1.Effect of heat-sensitive moxibustion plus psychological intervention on SP, 5-HT and quality of life in patients with irritable bowel syndrome

Objective: To observe the effect of heat-sensitive moxibustion plus psychological intervention on serum contents of substance P (SP) and 5-hydroxytryptamine (5-HT), as well as the quality of life (QOL) in patients with irritable bowel syndrome (IBS). Methods: A total of 120 IBS patients were divided into three groups by random sampling method, 40 cases in each group. The heat-sensitive moxibustion group received heat-sensitive moxibustion, the psychological intervention group received psychological intervention, while the observation group received both methods. The therapeutic efficacy was evaluated after 3 courses of treatment. Results: After treatment, the therapeutic efficacy of the observation group was better than that of the heat-sensitive moxibustion group and the psychological intervention group (P<0.01), and the heat-sensitive moxibsution group was better than the psychological intervention group (P<0.05). In comparing the bowel symptom scale (BSS), the observation group was better than the other two groups (P<0.05), and there was a significant difference between the heat-sensitive moxibustion group and the psychological intervention group (P<0.05). In the comparison of QOL, the observation group was significantly superior to the other two groups (P<0.05), and there was a significant difference between the heat-sensitive moxibustion group and psychological intervention group (P<0.05). In comparing serum SP and 5-HT, the observation group was markedly better than the other two groups (P<0.05), and there were significant differences between the heat-sensitive moxibustion group and psychological intervention group (P<0.05). The self-rating anxiety scale (SAS) and self-rating depression scale (SDS) were used to estimate the mental state of the three groups, revealing that the observation group was better than the heat-sensitive moxibustion group and the psychological intervention group (P<0.05), and there were significant differences between the latter two groups (P<0.05). Conclusion: Heat-sensitive moxibustion plus psychological intervention can regulate the levels of SP and 5-HT, improve BSS, SDS, SAS and QOL in treating IBS.
2.Rapid quantification of total nitrogen and end-point determination of hide melting in manufacturing of donkey-hide gelatin.

Hide melting presents itself as one of the most critical processes in the production of donkey-hide gelatin. Here a NIR-based method was established for the rapid analysis of in-process hide melting solutions as well as for end-point determination of this process. Near infrared (NIR) spectra of hide melting solutions were collected in transflective mode. With the contents of total nitrogen determined by the Kjeldahl method as reference values, partial least squares regression (PLSR) was employed to build calibration models between NIR spectra and total nitrogen. Model parameters including wavelength range and PLS factors were optimized to achieve best model performance. Based on the contents of total nitrogen predicted by calibration model, end point of hide melting was determined. The constructed PLS model gave a high correlation coefficient (R2) of 0.991 3 and a root mean square error of prediction (RMSEP) of 0.807 g x L(-1). With the predicted total nitrogen and predefined limit, decisions concerning the proper times of melting were made. This research demonstrated that NIR transflectance spectroscopy could be used to expeditiously determine the contents of total nitrogen which was subsequently chosen as the indictor for determining the end-point of hide melting. The proposed procedure may help avoid unnecessary raw material or energy consumption.

zoonoses, which is a serious threat to human health. Nowadays, the relationship between T. gondii infection and
neuropsychiatric diseases has attracted researchers' attention increasingly. T. gondii infection is related to the pathogenesis of
many neuropsychiatric diseases by affecting the nervous system, such as schizophrenia, depression, Alzheimer's disease, and so
on. This review will focus on the relationship between T. gondii infection and neuropsychiatric diseases and summarizes the
possible mechanisms of disorders resulting from T. gondii infection. It is expected that the study on the related pathogenic
mechanism of T. gondii will lead to new therapeutic directions and feasible solution for the clinical treatment of
neuropsychiatric diseases caused by T. gondii infection.
4.Degradation of refuse in hybrid bioreactor landfill.

