2.Curcumin inhibits telomerase activity of HeLa cells

Objective To investigate the inhibitory effect of curcumin on the telomerase activity of HeLa cells.Methods The growth inhibition rate of HeLa cells treated with curcumin was measured with MTT method.The expression of hTERT mRNA was studied using RT-PCR.Telomerse activity was examined by TRAP-ELISA.Results The growth of HeLa cells treated with curcumin was inhibited in concentration dependent manner and the expression of hTERT mRNA and the level of telomerase activity were down-regulated significantly.Conclusion Curcumin can inhibit telomerase activity of HeLa cells.The underlying mechanism might be related to the down-regulation of hTERT transcription.
3.Sequence analysis of p62~(dok) amino acid and cDNA clone

AIM:To analyse sequences of p62 dok amino acid and cDNA and to investigate p62 dok tyrosine phosphorylation and its relation with p21 ras GAP. METHODS:The purified p62 dok was extracted from CHO/IR cells. The peptide sequence of p62 dok was carried out on a high performance analyzer. PCR was performed with the primers designed from the sequence of p62 dok amino acid. Western blot and immunoprecipitation were used to identify tyrosine phosphorylation of p62 dok and the binding of p62 dok with p21 ras GAP. RESULTS:The p62 dok cDNA is a 1863 bp sequence and code 481 amino acid with 15 tyrosine residues and a putative pleckstrin homology domain. The p62 dok protein is rich in PxxP motif. The tyrosine-phosphorylated p62 dok can bind p21 ras GAP. CONCLUSION:Perhaps p62 dok is a new signaling molecule and play an important role in insulin signaling networks through RAS/MAPK pathway.

5.Nasal cavity volume and nasopharyngeal cavity volume in adults measured by acoustic rhinometry

Objective:To investigate the range of normal value of total nasal cavity volume(NV)and total na-sopharyngeal cavity volume (NPV)of healthy adults. Method: Six hundred and fifty-nine healthy adults andeighty-two adults of chronic rhinitis were measured with acoustic rhinometry. Result:The range of NV was29. 922~37. 481 cm3 and NPV was 29. 369~44. 159 cm3. Comparing healthy adults with adults of chronic rhini-tis,there was a significant difference in NV. Conclusion:Acoustic rhinometry suited for objective assessment ofthe nasal airways in adults. It was demonstrated that these data could provide available information for the studyof nasal physiology and pathophysiology,as well as for the diagnosis and judgement of therapeutic effectiveness ofnasal diseases.
6.Therapeutic Effect of Patent Doctus Arteriosus Closer for Low Body Weight Children

Objective To discusses the efficacy and safety of patent ductus arteriosus sealing for low body weight children. Methods From January 2008 to June 2012 in our hospital,32 cases (male14 and female 18) of PDA babies< 8 kg weight were given transcatheter closure treatment. The enrolled PDA babies aged 6 month-18 months, average (11 ±5.5 months),weighted (7.2 ±0.6) kg,aortic imaging showed the narrowest diameter of PDA was 1 ~ 8 mm (4.6 ±1.7) mm,descending aorta imaging was performed immediately and 30 min after transcatheter closure treatment. Echocardiography and X-ray radiography examination were performed 1 d,1,3, 6 months after treatment. Results 30 cases got successful patent ductus arteriosus sealing,all the successful patients were treated with domestic closer,and 1 case was complicated with the descending aorta stenosis and ventricular septal defect, and was transferred to surgerical treatment. 1 case of PDA gave up patent ductus arteriosus sealing as descending aorta stenosis caused by the closer. 2 cases were found a trace-residual shunt immediately after patent ductus arteriosus sealing. No serious complications were found in all patients during the follow-up period. Conclusions Application of domestic closer in treatment of PDA infants < 8 kg is a safe, effective and simple intervention treatment method with little trauma and fast recovery.
7.Effects of Posture and Phonation on Mallampati Test and Its Correlation with Improved Cormach-Lehane Score

