4.Effect of apoptosis and gp 130 expression by Cap preconditioning on rats of acute myocardial injured

Objective To study the influence of Captopril preconditioning on apoptosis and the expression of gp 130 in rats of acute myocardial injured. Method Wistar rats were randomly divided into three groups:control group, Iso group and Cap preconditioning group. Cardiomyocytes apoptosis was detected with TUNEL method. The expressions of Bcl-2 and Bax proteins in myocardium were tested by immunohistochemistry. The expression of gp 130 was tested with Western blot method. Results The index of cardiomyocytes apoptosis was decreased, the expressions of Bax proteins and gp 130 were decreased and the expression of Bcl-2 proteins was increased in Cap preconditioning group compared with Iso group (P<0.01 or P<0.05). Conclusion The expression of gp 130 could be decreased through Cap preconditioning, which can reduce the cardiomyocytes apoptosis.
5. Breeding of Dendrobium officinale var. 'Xianhu 2' and its characteristics

OBJECTIVE: To select and breed elite variety of Dendrobium officinale, thus to conserve the wild germplasms and promote the healthy developing of D. officinale industry. METHODS: System breeding method was employed for the breeding of new variety developed from wild germplasm oi Dendrobium officinale. Yield of fresh stems and leaves, quality characters including dried stem rate and raw polysaccharide content etc. were tested. ISSR molecular marker technique was used to distinguish genetic difference between new variety and other D. officinale materials. RESULTS: New elite variety 'Xianhu 2' was bred successfully with high polysaccharide content, fine adaptability, and high yield. 'Xianhu 2' had significantly higher yields of fresh stems-leaves and stems than D. officinale var. 'Yunnan Ruanjiao' and D. officinale var. 'Xianhu 1' and significantly higher dried stem ratio and raw polysaccharide content than 'Yunnan Ruanjiao' at 12, 24, and 36 m, respectively. ISSR analysis indicated that there were distinct differences in DNA level between 'Xianhu 2' and other D. officinale varieties and lines. D. officinale var. 'Xianhu 2' got its certificate for commerce releasing by Zhejiang Province in December 2011. CONCLUSION: 'Xianhu 2' is a new elite variety with good synthetic agronomic traits. Its breeding is beneficial for the cultivation of D. officinale.
6.Spleno-left adrenal vein shunt for portal cavernous transformation

Objective To evaluate the effect of spleno-left adrenal vein shunt for the treatment of portal hypertensive upper GI bleeding caused by portal vein cavernous transformation in children.Methods Spleno-left adrenal vein shunt was performed in 8 children with portal hypertension due to cavernous transformation.The clinical data was reviewed.Results Portal vein pressure decreased significantly from (30±11)mm Hg to(22±7) mm Hg after shunt.There was no mortality perioperatively and during the follow-up.There were no recurrent hemorrhage nor hepatic encephalopathy occurring in the follow-up and all the children have normal intelligence and normal liver function though blood ammonia level increased significantly from(18±7)μmol/L to (60±17)μmol/L in 4 cases.In 7 cases in which preoperative whole blood cell count significantly decreased,the postoperative WBC,RBC,Hb and PLT was (7.64 ±4.46)×10~9/L,(4.54±0.97)×10~(12)/L,(133±5) g/L and (355.40±107.36)×10~9/L respectively (all P <0.05).In one case suffering from preoperative low PLT count the postop PLT reached 333×10~9/L,which was significantly higher than that preoperatively.Esophageal varices ameliorated in 6 cases.No stenosis of anastomotic stoma and thrombosis developed.Conclusion Spleno-left adrenal vein shunt is an effective procedure to treat portal vein cavernous transformation induced portal hypertension in children.

8.The Mechanisms of Tcrolimus on the Migration of Langerhans' Cells

Objective To investigate the mechanisms of tacrolimus on the migration of Langerhans′ cells. Methods The concentration of MCP-1 in the cultured supernatants of HaCaT cell treated with tacrolimus was determined by ELISA. The expression level of MCP-1 mRNA in HaCaT cell treated with tacrolimus was studied by RT-PCR method. Results The concentration of MCP-1 in HaCaT cell treated with 625 ~ 5 000 ng/mL of tacrolimus was significantly decreased. The expression level of MCP-1 mRNA was also markedly decreased. Conclusions Tacrolimus may probably decrease the expression of MCP-1 in keratinocytes and suppress the chemotactic ability of Langerhans′ cells precursor and Langerhans′ cells, thus inhibiting or diminishing certain local immunoreaction.
9.Nanoknife ablation of the pancreas in vivo experiment

Objective Nanoknife,also called irreversible electroporation,is a new technique of tissue ablation.Short,microsecond electrical pulses with high voltage are applied to the cell membrane,causing pores to form within the membrane and finally leading to cell death.The current study was to investigate the efficacy and safety of the nanoknife in the ablation of the healthy pig pancreas.Methods Three healthy pigs underwent open pancreatic tissue ablation with nanoknife,and blood leukocytes and amylase were detected before and after treatment.Three pigs were sacrificed and gross specimens were collected on day 5,day 10 and day 15 after the procedure,respectively.HE staining and TUNEL staining were conducted and tissue,cellular and subcellular structures were observed under the ordinary microscope and transmission electron microscopy.Results Three experimental pigs recovered well after the procedure.No significant adhesions were found surrounding the pancreatic tissue,and the ablation zone was slightly harder.Transiently increased leukocyte count and amylase level were observed after the ablation,which decreased to the normal level on day 3 after treatment.Under light microscope,the pancreatic tissues in ablation zone appeared to be significantly different from the normal surrounding regions,with more cell death and more apoptotic cells detected by TUNEL staining.The subcellular structure changes also changed under electron microscope.But the main pancreatic duct and its large branches,together with arteriovenous distributions did not change much.Conclusions Nanoknife pancreatic tissue ablation can induce irreversible damage.In the ablation area,pancreatic duct and vascular structures are kept intact.Within a reasonable voltage range and appropriate electrical pulses setting,nanoknife ablation is safe in vivo experiment.
