1.Drug resistance of bacteria in simple periodontitis in 138 cases

Objective:To detect drug resistance of bacteria in simple periodontitis.Methods:Bacteria from periodontal pocket were cultured and identified with the aerobic,anaerobic and L-form bacterium series of synchronism culture technology in 138 patients with simple periodontitis. Drug resistance of bacteria was detected with a fast chemosensitivity test.Results:135 bacterium strains were found in 138 samples (97.82%),multiple bacterial infecton rate was 81.48%,and penicillin resistancce rate was 90.37%.Conclusion:Multiple bacterial synergistic infection is principal in the simple periodontitis.Heigher curative rate may be obtained if treat the disease according to drug resistance analysis.
2.Cell metabolism of Taxus chinensis var. mairei in suspension cultures treated with methyl jasmonate

Object To study the physiological changes of Taxus chinensis var. mairei (Lemee et L?vl.) Cheng et L. K. Fu in the case of methyl jasmonate (MJ). Methods TTC assay, soluble protein measurement and enzyme analysis were used. Results It was observed that MJ inhibited Taxus cell growth in the view of primary metabolism. MJ induced phenylalanine ammonia lyase(PAL) activity while it inhibited polyphenoloxidase (PPO) activity. Extracellular phenolic content after addition of MJ increased to the maximum at the three days than that of the control group. Conclusion It was demonstrated that MJ induced the transition of Taxus cell from primary metabolism to secondary metabolism. This is favorable for secondary metabolism of Taxus cells. It is important to study the physiology of Taxus cells for revealing the mechanism of MJ.
3.Effect of dihydromyricetin on proliferation and apoptosis of breast cancer MCF-7 cells

Objective To investigate the effect of dihydromyricetin on proliferation and apoptosis of breast cancer MCF-7 cells.Methods From March 2014 to February 2015,breast cancer MCF-7 cells were treated with 99% pure DMY as an inhibitor.MTT assay,flow cytometry and immunocytochemistry were used to analyze the proliferation,apoptosis and protein expression of breast cancer cell MCF-7.Results When the DMY concentration was higher than 20 μg/mL,the inhibitory effect appeared,but not good.When 40 and 80 μg/mL DMY were used,the proliferation of MCF-6 cells were significantly inhibited,and have different degrees of sensitivity to it.When DMY was 80 μg/mL,the IC50 was 226.9 μg/mL.The inhibition rate and IC50 were compared with 0 μg/mL DMY,there was significant difference(P<0.05).Indicating that the relatively high concentration of DMY could significantly improve the patient′s condition of breast cancer;MCF-6 cells treated with 40 and 80 μg/mL DMY could induce G2/M phase arrest in DMY with concentration greater than 20 μg/mL(P<0.01),and showed significant cell apoptosis death phenomenon.At the same time,G0/G1 phase was also blocked and S phase cells decreased significantly,the difference was significant(P<0.05).The positive rate of PCNA in breast cancer MCF-6 cells significantly decreased(P<0.01) when the DMY concentration was higher than 50 μg/mL,the positive rate was> 50%,especially in DMY with 80 μg/mL,the positive rate was 10.00%.Compared with 0 μg/mL DMY,the difference was significant(P<0.05).Conclusion The use of dihydromyricetin in breast cancer patients can effectively inhibit the rapid increase of cancer cells,accelerate apoptosis,slow down the patient′s condition,the effect is outstanding.

5.Expression of Heat Shock Protein 70 and Bag-1 Protein in Gastroenteric Cancer

Objective To observe the expression of bag-1 and heat shock protein (HSP) 70 in clinical malignant tumors and the correlation between them. Methods RT-PCR and Immunohistochemistry were employed to examine the expression of bag-1 and HSP70 in 69 cases with gastroenteric cancer (54 cancer tissues and 15 non-cancer tissues). Results The expression of Bag-1 was detected in 96.3% of the cases, much higher than that in the control group (P0.05), and no correlation was observed between histological grade and HSP 70 expression. Conclusion There exists definite relationship between the gastroenteric cancer and the expression of bag-1 and HSP70, and there are close correlation between the tumor progress and the expression of bag-1.
6.DNA detection of human papilloma virus and herpes simplex virus in pterygia

