2.Effect of antisense oligodeoxynucleotide of vascular endothelial growth factor C on lymphangiogenesis and angiogenesis of pancreatic cancer.

In order to investigate the effect of antisense oligonucleotide (ASODN) of vascular endothelial growth factor C (VEGF-C) on lymphangiogenesis and angiogenesis of pancreatic cancer, antisense and scamble-sense oligonucleotide of VEGF-C were constructed, and the model of nude mice with orthotopically xenografted human pancreatic cancer cells (Panc-1) was established. Thirty nude mice were randomly divided into 3 groups: PBS control group (group A), scramble-sense control group (group B) and antisense group (group C). All nude mice were treated once every 2 days as 3 times per week, for 3 weeks (oligonucleotide 10 mg/kg every time). After treatments were completed, ELISA method was used to examine the concentration of VEGF-C in plasma and immunohistochemical method to examine microvessel density (MVD), lymphtic vessel density (LVD) of pancreatic cancer. The results showed that the expression of VEGF-C was inhibited significantly in group C. The concentrations were 237.5+/-41.5, 221.5+/-52.3 and 108.6+/-14.9 pg/mL in groups A, B and C respectively (P<0.01). LVD in groups A, B and C was 13.8+/-2.1, 12.4+/-1.9 and 4.2+/-1.6 respectively (P<0.01). MVD in groups A, B and C was 27.5+/-8.7, 25.9+/-4.2 and 19.4+/-5.6 respectively with no significant difference among the groups (P>0.05). It was suggested that VEGF-C ASODN decreased the expression levels of VEGF-C in nude mice with orthotopically xenografted human pancreatic cancer, and it could inhibit lymphangiogenesis, but had no significant effect on angiogenesis.
3.Protective effect of Jiechangning decoction in treating experimental ulcerative colitis in guinea pigs.

OBJECTIVETo study the therapeutic effects and mechanism of Jiechangning (JCN) decoction on carrageenan induced experimental ulcerative colitis (UC).
METHODSAfter sensitizing guinea pigs with carrageenan, we established UC animal models by free drinking water containing 2% acid degraded carrageenan (ADC). JCN decoction was orally administered once a day for 2 weeks after carrageenan treatment. Salicylazosulfapyridine (SASP) and normal saline were given to the other two groups as control. The levels of colon lipid peroxide (LPO), acid phosphatase (ACP) activity and tumor necrosis factor-alpha (TNF-alpha) were measured; colitis activity score (CAS) was carried out for assessment of the degree of tissue inflammation and injury; the colonic pathological changes were examined simultaneously with hematoxylin and eosin (HE) and toluidine blue staining used to evaluate the therapeutic effects of JCN decoction and SASP.
RESULTSExperimental colitis models resembling human UC were successfully induced. The levels of tissue LPO, ACP activity and the content of tissue TNF-alpha were markedly increased in the model group as compared with the normal control group (P < 0.01) and were positively correlated with CAS. JCN decoction could reverse these changes like SASP. HE staining showed that JCN decoction and SASP could reduce CAS and the degree of tissue injury, toluidine blue staining revealed that mucosa and submucosa red metachromasia pellets in JCN group and SASP group were markedly fewer than those in the model group.
CONCLUSIONJCN decoction is effective in treating experimental UC, which provides theoretical basis for its clinical application.

4.The application of psychometric measures in diagnosis of minimal hepatic encephalopathy

Objective To establish the normal parameters of psychometric measures such as the number connection tests A(NCT-A)and digit symbol tests(DST)in assessment of minimal hepatic en- cephalopathy(MHE).Methods One hundred and sixty healthy volunteers(aged 25 to 64 years;educa- tional level>9 years)were divided into<35 ys,35~44 ys,45~54 ys and 55~64 ys groups.All of the healthy volunteers were assessed with NCT-A and DST to establish the normal value of age-related parameters,which can be used for diagnosis of MHE in patients with liver cirrhosis.Two standard devi- ation of the normal mean was used as a diagnostic criterion for MHE.One hundred and six cirrhotic patients were assessed with these parameters.Results The parameters of NCT-A were(25.1?4.6) sec in<35 ys group,(32.1?6.8) sec in 35~44 ys group,(38.6?7.1)sec in 45~54 ys group or (49.3?6.3)sec in 55~64 ys group.The scores of DST were 49.9?4.7 in<35 ys group,44.6?4.8 in 35~44 ys group,38.5?5.0 in 45~54 ys group or 35.4?4.7 in 55~64 ys group.Thirty one out of 106 cirrhotic patients were diagnosed as MHE based on these parameters.Conclusion The NCT- A and DST are psychometric assessments for diagnosis of MHE.Age-based normal paramerters of NCT- A and DST are needed to be established.
5.Effect of intravenous fluid infusion combined with water throuth gastrointestinal tract on treatment of patients with hyperosmolar hyperglycemic state

