2.Learning and Memory Capacity and NMDA Receptor Expression in Shen Deficiency Constitution Rats.

OBJECTIVETo explore material bases and neurobiological mechanisms of "Shen storing will" by observing learning and memory capacities and N-methyl-D-aspartic acid (NMDA) receptor expressions in Shen deficiency constitution (SDC) rats.
METHODSTotally 40 SD rats were randomly divided into the model group, the Zuogui Pill (ZP) group, the Yougui Pill (YP) group, the blank control group (consisting of normal pregnant rats), 10 in each group. SDC young rat model (inherent deficiency and postnatal malnutrition) was prepared by the classic way of "cat scaring rat". Medication started when they were scared by cat. Rats in the ZP group and the YP group were administered by gastrogavage with ZP suspension 0.1875 g/mL and YP suspension 0.0938 g/mL respectively. Equal volume of normal saline was administered to rats in the blank control group and the model group by gastrogavage. All medication was given once per day, 5 days in a week for 2 consecutive months. Learning and memory capacities were detected by Morris water maze test. Expressions of NMDA receptor subunits NR2A and NR2B in hippocamus were detected by immunohistochemical method.
RESULTSCompared with the blank control group, the latency period, total distance in Morris water maze test were longer in the model group (P < 0.05). All the aforesaid indices all decreased in the ZP group and the YP group, with statistical difference when compared with the model group (P < 0.05). The protein expressions of NR2A and NR2B in hippocamus were lower in the model group than in the blank control group (P < 0.05). But when compared with the model group, they were obviously higher in the ZP group and the YP group (P < 0.05).
CONCLUSIONSSDC rats had degenerated learning and memory capacities and lowered NMDA receptor expressions. ZP and YP could up-regulate learning and memory capacities and NMDA receptor expressions, thereby improving deterioration of brain functions in SDC rats.

3.Clinical observation of porcelain and composite veneer in repairing dental fluorosis

Objective To observe the clinical changes in resin and porcelain veneer in restoring dental fluorosis in order to provide a basis for the repair of dental fluorosis. Methods Fifty six severe dental fluorosis patients were divided into porcelain and resin teeth group in the department of Prosthetics, school of Stomatology, Harbin Medical University during 2005 to 2008. All 162 teeth of 25 patients in porcelain group were veneered with porcelain. 201 teeth of 31 patients in resin group were repaired with resin. To evaluat the clinic effect, the veneer surface color was detected by the Easyshade computer-aided colorimeter when the repair was completed and 18 months afterward. The edge of veneer adaptation, retention, secondary caries and abutment were examined after 18 months, and classified by color, shape, function and feeling. Results The color difference between the porcelain and resin teeth group was 0.27±0.20 and 0.21±0.15 when it was completed, and it was 0.28±0.21 and 0.77± 0.68 respectively after 18 months. The color difference value of the porcelain teeth group was lower when it was completed than 18 months later(t=-13.55, P<0.01). The color difference value of the resin teeth group was lower than the porcelain teeth group after 18 months(t=-12.60, P<0.01). The percentage of level A of veneer adaptation in the porcelain group[100%(162/162)] was higher than the resin group[91.04% (183/201), χ2=15.26, P< 0.01) after 18 months. The clinical effect was divided into three degrees of excellent, moderate or failed, into which the number of the teeth catergorized was 158, 4 and 0 in porcelain group, 148, 56 and 4 in resin group respectively. The clinical effect of the porcelain group was superior to the resin group(χ2=44.24, P<0.01). Conclusions The surface color of porcelain veneer last 16nger than the resin veneer, the adaptation and clinical effect is also superior to the resin veneer. But the long-term efficacy of two methods needs further study, especially of the resin veneer.

5.Chemical composition of fructus Liquidambaris--lulutong.

AIMTo study the chemical composition in the fruits of Liquidambar formana Hance.
METHODSVarious column chromatographic techniques were used to separate and purify the constituents. Their physical and chemical properties and spectral data were measured for structural elucidation.
RESULTSEleven compounds were isolated and identified as beta-sitosterol (1), 3-oxo-11 alpha, 12 alpha-epoxyleanan-28, 13 beta-olide (2), 3-oxo-12 alpha-hydroxy-oleanan-28, 13 beta-olide (3), 3 alpha-acetyloxy-25-hydroxyolean-12-en-28-oic acid (4), oleanolic acid (5), ursolic acid (6), daucosterol (7), betulonic acid (8), gallic acid (9), nonacosane (10) and n-triacontanoic acid (11).
CONCLUSIONAmong the isolated constituents, compound 4 is new compound, compound 3 is firstly isolated from the natural product and compound 5, 6, 7, 9, 10 and 11 are isolated from LuLuTong for the first time.

