OBJECTIVETo study the overexpression of Sox9 gene on rabbit bone marrow mesenchymal stem cells for repairing articular cartilage injury in vivo.
METHODSRabbit bone marrow mesenchymal stem cells (BMSCs) were transduced with lentivirus vector containing Sox9 gene and then cartilage specific molecule was detected by RT-PCR in vitro. Total 48 knee joints of 24 mature New Zealand white rabbits were randomly divided into 3 groups according to different defect treatment. After animals anesthesia,a full-thickness cylindrical cartilage defect of 4 mm diameter and 3 mm deep was created in the patellar groove using a stainlesssteel punch. Meanwhile, the transfected cells were implanted to repair the rabbit model with full-thickness cartilage defects. Cartilage defects tissue was observed with light microscope, electron microscope, HE and immunohistochemistry staining to assess the repair of defects by the complex at 6 weeks or 12 weeks after the implantation.
RESULTSAt 3 days after the transfection, Sox9 gene expression was highest and Sox9 gene expression decreased with the increase of time. At 3 days after the transfection, the expression of collagen type II began and reached the peak at 14 days. It showed that the bone marrow mesenchymal stem cells went into chondrogenic differentiation after transfected by Sox9 gene. Histological observation showed that at 6 weeks after the operation, the defects in the experimental group was filled with hyaline like cartilage tissue, 12 weeks after operation,the defects of cartilage and subchondral bone had satisfactory healing. Both at 6 and 12 weeks postoperatively, the defects were filled with fibrous tissues in control groups. Meanwhile, immunohistochemical staining of sections with type II collagen antibodies showed the proteins in the regenerated tissue stained positive for type II collagen and stronger than the control groups. The histological scoring system indicated that the cartilage repair of experiment groups were better than the two control groups with statistical significances.
CONCLUSIONOverexpression of Sox9 gene on rabbit bone marrow mesenchymal stem cells (BMSCs) promote the repair of cartilage defect.

4.Analysis of 105 cases of pregnancy -induced hypertension patients with Ⅲphase of retinopathy

To explore the related risk factors and outcome in pregnancy- induced hypertension patients with Ⅲ phase of retinopathy.
●METHODS: A total of 105 pregnancy - induced hypertension patients with Ⅲ phase of retinopathy in our hospital from Januany 2012 to December 2013 were enrolled. Clinical date of them were collected to analyze.
●RESULTS: The occurrence of pregnancy - induced hypertension patients with Ⅲ phase of retinopathy were positively correlated with the course of the disease, blood pressure, proteinuria, and it was higher occurred in cold winter and spring, timely termination of pregnancy and appropriate hormone therapy can promote the recovery of vision, and improve outcomes of pregnancy.
●CONCLUSlON: The occurrence of pregnancy - induced hypertension patients with Ⅲ phase of retinopathy associated with season and disease severity. Timely treatment can restore normal vision, improve maternal and neonatal prognosis. Routine examination of fundus examination should be used as the pregnancy induced hypertension syndrome.
5.Chemical constituents from Morus notabilis and their cytotoxic effect.

Une new flavonoids named as notabilisin K (1), together with four known compounds, morusin (2), mulberrofuran A (3), neocyclomorusin (4) and mornigrol F (5) are separated from 95% ethanol extracts of the twigs of Morus notabilis. Compounds 2-5 are separated from this plant for the first time. Notabilisin I, notabilisin J exhibits certain effect against cells of HCT-116, HepG2 and A2780 with IC50 values ranging from 1.47 μmol x L(-1) to 5.46 μmol x L(-1). Morusin exhibits strong effect against five kinds of human cancer cells (BGC823, A2780, HCT-116, HepG2 and NCI-H1650) with IC50 values ranging from 0.74 μmol x L(-1) to 1.58 μmol x L(-1).

7.MRI diagnosis of cavernous transformation of the portal vein

ObjectiveTo investigate the value of MRI in the diagnosis of cavernous transformation of the portal vein (CTPV).MethodsPlain MRI,dynamic enhanced and (or) dynamic contrast enhanced magnetic resonance angiography(DCE-MRA) findings in 30 patients of clinical-proved CTPV were retrospectively analyzed.ResultsAmong 30 CTPV patients on plain MRI,obliteration of main and (or) branched portal vein were found,and mass-like or reticular abnormal soft-tissue signals were around the vein,which were produced by collateral vessels.On dynamic enhanced MRI,abnormal hepatic perfusion during arterial phase and abnormal enhanced collateral veins during portal phase could be seen.The above signs became more obvious on DCE-MRA.ConclusionsMRI and DCE-MRA can clearly visualize the anatomical features of CTPV.It is important and can provide the reliable evidence for planning properly therapeutic protocol to recognize and directly evaluate the CTPV.
8.Clinical effects of arthroscopic-assisted allograft reconstruction of anterior cruciate ligament injuries

