Objective To explore the value of contrast-enhanced ultrasound(CEUS) in the diagnosis of hematoma and active hemorrhage of renal trauma. Methods Totally 28 patients with renal trauma were examined by conventional ultrasound and CEUS,respectively, including 24 cases caused after renal biopsy,4closed trauma. The detectability of renal hematoma and active hemorrhage with these two methods were compared. All patients were identified by CT or follow-up studies with ultrasound. Results The detectability of renal hematoma with conventional ultrasound and CEUS were 67. 86% (19/28), 92. 86%(26/28), respectively. There was statistically different for detectability in the diagnosis ( P <0.05), and the extent of hematoma was more obvious with CEUS. CEUS diagnosed 7 of 26 were renal hematoma with active hemorrhage, which were difficult to be detected with conventional ultrasound. For renal hematoma, the features of CEUS were no enhancement found in every phase; for renal hematoma with active hemorrhage, the contrast agents overflowed from injured blood vessels and formed irregular remarkable enhanced regions. Conclusions CEUS is useful in diagnosing hematoma and identifying the extent and active hemorrhage of renal trauma,in addition,CEUS is valuable in detecting complications after renal biopsy.

AIM To investigate the effect of 4-aminopyridine (4-AP) on ion channels of myocytes. METHODS L-type calcium channel and sodium channel currents were recorded in guinea pig single ventricular myocyte using whole-cell patch-clamp techniques. RESULTS 4-AP, 0.1, 0.5 and 1.0 mmol*L-1 were shown to inhibit L-type calcium channel currents (ICa,L) and sodium channel currents (INa) concentration-dependently. The percentage of inhibition were (11.6±1.7)%, (37.5±8.3)% and (54.5±6.9)% (P<0.01) respectively for ICa,L, and (22.1±14.3)% (P<0.05), (39.4±8.8)% and (62.3±6.8)% (P<0.01) respectively for INa. 4-AP 0.5 mmol*L-1 shifted the I-V curves of ICa,L and INa upwardly. CONCLUSION 4-AP blocked L-type calcium channel and sodium channels in guinea-pig ventricular myocytes concentration-dependently.

Objective To assess the differential diagnosis and characteristics of renal benign and malignant tumors with three-dimensional contrast-enhanced ultrasound (3D-CEUS). Methods Totally 68 patients with renal tumors were examined by conventional ultrasound and two-dimensional contrastenhanced ultrasound(2D-CEUS). 3D imaging was reconstructed from 2D imaging, the differential diagnosis of renal tumors with 3D-CEUS was analyzed by comparing with 2D-CEUS. All patients with renal tumors were proved by operational pathology. Results Eighteen patients with renal benign tumors mostly displayed equal or low enhancement, showed "slowly in and slowly out" with 2D-CEUS, while displayed regular peripheral and internal vessels with 3D-CEUS. Fifty patients with renal malignant tumors mostly displayed high enhancement, showed "rapidly in and rapidly out" with 2D-CEUS,displayed winding peripheral vessels and disordered internal vessels with 3D-CEUS. 3D-CEUS may display the vascular characteristics of tumors and showed superior imaging quality to 2D-CEUS ( P < 0.05). Conclusions 3D-CEUS can display the vascular characteristics of tumors and their spatial positions, it plays an important role in differential diagnosis between renal benign and malignant tumors.

OBJECTIVE To study the optimal reagent for regeneration of aptamer-coated piezoelectric quartz crystal chips and detect storage ability of aptamer-coated chips. METHODS Anti-IgE aptamers were immobilized on the gold surface of piezoelectric quartz crystal biosensor with biotin-avidin method. The reaction between anti-IgE aptamers and IgE was monitored in time on piezoelectric sensor. We tried to regenerate the sensor surface after binding IgE by rinsing the aptamer-coated chips with HCl, NaOH, EDTA, urea and formamide individually. Aptamer-coated chips were stored in PBS with 0.1% sodium azide and the stored chips were used to detect IgE in 35 days. RESULTS Of the five reagents, EDTA was the best one for regeneration of aptamer-coated chips and the sensor could retain 91.5% of the original detecting signals after five regeneration cycles. Moreover, the aptamer-coated chips could be stored in the binding buffer for 21 days without obvious loss of activity. CONCLUSIONS Compared with antibody-based piezoelectric sensor, aptamer-based sensor has lower cost, more regeneration cycles and longer time for the storage of chips. This series of experiments shows the superiority of aptamer detection.

OBJECTIVE To establish an aptamer-based piezoelectric sensor assay to detect human immunoglobulin E (IgE) and acquire the parameters about this kind of biosensor. METHODS Anti-IgE aptamers were immobilized on the gold surface of piezoelectric quartz crystal sensor with biotin-avidin method. The reaction between anti-IgE aptamers and IgE was monitored in time on piezoelectric sensor. Different concentrations of IgE were detected and the calibration curve of IgE was drawn. Non-specific response signals of BSA, IgG and IgE were monitored to show the detecting specificity of aptamer-based sensor. RESULTS For the aptamer-based sensor, the lowest limit of detection was 0.055mg/L and the linear range was 0.1-2.5mg/L. Non-specific signals of this sensor were less than 5% of specific signals of IgE at the same concentration. CONCLUSIONS The aptamer-based piezoelectric sensor can detect 5.5 ng IgE while the non-specific signals are very low. So this aptamer-based sensor is hopeful to be applied to clinical laboratory diagnosis.
