher than allying SCC or TSGF alone. Conclusions SCC and TSGF levels are helpful in the diagnosis of cervix carcinoma. The combined determination can improve the sensitivity and accuracy of cervical carcinoma diagnose.
10.Simulation of sparse views CT image iterative reconstruction based on defective projection data

Objective To reconstruct sparse views CT image based on defective projection data.Methods The position of bad bins in detector determined whether the linear interpolation was applied to the defective projection data.Moreover,reconstruction of air pixels in CT image was achieved rapidly and accurately.Results he experimental results showed that the proposed method could solve the problem from classical ART-TV method that the robustness was unstable due to the different positions of bad bin in CT detector.Conclusion Compared to analytical reconstruction methods,iterative methods can solve the reconstruction problems in this modality so that the radiologist is facilitated to perform image processing and quantitative analysis.
