With the development of drug discovery, more and more candidate compounds need to be studied. Methods that can screen compound rapidly received significant attention. Parallel artificial membrane permeability assay (PAMPA) as a powerful tool has been applied to drug studies. It uses an artificial lipid membrane to mimic the barrier for drug permeability studies. This article introduces the establishment and characteristics of PAMPA, as well as its applications in screening compounds. It can be used as models (e.g. predicting the ability of compound in gastro-intestinal absorption, blood-brain barrier transportation and skin penetration) by changing the component of artificial lipid membrane. PAMPA has advantages in high throughput selection of valuable compound with low cost, versatile, low dose, and good reproducibility.

Objective To investigate the relationship between the gene polymorphisms of thromboxane A2 receptor (TXA2R) and cerebral infarction. Methods A genetic association study of one single nucleotide polymorphism (rs768963) in the human TXA2R gene was performed in 334 patients with cerebral infarction and 135 healthy controls by polymerase chain reaction-amplification and restriction fragment length polymorphism(PCR-RFLP) method. Results The levels of blood pressure,blood-fat and serum glucose were independent risk factors for cerebral infarction. No significant differences in the overall distribution of genotypes (T/T, T/C and C/C) were noted (P>0.05); however, significant differences in T or C gene frequency of rs768963 of TXA 2R gene between patients with cerebral infarction and noninfarction controls were found (P<0.05). Logistic regression analysis showed that no association between the mutant of rs768963 of TXA 2R gene and such factors as gender, age and levels of blood fat, blood pressure and serum glucose was noted. Conclusion The rs768963 of TXA 2R gene in human thromboxane A2 receptor may be a risk factor for cerebral infarction and patients carried C allele are much likely to have cerebral infarction.

This study was aimed to explore the level of glycosylated ferritin in the patients with secondary hemophagocytic lymphohistiocytosis (HLH) and its diagnostic significance. 29 suspected HLH patients from October 2007 to October 2008 were enrolled in the study, and 25 healthy subjects were selected as control. The 29 suspected HLH patients were divided into confirmed group (22 out of 29) and unconfirmed group (7 out of 29) according to HLH-2004 diagnostic criteria. The percentage of glycosylated ferritin in peripheral blood was determined by phytohemagglutinin adsorption assay. The results showed that the median level of total serum ferritin in patients of confirmed group (2897.6+/-1837.2 microg/L) was significantly higher than that in patients of unconfirmed group (653.1+/-249.1 microg/L) (p<0.01), and was also higher than that in controls (414.6+/-212.6 microg/L) (p<0.01). The median percentage of glycosylated ferritin in patients of confirmed group was significantly lower (17.0+/-4.2%) than that in patients of unconfirmed group (40.7+/-4.5%) (p<0.01) and was lower than that in controls (53.6+/-13.3%) (p<0.01). The sensitivity (86.4% vs 77.3%) and specificity (71.4% vs 42.9%) of glycosylated ferritin for the diagnosis of HLH were higher than that of total serum ferritin. In conclusions, glycosylated ferritin may be a helpful marker for the diagnosis of HLH.