Objective To evaluate the effects of various head and neck postures and phonation on oropharyngeal view(Mallampati score),and ex-plore the correlation with laryngoscopic view tested by the improved Cormach-Lehane score(MCLS). Methods Following the local ethics commit-tee approval and patients′informed consent to anesthesia,124 patients were enrolled for this study. Prior to anesthesia,these patients were placed in supine position with various head and neck positions for oropharyngeal structures visualization and evaluation,including two head positions(neu-tral and full extension),two tongue positions(in and out),and with and without phonation,according to the modified Mallampati test score(MMT). Following induction,laryngoscopic view scores according to MCLS were recorded,then the sensitivity,specificity,positive and negative predictive value and accuracy of the various MMT scores were calculated. Correlation coefficient(r)of MMT scores with MCLS were also studied. Results Mallampati score was decreased in all the postures of head full extension,tongue outside and phonation,which makes oropharyngeal structures to be better visualized. There is no correlation between MMT scores and MCLS. The sensitivity,specificity,positive and negative predictive value and accu-racy of the various MMT scores is most satisfied in the posture of head full extension,tongue outside and phonation. Conclusion During airway as-sessment in supine position,the best posture is head full extension,tongue outside and phonation.

9.Experimental Study of Nerve Growth Factor of Sciatic Nerve of Rats with Diabetes Peripheral Neuropathy Accommodated by Electro-acupuncture

Objective To explore the mechanism of the nerve growth factor (NGF) accommodated by electro-acupuncture in rats with diabetes peripheral neuropathy (DPN).MethodsThe rat DPN model was established with streptozotocin (STZ). The rats were randomly divided into the normal group, model group, western medicine group and electro-acupuncture group. The animals of the electrol-acupuncture group were treated with electrol-acupuncture on "Zusanli" and "Shenshu" after DPN model established. The nervous tissues of all animals were examined with the techniques of immunity and fluorescent quantitation PCR to detect the albumen and mRNA expression of NGF.ResultsThe albumen and mRNA expression of NGF of the animals in the model group, western medicine group and electro-acupuncture group were significantly lower than that in the normal group ( P<0.05~0.01), and the albumen and mRNA expression of NGF was up-regulation in the electro-acupuncture group after electro-acupuncture treatment and was significantly higher than that in the model group ( P<0.01).ConclusionElectro-acupuncture on "Zusanli" and "Shenshu" can up-regulate the albumen and mRNA expression of NGF, and improve reparation of the sciatic nerve in DPN animals.
10.Construction of eukaryotic expression plasmid of bFGF gene in rats and its expression in tenocytes.

The bFGF plays an important role in embryonic development of tendons and ligaments and in the healing of injuried tendons and ligaments. The eukaryotic expression plasmid of rat basic fibroblast growth factor (bFGF) gene was constructed in order to further investigate the bFGF function in molecular regulatory mechanism in the repair of tendons and ligaments and to provide the foundation for the clinical application. The cDNA fragments of bFGF were cloned from the skin of rats by RT-PCR, and recombinated to the pMD18-T vector. The cDNA encoding bFGF was cloned from the pMD18-T vector by RT-PCR, digested with restriction enzyme EcoR I Pst I and bound to eukaryotic expression plasmid pIRES2-EGFP to construct eukaryotic expression plasmid pIRES2-EGFP-bFGF. The pIRES2-EGFP-bFGF was transfected into the tenocytes by lipid-mediated ransfection technique. MTT test was used to detect the biological activity of bFGF in supernatants after the transfection. The expression of type I and III collagen genes was detected by using RT-PCR. It was verified that the pIRES2-EGFP-bFGF was successfully constructed, and its transfection into tenocytes could significantly enhance the biological activity of bFGF, and increase the expression of type I and III collagen mRNA, suggesting that pIRES2-EGFP-mediated bFGF gene therapy was beneficial to the repair of tendons and ligaments.