Background To explore the roles of human papilloma virus(HPV)and herpes simplex virus (HSV)in pathogenesis of pterygia are very important for provide more basis for the study of pterygia mechanisms.Objective The aim of the study was to detect the expression of HSV and HPV in pterygia. Methods A total of 68 specimens of pterygia and specimens of normal conjunction tissue were obtained during the operation.The expression of HSV DNA and HPV DNA in pterygia specimens and conjunetival tissue were detected and compared by real time fluorescence quantitative polymerase chain reaction(PCR).Written informed consent was obtained from each patient before the any medical procedure related to this study. Results Real time fluorescence quantitative PCR showed that HPV DNA was detected in 12(17.65%)pterygia specimens,and HSV DNA was found in 15 (22.06%)pterygia specimens;however them were detected in 6(8.82%)and 1(1.47%)in conjunctival tissue respectively.Significant differences were found in the expression both HSV DNA and HPV DNA between pterygium tissue and eonjunctival tissue(HPV:χ2=10.291,P<0.05;HSV:χ2=4.561,P<0.05).The co-expression of HSV DNA and HPV DNA was seen in 5(7.35%)pterygium specimens,but no any co-expression of HSV DNA and HPV DNA was in conjunctival specimen. Conclusion HSV and HPV may participate in the pathogenesis and development of pterygium.The detection of virus DNA can offer an experiment evidence of HSV and HPV in pterygium generation.
7.Determination of phthalate esters in physiological saline solution by monolithic silica spin column extraction method

Monolithic silica spin column extraction (MonoSpin-SPE) was developed as a simple, sensitive, and eco-friendly pretreatment method which combined with ultra-fast liquid chromatography-mass spectrometry (UFLC-MS) to determine the levels of six phthalate esters, dimethyl- (DMP), diethyl- (DEP), dipropyl- [ DPrP], butyl-benzyl- (BBP), dicyclohexyl- (DcHP), and di-n-octyl-(DOP) phthalate in physiological saline samples. Under optimized experimental conditions, the method was linear in the following ranges: 0.2- 50 μg/L for DMP, DEP, DPrP, DcHP and DOP; 5- 100 μg/L for BBP. The correlation coefficients (R2 ) were in the range of 0. 9951 - 0. 9995 for all the analytes and the limits of detection (LODs) and limits of quantification (LOQs) were in the ranges of 0.02 - 0.9 μg/L and 0.08 - 2.7μg/L, respectively. The pretreatment process showed good reproducibility with inter-day and intra-day relative standard deviations (RSDs) below 8.5% and 11.2%, respectively. This method was used to determine the levels of six phthalate esters in physiological saline samples and the recoveries ranged from 71.2% to 107.3%. DMP and DEP were found in actual physical saline samples (brand A and brand B).

9.Morphosis of the epiphyseal plate and expression of vascular endothelial growth factor during healing of injured epiphyses

Objective To investigate morphosis of the epiphyseal plate and the expression of vascular endothelial growth factor (VEGF) in the epiphyseal plate during healing of injured epiphysis.Methods Thirty SD rats aged 4-5 weeks were used to make models of proximal tibia epiphysis injury.These models were equally divided into 5 groups in random,with 6 rats in each group,and these groups of rats were killed separately at 2,4,6,10,21 days postoperatively to harvest the proximal tibia epiphysis,in order to explore the morphosis of the epiphyseal plate and VEGF expression pattern in the epiphyseal plate at different healing phase.The structure of the epiphyseal plate was evaluated by measuring the length of the limb and radiographic examination of the limb.The ratio of length of injured tibia and that of the other side was used to describe the tibia length discrepancy.The micro structure of the epiphyseal plate and situation of the bone bridge in the epiphyseal plate were measured by using Micro CT.By using HE stain and VEGF immunohistochemistry,the changing of chondrocyte and VEGF expression pattern in the epiphyseal plate were observed at different healing phase.Results The limb discrepancy appeared at 4 days postoperatively,became biggest at 10 days postoperatively,and tended to decrease at 21 days postoperatively.Micro CT demonstrated the fibrous bone tissue formation in the epiphyseal plate at about 6 days postoperatively,which became bone bridge finally.HE stain showed chondrocyte in the rest zone had the tendency to aggregate after injury,and the highly ordered structure of chondrocyte in the epiphyseal plate no longer existed,chondrocyte differentiation was accelerated.VEGF immunohistochemistry stain showed the high reaction of VEGF in the epiphyseal plate appeared after surgery,and the positive zone of VEGF expanded through the physis with time,which finally led to angiogenesis and ossification.Conclusion A serial alteration of morphosis of the epiphyseal plate occurred during the healing process.Fibrous bone tissue formation in the epiphyseaL plate could be observed in the early and mid-term of the healing process.With progress of healing,VEGF expression zone gradually expanded across the epiphyseal plate,which is related to the formation of bone bridge.
