3.Acquiring and analyzing of ornithine aminotransferase gene of Schistosoma japonicum

Objective To screen and identify the novel genes from the adult worm cDNA library of Schistosoma japonicum by the expressed sequence tags (EST) strategy.Methods The cDNA clones were selected randomly and the ESTs were obtained.The novel gene was searched for homologue identity with NCBI blast programmer.The homologue of the two sequences with a high identity was compared at amino acid and nucleotide level with the BLAST programmer,and the analysis of protein was carried out with the pcgene software. Results There was a novel gene of Schistosoma japonicum which included 1 677 bp coding for 424 amino acid residues, and it was given the accession number of sequence in Genbank(AY336497).The cDNA sequence was homologous to the known ornithine aminotransferase gene of Xenopus laevis. The identity of amino acid sequence was 66%. The academic pI was 8.52, and the antigen determinant was probably from 795 to 846 on the cDNA sequence. Conclusion EST strategy is an effective measure to discover new genes of Schistosoma japonicum. The novel gene is homologous to the ornithine aminotransferase gene of Xenopus laevis.This study is helpful to further sequential and functional search of the gene.[

5.Relationship between age and refractive predictability of excimer laser in situ keratomileusis

Objective To explore the relationship between age and refractive predictability of excimer laser in situ keratomileusis (LASIK)for the correction of myopia.Design Retrospective case series.Participants 362 patients(703 eyes)who had performed LASIK.Methods According to the age,patients were divided into four groups(≤30y,31~34y,35~39y and≥40y).Their preoperative and postoperative refractive powers examined with automatic refactor as well as manifest optometry were analysed and followed up from 6 months to 2 years.According to pre-operative refractive power,patietns were divided into low and moderate myopia group(≤-6.00D), high myopia group(-6.25~-12.00D)and super-high myopia group(≥-12.25D).The eyes with refractive power at post-operative 2 years≥-1.00D were regressive eyes.Main Outcome Measures Refractive error before surgery,intended residual refractive power and 3,6, 12, 24 months post-operative refractive powers.Results There were no significant differences between intended residual refractive powers and refractive powers at post-operative 6 month in 3 groups(
6.INDUCTION OF PROTECTIVE IMMUNITY AGAINST SCHISTOSOMA JAPONICUM WITH pcDNA3.1(+)/MLP NUCLEIC ACID VACCINATION

Objective To investigate the immune efficacy of nucleic acid vaccination in rabbits against Schistosoma japonicum with pcDNA3 1(+)/MLP(SJP) Methods The 24 New Zealand rabbits were randomly divided into 2 groups. The rabbits of experimental group were vaccinated by each quadriceps muscle of leg with pcDNA3 1(+)/MLP nucleic acid vaccination and the control group rabbits were vaccinated with pcDNA3 1(+). Each rabbit was immunized four times with 2 weeks interval. The rabbits were challenged 2 weeks after final DNA boosting by percutaneous infection with cercariae. Sixty days after infection the rabbits were sacrificed, the livers were investigated, and the worms and eggs in livers were counted. Blood sera were collected from rabbits and investigated. Results In the experimental group,the egg reduction rate was 28 10%. The rabbits of experimental group produced IgA, IgG 1, INF-?. Conclusion DNA vaccination with pcDNA3.1(+)/MLP could induce partial protective immunity against Schistosoma japonicum in rabbits.

8.Phytoestrogen Genistein induces osteoblastic differentiation of mouse bone marrow-derived mesenchymal stem cells through p38 MAPK

Aim To investigate the role of p38 mitogen-activated protein kinases(MAPKs) in genistein-induced osteoblastic differentiation of mouse bone marrow-derived mesenchymal stem cells(BMSCs).Methods Mouse BMSCs cultured in phenol red-free ?-MEM containing 10% V/V FBS,were added ?-glycerophosphate and ascorbic acid for inducing osteoblastic differentiation,and treated with 0.01~1 ?mol?L~(-1) genistein and/or SB203580 and PD98059.The temporal sequence of osteoblastic differentiation in BMSCs cultures was assayed by measuring alkaline phosphatase activity(ALP) and calcium deposition.The MAPK phosphorylation level was detected by Western-blot.Results Genistein(0.01~1 ?mol?L~(-1)) showed a dose-dependent effect on osteoblastic differentiation as evidenced by increased alkaline phosphatase(ALP) activity and calcium deposition in mouse BMSCs cultures.Genistein(1 ?mol?L~(-1))-induced osteoblastic differentiation of BMSCs was diminished by p38 MAPK inhibitor but not the p44/42 MAPK inhibitor.The effects of Genistein were associated with rapid and sustained activation of p38 MAPK in the BMSCs cultures,which was blocked by SB203580(1 ?mol?L~(-1)).In contrast,Genistein treatment resulted in inactivation of p42/44MAPK,which was further attenuated by PD98059(25 ?mol?L~(-1)).Conclusion p38 MAPK plays an important role in genistein-induced osteoblastic differentiation of mouse BMSCs cultures.
9.Prenatal ultrasound diagnosis and analysis of fetal limb deformities

Objective To explore the ultrasonographic characteristics and methodology characteristics of fetal limb deformities . Methods A retrospective analysis was made on 6 336 pregnant women accepting systematic prenatal ultrasonography during the period of May 2010 to May 2012 in the General Hospital of Chengdu Military Region .The long bones of the upper and lower limbs were scanned by using the systematic continuous sequence approach (SCSA) and by adding the arc rotaryprocess for observing the hands and feet ,fetal limbs morphology ,structure ,posture and movement ,etc .were dynamically observed .The suspected position or fetal limb deformities were examined with three-dimensional(3D) ultrasonography as the supplementary .The prenatal ultrasound examination results of fetal limb deformities were analyzed .Results Among 6 336 pregnant women ,20 cases of fetal limb deformi-ties were confirmed by induced labor or after birth ,18 cases were diagnosed by the prenatal ultrsound ,including 5 cases of short limbs deformity ,3 cases of fatal dwarf ,1 case of phocomelia ,1 case of double radiuses absence ,1 case of unilateral hand missing ,1 case of double shank absence ,2 cases of polydactyly ,3 cases of foot varus and 1 case of overlapping finger .2 cases of missed diagno-sis were multiple fingers and overlapping finger .Conclusion The abnormality of terminal fingers(toes) number and posture is easy to be missed diagnosis ,which are the difficulty and the key point of examination .
10.Transcatheter arterial embolization for traumatic hepatic hemorrhage

Objective To investigate the clinical efficacy,indications and complications of transcatheter arterial embolization for the treatment of traumatic hemorrhage of liver.Methods Retrospectively analyzed 33 cases of traumatic hemorrhage of liver admitted to West China Hospital from May 1996 to May 2006.Twenty-four cases underwent hepatic arterial angiography and were diagnosed as hepatic arterial pseudoaneurysms and followed by superselective embolization of the feeding vessel with gelatin sponge and/or spring coils.Results Embolization was succeeded technically in 28 pseudoaneurysms of 24 cases.Rebleeding occurred in 2 cases and a second embolization was performed with one recovered uneventfully.The other died of rebleeding and severe abdominal infection one week after the second embolization.Fever unrelated to embolization occurred in 9 cases.Conclusion Transcatheter arterial embolization is safe,prompt and effective for the treatment of hemorrhage due to hepatic injury.(J Intervent Radiol,2007,16:226-228)
