1.Simultaneous HPLC Determination of Hesperidin, Magnolol, Honokiol and Liquiritin in Soft Capsule Jia-Wei Huo-Xiang Zheng-Qi

This study was aimed to develop an HPLC method for the determination of hesperidin,magnolol,honoki-oland liquiritin in Soft Capsule Jia-Wei Huo-Xiang Zheng-Qi (JWHXZQ). AKromasil C18 column (250 mmí4.6 mm, 5 μm) was used with water-methanol as mobile phasegradient elution. The flow rate was 1.0 mL·min-1, and the de-tecting wavelength was at 287 nm. The results showed that the linearity ranges ofhesperidin,magnolol,honokioland liquiritinwere 4.47-178.70 μg·mL-1, 3.42-136.96 μg·mL-1, 3.49-139.48 μg·mL-1, 3.92-157.20 μg·mL-1, respec-tively (r>0.999). The average recoveries of them were 99.48%, 99.05%, 99.57% and 99.79%, respectively. It was concluded that the method was accurate, sensitive and specific for quality control of Soft Capsule JWHXZQ.

Catalpol is a kind of iridoid,which has wide pharma-cological activities,including anti-cerebral ischemia,improving senile dementia,anti-inflammation,inhibiting capillary permea-bility,relieving pain,anti-tumor,antidiarrheal,reducing blood sugar level,protecting liver,and anti-aging.The mechanisms of Catalpol effects have been well studied.Signaling pathways in-clude NF-κB signaling pathway,PI3K/AKT signaling pathway, BDNF /TrkB signaling pathway,JAK2 /STAT3 /angiogenesis sig-naling pathway,MAPK signaling pathway,TGF-β/Smad signa-ling pathway,and ACh signaling pathway.We reviewed related signaling pathways of Catalpol effects,in order to broaden the understanding of molecular mechanism and signaling pathways of Catalpol,to know the status of catalpol,and to provide new di-rection to study Catalpol.

4.Dissolution testing combined with computer simulation technology to evaluate the bioequivalence of domestic amoxicillin capsule.

Re-evaluation of bioequivalence of generic drugs is one of the key research focus currently. As a means to ensure consistency of the therapeutic effectiveness of drug products, clinical bioequivalence has been widely accepted as a gold standard test. In vitro dissolution testing based on the theory of the BCS is the best alternative to in vivo bioequivalence study. In this article, the conventional dissolution method and flow-through cell method were used to investigate the dissolution profiles of domestic amoxicillin capsules in different dissolution media, and the absorption behavior of the drugs with different release rates (t85% = 15-180 min) in the gastrointestinal tract was predicted by Gastro Plus. The flow-through cell method was thought better to reflect the release characteristics in vivo, and amoxicillin capsules with regard to the release rates up to 45 min (t85% = 45 min) were having a satisfied bioequivalence with the oral solution according to the C(max) and AUC. Although two different dissolution profiles of domestic amoxicillin capsules were found by flow-through cell methods, prediction results revealed that domestic capsules were probably bioequivalent to each other.

7.Construction and confirmation of the fused plasmid with Rb bait gene in yeast two-hybrid system.

OBJECTIVETo screen and clone the novel genes related to cellular proliferation of oral squamous cell carcinoma.
METHODSWe selected the Rb gene as the bait protein gene to construct the fusion bait plasmid of yeast two-hybrid. The whole code sequence of Rb gene was acquired by digestion with restricted enzyme EcoRI and BamH1 and reclaimed from its original vector pGBT9-pRb. After being confirmed by electrophoresis, the Rb gene was cloned into the MCS of the plasmid pGBKT7 to construct a recombined plasmid pGBKT7-pRb and the sequence of the recombined plasmid was detected in company. According to the protocol of yeast two hybrid system III, the competent Y187 yeast was prepared, and transformed with recombined plasmid pGBKT7-pRb. Following that, the toxicity and transcriptional activation of this recombined plasmid pGBKT7-pRb in Y187 yeast were tested.
RESULTSThe sequence of the recombined plasmid was correct compared with the sequence provided in Genbank. The protein could be correctly synthesized in vitro, and no self-activating transcriptional activation and toxicity was observed in Y187 yeast.
CONCLUSIONSThe construction of the recombined plasmid was capable to be used as the fusion bait plasmid in yeast two-hybrid system III, and the recombined Rb-protein could be used as the bait protein successfully.

8.Muscle fatigue: general understanding and treatment.

Muscle fatigue is a common complaint in clinical practice. In humans, muscle fatigue can be defined as exercise-induced decrease in the ability to produce force. Here, to provide a general understanding and describe potential therapies for muscle fatigue, we summarize studies on muscle fatigue, including topics such as the sequence of events observed during force production, in vivo fatigue-site evaluation techniques, diagnostic markers and non-specific but effective treatments.

Objective To evaluate the use of gated SPECT in patients with hypertrophic obstructive cardiomyopathy (HOCM) and the effects of percutaneous transluminal septal myocardial ablation (PTSMA) on myocardial perfusion.Methods 99 Tcm-methoxyisobutylisonitrile (MIBI) and 18F-fluorodeoxyglucose (FDG) images were performed in 31 HOCM patients before PFSMA and in 15 patients 3-7 d after PTSMA.The images in different left ventricular segments were analysed by using scores.Results In 99Tcm-MIBI images, uptake decreased at the septal regions in 12 HOCM patients (80.0%, 12/15) after PTSMA, 18F-FDG images also showed decreased uptake at the septal regions in 5 HOCM patients (33.3%, 5/15) after PTSMA.Conclusion 99Tcm-MIBI images might be an important method to evaluate PTSMA results, and 18 F-FDG images showed important value as reference.
10.Study on effect of different combinations of traditional Chinese medicine preparation active parts on metabolism of puerarin in plasma of rats.

OBJECTIVETo build an experimental method to determine the content of puerarin in plasma, observe the effect of different combinations of traditional Chinese medicine preparation active parts on metabolism of puerarin in plasma of rats suffering cerebral ischemia attack after being orally administered the main active parts of Yangyin Tongnao granules.
METHODAbsorption of puerarin in plasma was extracted by acetonitrile. Using hydroxybenzoate as interior standard, methanol-0.5% acetic acid water solution as mobile phase, ODS C(18) chromatography column (4.6 mm x 150 mm, 5 microm) as stationary phase, puerarin was detected at 248 nm.
RESULTThere is a good linear relationship for puerarin in the range 0.025-3.2 microg x mL-1, RSD < 10%, and the average recovery is 97.6%.
CONCLUSIONThe method is sensitive, reliable, and can be well applied in this experiment. It can provide a method for determination of active components in TCM preparation in vitro. Different compatibility influences metabolism of components.
