1.The characteristics of IL-22 in Helicobacter pylori infection

Objective To investigate the characteristics of IL-22 in Helicobacter pylori (H.pylori)infection.Methods Thirty H.pyloripositive and fifteen H.pylori negative gastric biopsy specimens were enrolled,IL-22 mRNA expression was detected by real-time PCR and the protein level of IL-22 was determined by ELISA in gastric tissue.The H.pyloriand cell coculture system was established and IL-22 expression was measured by real-time PCR to investigate the main source of IL-22 in gastric tissue.The IL-22-producing T cell was examined by FCM in the gastric mucosa tissue.Results Gastric mucosal IL-22 mRNA and protein levels were significantly higher in H.pylori-positive patients than uninfected patients (P<0.05).A H.pyloriand cell coculture system was established successfully and gastric epithelial cell and T cell were the main source of IL-22 in gastric tissue.IL-22 was produced by CD4+ and CD8+ T cells and these T cells were increased in H.pylori-infected gastric mucosa (P<0.05).Conclusion IL-22 took part in H.pyloriinfection induced immune response and increased IL-22-producing T cells was the important feature of H.pyloriinfection.
2.Research on correlation between odor and chemical compounds of Lonicera japonica.

This study aims to investigate the relationship between odor and contents of the chemical compounds in Lonicera japonica, including chlorogenic acid, galuteolin and polyphenols. High performance liquid chromatography (HPLC) was applied to determine the contents of chlorogenic acid and galuteolin in L. japonica. The ponptent of polyphenols was determined by UV-Vis Spectrophotometry. Electronic nose was used to extract and measure the odor of L. japonica. Then SPSS 17.0 software was employed for data processing. There is a significant positive correlation between the comprehensive index value of aroma and the contents of chlorogenic acid and polyphenols. The regression equations have been established. However, the relationship between the comprehensive index value and the content of galuteolin is not obvious. This is proof that the odor of L. japonica has close connection with the chemical compounds. Therefore, this research offered a new method for initially determine or predict the content of the chemical composition in L. japonica,

3.Observation on therapeutic effect of electroacupuncture combined with acupoint-injection on acute gouty arthritis.

OBJECTIVETo search for a new method for treatment of acute gouty arthritis.
METHODSSixty cases of acute gouty arthritis were randomly divided into a treatment group and a control group, 30 cases in each group. The treatment group were treated with electroacupuncture combined with point-injection, and the control group with western medicine. Their therapeutic effects were observed and serum uric acid contents were detected before and after treatment.
RESULTSThere was a very significantly difference between the two groups in the therapeutic effect (P<0. 01) and the analgesic effect in the treatment group was better than that in the control group (P<0. 01). The treatment group was superior to the control group in decrease of blood uric acid (P<0. 01).
CONCLUSIONElectroacupuncture combined with point-injection is an effective method for treatment of acute gouty arthritis, and it can decrease blood uric acid content.

4.Human HSF1 mutants and their applications.

Heat shock factor 1 (HSF1) is the key protein in regulating stress response. It can be activated under heat, oxidative or another stress conditions. Dominant-positive and dominant-negative HSF1 are two types of HSF1 mutants. Both of them gain the DNA binding activity in the absence of stress. In addition, dominant-positive HSF1 acquires transcriptional activity, which dominant-negative HSF1 does not acquire. In this paper, the progress of using these HSF1 mutants in the research of cancer, neurodegenerative disorders and cardiovascular diseases will be discussed.

5.Study of leptin-treated mesenchymal stem cells modulate subsets of immune cells