Objective To investigate the effect of intravenous fluid infusion combined with water throuth gastrointestinal tract in treating the patients with hyperosmolar hyperglycemic state(HHS).Methods 30 HHS pa- tients were recruited.All the patients were given water throuth gastrointestinal infusion while they were administrat- ed continuously intravenous infusion.Laboratory parameters such as serum natrium,serum potasium,serum glucose and serum osmolarity ect were monitored at the admission and after treatment.Results Serum glucose and serum osmolarity of HHS patients were decreased smoothly at the speed of less than 3 mmol?L~(-1).h~(-1)during the first 12 hours after treatment.After 48 hour-treatment,serum natrium,serum potassium and serum osmolarity recovered to normal levels except 2 deaths,serum glucose decreased to(10.8?5.2)mmol/L.Conclusion Intravenous fluid infu- sion combined with water throuth gastrointestinal tract for the patients could lower smoothly serum glucose and serum osmolarity and decrease the mortality of the HHS patients.
6.Cross-sectional study on rates and influential factors of standard medication for antiretroviral therapy in preventing mother-to-child transmission of HIV.

OBJECTIVETo understand the status of standard medication of antiretroviral therapy and identify its correlationship with preventing mother-to-child transmission of HIV (PMTCT).
METHODSQualitative and quantitative methods for measuring of standard medication towards antiretroviral therapy of PMTCT were used in a cross-sectional study of 167 pregnant women who had accessed to PMTCT services and 57 physicians providing service from January 2005 to June 2007 in 5 HIV epidemic counties in Yunnan province.
RESULTSA total of 167 mothers were recruited, of which 65.87% (110/167) demonstrated for standard medication. The percentage of rational selection of antivirus-therapy regimen was 88.02% (147/167). HIV positive mothers with rational using medicine during labor covered 81.37% (131/161). 87.42% (146/167) of women demonstrated good compliance. The main correlations with the standard medication level of HIV positive pregnant women and their infants were as follows: diagnose time (OR = 2.617; 95% CI: 1.184 - 5.783), place of delivery (OR = 0.064; 95% CI: 0.007 - 0.607), minorities (OR = 0.344; 95% CI: 0.162 - 0.730), understanding of HIV women for antiretroviral therapy (OR = 6.843; 95% CI: 1.449 - 32.312), and doctors' cognition about the regimen for antiretroviral therapy was not enough; 5 key knowledge points (purpose of PMTCT, the consequence of not requiring standard medication, the effect of knowing regimens, the relationship of CD4 levels and selected regimens, the side effect of the relevant medicine) score rate was 47.72% (136/285).
CONCLUSIONThe proportions of standard medication in PMTCT antiretroviral therapy were not high. Standard medication might be associated with multiple factors from doctors, patients and society. It is necessary to make some steps to improve standard medication of antiretroviral therapy in PMTCT.

7.Effect of T-cell vaccination in murine experimental autoimmune hepatitis.

OBJECTIVETo investigate the effect of T-cell vaccination in murine experimental autoimmune hepatitis (EAH).
METHODSTo induce the EAH model, the syngeneic S-100 antigen emulsified in complete Freud's adjuvant was injected intraperitoneally to C57Bl/6 at day 1 and day 7. For T-cell vaccination, splenocytes were removed from animal 2 weeks after induction of EAH and from control animals, and activated in vitro by mitogen stimulation with Concanavalin A (Con A), then inactivated by mitomycin and injected at 5 10(7) cells per animal as T-cell vaccination at 14 and 7 days before first induction of EAH.
RESULTSThe histological grade and serum ALT level of the mice who received T-cell vaccination were decrease significantly, compared with that of model group (1.44+/-0.88 vs. 2.33+/-0.87, t=2.24, P<0.05; 63.0U/L+/-23.4U/L vs. 115.0U/L1+/-39.6U/L, t=2.37, P<0.01, respectively); there was no significant change in mice who received irrelevant T-cell vaccination.
CONCLUSIONT-cell vaccination with T cells from EAH animals, but not with irrelevant T cells, was able to protect animals from EAH.

8.Treatment of herpes zoster with cotton sheet moxibustion: multicentral randomized controlled trial.