Objective To determine 2-4 year intermediate-term clinical effects of deep-freezing bone-ACL-bone allograft plus endobutton plate fixation in reconstruction anterior cruciate ligament(ACL) injuries after ACL injury and estimate the feasibility and validity of the technique.Methods We car- ried out a retrospective study on 15 cases with ACL injury of lateral knee joint reconstructed with deep- freezing bone-ACL-bone allograft plus endobutton plate fixation method from September 1999 to October 2002.The average follow up was 36.9 months.The myodynamic recovery,muscular activity,Rom Lach- man test,Pivot shift test,Lysholm score and X-ray of tunnels enlargement at intermediate term were com- pared with normal ones.Results The circumference of thigh was(0.976?0.119)cm shorter than that of the normal side.Extension limitation of less than 3?was found in 12 cases and knee flexion limita- tion for below 5?in 13.Lachman test showed negative results.Lyshohn score was increased from preoper- ative 66.2?4.6 to postoperative 89.4?3.2.X-ray showed no tunnel enlargement.Conclusions Re- construction of ACL with deep-freezing bone-ACL-bone allograft plus endobutton plate fixation under ar- throscopy can attain equal length and anatomic reconstruction of ACL.Deep-freezing bone-ACL-bone is beneficial to functional recovery of ACL.
9.Effect of fengshiqing on interleukin-4, gamma-interferon and chemotactic factor in rats with collagen type II.

OBJECTIVETo study the regulatory effect of fengshiqing (FSQ) on interleukin 4 (IL-4), gamma-interferon (gamma-IFN), macrophage inflammatory protein 1alpha (MIP-1alpha) and collagen type II antibody (C II Ab) in serum and supernatant of synovial cell culture of rat with rheumatoid arthritis (RA), to explore the mechanism of action of clearing-heat and activating blood method for treatment of RA.
METHODSRA rat model was induced by C II Ab combined with Freund's complete adjuvant, and the levels of IL-4, gamma-IFN, MIP-1alpha and C II Ab in serum and supernatant of synovial cell were detected by ELISA.
RESULTSAs compared with the normal group, serum level of C II Ab in the model group was significantly higher (P < 0.01), serum and supernatant contents of IL-4 on the 14th and 28th day of modeling were lower and those of gamma-IFN and MIP-1alpha were higher, the difference showed statistical significance (P < 0.05 or P < 0.01). After being treated with FSQ and IL-4 contents in serum and supernatant as well as MIP-1alpha in supernatant restored on the 14th day (P < 0.05 or P < 0.01), while all the indexes restored on the 28th day.
CONCLUSIONFSQ could evidently up-regulate the level of IL-4 and down-regulate that of MIP-1alpha in serum and local synovial membrane in RA rats, and shows a suppressive trend of gamma-IFN, so as to maintain the Th1/Th2 equilibrium, suppress the cellular and humoral immune response in the local synovial membrane, and alleviate the chronic changes of arthritis, synovitis and vasculitis.

10.Medial plica after reconstruction of anterior cruciate ligament.

BACKGROUNDThe medial plica may be caused by direct trauma or joint degeneration, which also could be iatrogenic. There have been few reports in the literature discussing incidence of the medial plica caused by an operation on the knee joint, specifically after the reconstruction of anterior cruciate ligament (ACL). In this study, we aimed to evaluate and analyze the relationship between the incidence of the medial plica and reconstruction of the ACL.
METHODSA retrospective case series study was conducted to review the findings of 1085 patients between 2003 and 2007, who underwent second-look arthroscopy after reconstruction of the ACL (between 2002 and 2006). The correlation of the incidence of medial plica with the stability of the knee joint, the time from onset of injury to reconstruction surgery, the associated injuries, and the rate of progress during postoperative rehabilitation were analyzed.
RESULTSWe found that 722 patients had the structure of a medial plica. The incidence after reconstruction of the anterior cruciate ligament (66.5%) was significantly higher than usually reported. All these medial plica had avascular fibrotic and thickened edges. An excision of pathologic medial plica and fat pad synovial fringes were done. The incidences were significantly different between the two groups with their reconstruction operation time, from onset of injury to surgery (less than one month or over 2 years), and the progress rate of postoperative rehabilitation (knee flexion could not be over 90 degrees in four weeks). The incidence was not different between the groups with knee stable conditions.
CONCLUSIONSMedial plica is more common in patients after reconstruction of ACL. More associated injuries and more rehabilitation difficulties can increase the medial plica incidence.