Objective:Our previous study demonstrated serum leptin in patients with systemic lupus erythematosus(SLE) ac-celerated the senescence of mesenchymal stem cells (MSC),the aim of this study is to observe the effects of leptin-treated MSC on immune cells.Methods:Umbilical cord derived MSCs were isolated,and peripheral blood mononuclear cells (PBMC) in SLE patients were obtained by density gradient centrifugation with Ficoll.Three groups were divided,PBMC only,the co-culture of PBMC and MSC treated with or without leptin (100 ng/ml).The frequencies of immune cells were detected by flow cytometry,including CD4+CD25+Foxp3+regulatory T (Treg) cells,CD4+IL-17+Th17 cells,CD4+CXCR5+PD-1+T follicular helper (Tfh) cells,and CD19+B cells.Results:The frequency of regulatory T cells was lower in leptin-treated MSC group,compared with MSC group.The frequency of Th17 cells was increased by MSC pretreated with leptin.The frequency of Tfh cells was elevated by leptin-treated MSC compared with MSC control,but there was no statistical significance.MSC pretreated with leptin also restored the activation of T cells evaluated by CD25 and CD69,compared with untreated MSC.In addition,leptin-treated MSC increased the median fluorescence intensity of CD25, but not CD69 on B cells.Conclusion:Leptin not only accelerated cell senescence of MSC in vitro,but also impaired the capacity of MSC modulating the immune cells.
6.Correlation between polymorphisms in TNF-α and IL-10 genes and plasma cincentration with diffuse large B-cell lymphoma

Purpose To explore the association between single nucleotide polymorphisms (SNPs) of TNF-αand IL-10 genes as well as their plasma levels and pathogenesis of diffuse large B-cell lymphoma (DLBCL).Methods Snapshot SNP genotype technique was used to assess genetic variation in 6 SNPs for TNF-α and IL-10 in 38 DLBCL cases and 77 healthy controls,and enzyme-linked immunosorbent assay (ELISA) was used to measure the plasma concentrations of TNF-α and IL-10 in the above population.Results Plasma IL-10 level in DLBCL cases was higher than that in controls (P < 0.05),but there was no significant difference in plasma TNF-αlevel between the two groups (P > 0.05).Of all the candidate SNPs,the geneotype distribution for TNF-α-863C/A showed significant differences between the case and control groups (P < 0.05),and carriers with CA/AA genotypes had more two-fold risks of DLBCL than those with the CC genotype (95% CI =1.091-4.765,P =0.028).However several other candidate SNPs showed no significant difference in the geneotype distributions between the cases and controls (P > 0.05).After excluding the effects of disease status on plasma TNF-alpha and IL-10 levels,the polymorphism of each gene had no significant effect on the corresponding plasma concentration (P > 0.05).Conclusion TNF-α-863C/A polymorphism may be associated with the risk of DLBCL,and IL-10 play an important role in occurrence and development of DLBCL.
7.Incidence and etiology of stent associated respiratory tract infection caused by coated metal and silicone airway stents

Objective To investigate the incidence and bacterial etiology of stent associated respiratory tract infec-tion (SARTI) caused by two types of airway stents.Methods Silicone and coated metal airway stent were placed into patients with central airway stenosis caused by varied pathologies. The incidence of stent related respiratory tract infection,bacteria etiology of SARTI and improved dyspnea score were compared between two groups receiving different airway stent.Results 1)Totally 171 patients received airway stents, and among them, 39 patients (22.81%) developed SARTI.2)The incidence of SARTI in metal stent group and silicone stent group was 29.21% (26/89) vs.15.85% (13/82),P<0.05;3)Bacterial spectrum of SARTI was different in metal and silicone stent groups:staphylococcus aureus was 38.46% vs. 69.23%,respectively;candida albicans was 23.08% vs. 0%,re-spectively;Singular proteus was 7.26% vs.0%,respectively;4)The narrowed lumen was improved from 74.27%± 7.13% to 17.64%±6.22%in the metal stent group,while the data was improved from 74.94%±9.18% to 12.68%± 8.32% in the silicone stent group (P<0.01). Accordingly, the dyspnea symptomscore was improvedfrom 2.85 ± 0.89 to 0.85±0.68 in metal stent group,and from 2.88±0.91 to 0±0.61 in the silicone stent group (P<0.05). Conclusions Compared with metal airway stents,silicone stents have a lower incidence of SARTI,which mightbe due to the projections in the silicon stent surface and wider expanded in the bronchial stenosis.
8.Effects of long-term microwave irradiation on NMDAR, BDNF and related molecular expression in their signal pathways in rat hippocampus