6.Research progress on solid dispersion technique in preparative field

Solid dispersions (SD) as a preparation of intermediates have played an important role in improving the dissolution of insoluble drugs and its bioavailability.SD technique is one of the most promising techniques to improve the dissolution and solubility of insoluble drugs,and the development of SD technique will promote the gradual perfection in preparative field.This review focuses on the carrier materials of SD,various new preparation techniques and their comparisons,application of solid dispersion formulations,and stability problems of SD.The factors influencing the stability of SD are described,and the effective measures to prevent the aging of SD are put forward.Finally,the review puts forward the practical suggestions of the solid dispersion technique.
7.Effect of excessive iodine intake on sodium-iodide symporter mRNA and protein expression of breast in lactating rats

Objective To study effect of excessive iodine intake on sodium-iodide symporter(NIS)mRNA and protein expression of breast in lactating rats.Methods60 Wistar rats,having been weaned for one month,were randomly divided into three groups according to their body weights,I.e,①normal iodine(NI,30 rats);②ten fold high iodine(10 HI,15 rats);③one hundred fold high iodine(100 HI,15 rats).Eating food containing iodine of 300μg/L and drinking water of iodine at 5,1845,20 295μg/L,respectively.After fed for 3 months,the rats mated and had offspring,and urine and milk iodine of lactating rats were determined by As-Ce-catalytic spectrophotometric method.Their marmnary glands were sampled at lactation day 10.Then NIS mRNA expression by RT-PCR was determined and NIS protein by immunohistochemistry(SABC)was observed.Results The urine iodine of 10 HI group(3597.5μg/L)and 100HI group(25 404.3μg/L)increased obviously compared with that of NI group(344.7μg/L).The milk iodine of 10HI group(27.1×103μg/L)and 100HI group(191.0×1μg/L)was higher than that of NI group(6.0×103μg/L),but the increased fold of milk iodine was not paralleled with that of urine iodine.Difference of NIS mRNA expression was significant(F=24.19,P<0.01)among the groups,and the NIS mRNA expression in 10HI(1.250±0.034)and 100HI(1.272±0.039)group were less than that in NI (1.532±0.044)group(P<0.01).The breast NIS mRNA expression in lactating rats(1.532±0.044)was significantly higher than that in unlactating rats(0.879±0.018,P<0.01).With the increasing iodine uptake,NIS protein expression decreased.Conclusions The NIS mRNA and protein in rat breasts is down-regulated by excessive iodine intake.So increasing extent of milk iodine concentration is inhibited,which is important to prevent off-spring from getting excessive iodine intake from parental generation.
9.Therapeutic effect of Xinmailong injection on advanced aged patients with acute myocardial infarction heart failure/

To explore influence of Xinmailong injection on cardiac function and vascular endothelial func‐tion in advanced aged patients with acute myocardial infarction (AMI) heart failure (HF).Methods : A total of 122 advanced aged AMI‐HF patients treated in our hospital from Mar 2015 to Jul 2017 were selected ,randomly and e‐qually divided into routine treatment group (received routine anti‐HF medication ) and Xinmailong group (received Xinmailong injection based on routine treatment group ) ,both groups were treated for 10d and then followed up for six months .LVEF ,wall motion score index (WMSI) ,serum levels of BNP , hsCRP , cTnI ,nitric oxide (NO) ,en‐dothelin (ET)‐1 and inner diameter of brachial artery were observed and compared between two groups before and after treatment .Results : Compared with before treatment ,there were significant rise in LVEF [routine treatment group : (35.42 ± 5.11)% vs. (44.62 ± 5.88)%;Xinmailong group : (36. 13 ± 5.64)% vs. (49.75 ± 6. 18)%] and serum NO level [routine treatmentgroup : (108. 35 ± 21.67) μmol/L vs. (129.68 ± 25. 83 ) μmol/L ; Xinmailong group : (110.26 ± 22.71) μmol/L vs. (156.82 ± 24. 19) μmol/L] ,and significant reductions in WMSI [routine treatment group : (2.49 ± 0.51) score vs. (1.45 ± 0.34) score ; Xinmailong group : (2.51 ± 0.42) score vs. (1. 12 ± 0.27) score] ,NYHA class (routine treatment group :3.19 ± 0. 44 vs. 2.65 ± 0.36 ;Xinmailong group :3. 21 ± 0.42 vs. 2.11 ± 0.28) ,serum levels of BNP [routine treatment group : (986. 72 ± 245.63 ) pg/ml vs. (764.42 ± 217.33) pg/ml ; Xinmailong group : (989.99 ± 243. 54 ) pg/ml vs. (607. 82 ± 198. 65 ) pg/ml ] , hsCRP [routine treatment group : (24.31 ± 3.82) mg/L vs. (14.52 ± 1. 69 ) mg/L ; Xinmailong group :(24.93 ± 3.74) mg/L vs. (7. 81 ± 0. 96) mg/L] ,cTnI [routine treatment group :(5.29 ± 0.43) μg/L vs. (3.01 ± 0. 27) μg/L ;Xinmailong group :(5.37 ± 0.64) μg/L vs. (1.42 ± 0.33) μg/L ] and ET‐1 [routine treatment group :(95. 67 ± 16. 32) ng/L vs. (76. 34 ± 17.82) ng/L ;Xinmailong group :(96.13 ± 17.21) ng/L vs. (57.29 ± 14. 63) ng/L] in two groups ;and LVEF and serum NO level of Xinmailong group were significantly higher than those of routine treatment group , WMSI ,NYHA class ,serum levels of BNP ,hsCRP ,cTnI and ET‐1 of Xinmailong group were significantly lower than those of routine treatment group , P=0.001 all.Compared with before treatment ,there were significant rise in reactive hyperemia brachial diameter (Dhyperemia ) , fore brachial artery endothelium dependent diastolic‐systolic function (FMD) in Xinmailong group after treatment , and they were significantly higher than those of routine treatment group , P<0. 05 or <0. 01 .Conclusion : Xinmailong injection can significantly improve cardiac function and vascular endothelial function in AMI‐HF patients ,which is worth extending .