OBJECTIVETo compare the therapeutic effects and safety of herpes zoster treated by the cotton sheet moxibustion combined with the plum-blossom-needle tapping therapy to western medicine.
METHODSThe multicentral random controlled method was adopted, 120 cases of herpes zoster were randomly divided into a comprehensive treatment group and a western medication group, 60 cases in each one. In the comprehensive treatment group, the tapping therapy of plum blossom needle was applied to the foci, corresponding Jiaji (EX-B 2), Quchi (LI 11), Waiguan (TE 5), Zusanli (ST 36), Taichong (LR 3), etc. Afterward, the cotton sheet moxibustion was given. In western medication group, Acyclovir ointment for external application, Valaciclovir Hydrochloride tablets and Vitamin B1 for oral administration were prescribed. In 7 days of treatment, the clinical symptom score, effect time, efficacy and safety were observed before and after treatment between two groups. The recurrence of disease was followed up for 1 month.
RESULTSIn the comprehensive treatment group, the cured rate and the total effective rate were 80.0% (48/60) and 98.3% (59/60) separately, which were significantly better than 45.0% (27/60) and 71.7% (43/60) in western medication group separately (P < 0.01, P < 0.05). After treatment, in either group, the scores of clinical symptoms such as pain rating index (PRI), Visual Analogue Scale (VAS), present pain intensity (PPI), skin lesion and sleeping score, etc. were all reduced significantly (P < 0.01, P < 0.05). The score reducing was much more obvious in the comprehensive treatment group (P < 0.01, P < 0.05). In the comprehensive treatment group, the time of pain stopping, the time of blister stopping, the time of scarring and the time of healing were all shorter tha tn those in western medication group (P < 0.01, P < 0.05). In the follow-up observation, 1 case (1.6%) was recurred in the comprehensive treatment group and 8 cases (13.3%) were in western medication group. In western medication group, 6 cases presented mild adverse reactions.
CONCLUSIONThe cotton sheet moxibustion combined with the plum-blossom-needle tapping therapy is advantageous at good efficacy, quick effect and short-time treatment for herpes zoster, which is apparently superior to the treatment with Acyclovir ointment for external application, Valaciclovir Hydrochloride tablets and Vitamin B1 for oral administration. It is the safe and effective therapy.

9.Influence of staring at mobile phone on static balance, plantar pressure gait characteristics and lower limb joints in young men

BACKGROUND:Smartphones are a double-edged sword,which not only bring convenience,but also result in excessive reliance.Especially the youth often gaze at their phones when standing or walking,which may affect their static standing balance and plantar pressure gait characteristics.OBJECTIVE:To investigate the influence of staring at mobile phones when standing or walking on the static balance,plantar pressure and gait characteristics in young men,thus informing its negative effect on gait characteristics and lower limb joints.METHODS:Twenty health young men were selected,the plantar pressure when normal standing and walking was measured using Loran plantar pressure analysis system (NO.MPS,Italy).After 20 minutes of rest,the same measurement was performed in all participants staring at mobile for 10 minutes during standing or walking.The relative indexes were compared between two conditions after data collection.RESULTS AND CONCLUSION:When young men staring at mobile phone for 20 seconds during standing,the fluctuation speed,displacement and track area were significantly larger than those during normal standing (P < 0.05);moreover,the contact area,pressure peak and average pressure of the first and second metatarsus in plantar area were significantly larger than those during normal standing (P < 0.05),while the parameters of heel lateral area were significantly lower than those during normal standing (P < 0.05).The stride and speed when staring at mobile phone during walking were significantly smaller than those during normal walking (P < 0.05);the support time of two feet and angle between feet were significantly larger than those during normal walking;the contact area,pressure peak and average pressure of the 4th and 5th metatarsus in plantar area and heel lateral area were significantly larger than those during normal walking.These results suggest that the gravity is less stable and more anterior when young men staring at mobile phone during standing than during normal standing.Staring at mobile phone during walking can alter the gait characteristics,resulting in longer support time,shorter stride,slower step speed,larger walking angle and higher lateral plantar pressure.
10.The event-related potentials study on attentional deployment in major depressive disorder

Objective:To investigate the attention deployment emotion regulation disorder and its neural mechanisms in major depressive disorder(MDD).Methods:Twenty-six MDD patients(MDD group) and twenty-nine healthy control individuals(control group) were enrolled in this study.The accuracy and reaction time were recorded in the attention deployment behavioral paradigm.Neuroscan 64 Brain Evoked Potentiometer was used to detect the N1 and P3 components of the above two groups of subjects.SPSS 16.0 software were used to analyze the data.Results:(1)In the accuracy of attention deployment paradigm, the group main effect was significant ( F=11.626, P=0.001), indicating that the MDD group (0.82±0.01) was significantly lower than the control group(0.89±0.01)( P=0.001). In the reaction time of attention deployment paradigm, the group main effect was significant( F=16.55, P<0.01), indicating that the MDD group (1 460.82±41.86)ms)was significantly higher than the control group(1 226.31±39.63)ms)( P<0.01). (2)In the event-related potential of attention deployment paradigm, the group main effect of N1 was not significant ( F=2.949, P=0.092), and the interaction between task and group was significant ( F=4.756, P=0.034), indicating that the N1 amplitude induced by the MDD group in the calculation task(-3.699±0.441)μV) was significantly smaller than that of the control group (-5.055±0.418)μV)( P=0.030). The group main effect of P3 was not significant ( F=1.165, P=0.285), and the interaction between task and image attributes was significant ( F=11.602, P=0.001), indicating that the P3 amplitude induced by negative images(2.757±0.438)μV) was significantly higher than that of neutral images (1.963±0.460)μV)( P=0.002). Conclusion:Patients with MDD have difficulty transferring attention in emotional strategies, and it occurs in the early stages.