Objective To evaluate the effect of long-term microwave radiation on the expression of N-methyl-D-aspartate receptor(NMDAR),brain derived neurotrophic factor(BDNF) and related molecules in signal pathways in the hippocampus of rats.Methods Fifty male Wistar rats were exposed to microwave radiation at an average power density of 0,5,10,20 and 30 mW/cm2for 6 min/time,3 times/week,and for 6 weeks,which were sacrificed and the hippocampus was quickly removed at 14 d and 28 d after exposure.The changes in NMDAR (NR1,NR2A,NR2B),postsynaptic density protein(PSD)-95,cortactin,BDNF and tyrosine kinase receptor B (TrkB) in hippocampal neurons of each group were detected by Western blotting and image analysis techniques.Results Compared with the control group,the expressions of related proteins did not change significantly after microwave irradiation of 5 mW/cm2 at each time point.After 20 mW/cm2 microwave radiation,the expression of NR1 was increased at 14 and 28 d (P <0.05),the expression of NR2A was increased at 28 d (P < 0.05),but the expression of NR2B was decreased at 14 and 28 d (P < 0.05).At a average power density of 30 mW/cm2,the expressions of NR1,NR2A and PSD-95 and the expression of NR2B were decreased at 14 and 28 d(P <0.05),and cortactin,BDNF and TrkB were increased at 14 d after irradiation (P < 0.05).Conclusion The effect of different dosages of long-term microwave radiation on the proteins of NMDAR and its signal pathway related molecules is different.Microwave radiation may affect the NMDAR of postsynaptic information transmission through the BDNF-TrkB signaling pathways,which might play an important role in the impediment of learning and memory function caused by microwave radiation.
9.Ecology suitability study of Lomatogonium rotatum in Inner Mongolia.

The distribution information of Lomatogonium rotatum. was collected by interview investigation and field survey, and 55 related environmental factors were collected, the habitat suitability study was conducted based on geographic information system (GIS) and maximum entropy model. The AUCs of ROC curve were both above 0.99, indicating that the predictive results with the maximum model were highly precise. The results showed that 13 major environmental factors have obvious influence on ecology suitability distributions of L. rotatum, including month average temperature of February et al., the suitable distribution areas are mainly concentrated in the east-central of Inner Mongolia, including Hexigten banner, Duolun county, Zhenglan banner et al., The zoning results basically coincide with the genuine producing areas, and further afford new suitable distribution areas, which can provide reference for L. rotatum's wild nursery and the siting of introduction and cultivation.

10.Effect of electro-acupuncture on clinical outcomes and ovarian hyperstimulation syndrome in in vitro fertilization and embryo transplantation.

OBJECTIVETo observe the effect of electro-acupuncture (EA) on clinical outcomes and the occurrence of ovarian hyperstimulation syndrome (OHSS) in in vitro fertilization and embryo transplantation.
METHODSTotally 109 patients who routinely received in vitro fertilization/intracytoplasmic sperm injection (IVF/ICSI) at Reproductive Center were assigned to the control group (56 cases) and the EA group (53 cases) according to even and odd-numbered date. Patients in the control group received controlled ovarian hyperstimulation (COH) referring to GnRH-a long protocol. On the basis of COH, those in the EA group received EA from the day of Gn injection to the day of embryo transfer. Estradiol (E2), vascular endothelial growth factor (VEGF), interleukin-6 (IL-6), and angiotensin (AT) II were measured in all patients on the day of hCG injection, the day of ovum pick up (OPU), and the day of embryo transfer (ET), respectively. The oocyte retrieval rate, good quality embryo rate, clinical pregnancy rate, the abortion rate, and the occurrence of OHSS were compared between the two groups.
RESULTSCompared with the control group, serum E2 levels on the day of OPU and the day of ET were significantly lower in the EA group (P < 0.05). On the day of OPU levels of VEGF and IL-6 also significantly decreased (P < 0.05). Serum levels of VEGF and IL-6 reached the highest line on the day of hCG in the two groups, and then showed a decreasing trend. Compared with the control group at the same time point, serum levels of VEGF and IL-6 obviously decreased on on the day of OPU, hCG, and ET (P < 0.05). The occurrence of OHSS and the canceling rate of transplant cycle were significantly lower in the EA group than in the control group (P < 0.05).
CONCLUSIONSEA, as an adjunctive therapy, could reduce the occurrence of OHSS in IVF. Besides, it did not decrease good embryo rates and pregnancy rates in IVF-ET, which might be associated with lowering local vascular permeability of ovaries.