OBJECTIVETo explore the process of refuse decomposition in hybrid bioreactor landfill.
METHODSThe bioreactor landfill was operated in sequencing of facultative-anaerobic and aerobic conditions with leachate recirculation, pH, COD, and ammonia in the leachate and pH, biodegradable organic matter (BDM), and cation exchange capacity (CEC) in refuse were detected.
RESULTSCEC increased gradually with the degradation of refuse, which was negatively correlated with BDM. COD and ammonia in the leachate was declined to 399.2 mg L(-1) and 20.6 mg N L(-1), respectively, during the 357-day operation. The respective concentrations of ammonia and COD were below the second and the third levels of current discharge standards in China.
CONCLUSIONThe refuse is relatively stable at the end of hybrid bioreactor landfill operation. Most of the readily biodegradable organic matter is mineralized in the initial phase of refuse degradation, whereas the hard-biodegradable organic matter is mainly humidified in the maturity phase of refuse degradation.

The content of benzyl isothiocyanate (BITC) which as the enzymatic hydrolysis product of benzyl glucosinolate through thioglucosidase was determined by HPLC. The content of benzyl isothiocyanate (BITC) which as the enzymatic hydrolysis product of benzyl glucosinolate through thioglucosidase was determined by HPLC. The chromatography condition was as follows: Kaseisorb LC ODS 2000 (4.6 mm x 150 mm, 5 min) column with the mobile phase of acetonitrile(A)-water( B) under gradient elution (0-5 min, 3%-8% A; 5-9 min, 8%-48% A; 9-23 min, 48%-62% A; 23-28 min, 62%-99% A); the flow rate was 1.0 mL x min(-1) with 10 microL injection volume; detection wavelength was 246 nm and temperature of column was 40 degrees C. The content of benzyl glucosinolate was in the range of 10.76-17.91 g x L(-1). The method is simple, accurate and good reproducibility which can be used for the determination of benzyl glucosinolate in Lepidium meyenii, effectively.

8.Construction of T vectors based on Xcm I recognition site and optimization of PCR fragments for ligation.

OBJECTIVETo construct T vectors based on Xcm I recognition site and optimize the PCR fragments for its ligation.
METHODSWe firstly cloned the human histone H4 cDNA containing one Xcm I recognition site at both its 5' and 3' end into pCDNA 3.0 vector and then generated T vector with pCDNA 3.0 backbone by cutting the recombinant plasmid with Xcm I. To increase the ligation efficiency, the primers were firstly phosphorylated before DNA fragments amplification and then the PCR products were treated with Taq DNA polymerase and dATP after PCR amplification. Two DNA fragments with the length of 312 bp and 1 329 bp were ligated to it and the ligation mixture was transformed into E. coli DH5α competent cells and the positive rates of the transformants were evaluated by PCR and DNA agarose gel electrophoresis.
RESULTSOur results showed that the T vector produced by our method could ligate to the target DNA fragments with high efficiency. Besides, the phosphorylation state of the primers used for PCR amplification is also an important factor determining the cloning efficiency. What's more, as for longer DNA fragments, retreatment with Taq DNA polymerase and dATP after PCR amplification and purification could improve the ligation efficiency significantly.
CONCLUSIONOur protocol may overcome the dependence on blue/white screening to get positive clones and provide a potent way to generate T vectors and ligate them to the target PCR fragment.

9.Cardiopulmonary protection of ischemic postconditioning in cardiac surgery in children with tetralogy of Fallot.

OBJECTIVEIschemic postconditioning effectively minimizes the ischemic/reperfusion injury, and the large series of case reports on its protective effects in cardiac surgery are limited. A randomized trial was conducted to investigate the effect of ischemic postconditioning on cardiopulmonary protection in children undergoing cardiac surgery for tetralogy of Fallot.
METHODSOne hundred and five-children with tetralogy of Fallot undergoing surgery were randomly assigned to control (n=58) and ischemic postconditioning groups (n=47). Ischemic postconditioning was performed by intermittent aortic clamping after reperfusion. After surgery, the duration of intensive care unit (ICU) stay, capacity of blood transfusion, hemodynamics, inotropic scores, respiratory function, and release of blood lactate were assayed.
RESULTSThere was a significant decrease in the ICU stay in the postconditioned group compared with the control group (37+/-21 hrs vs 54+/-26 hrs; P<0.05 ). The capacity of blood transfusion (308+/-230 mL vs 526+/-515 mL; P<0.05) and the inotropic scores (5.9+/-5.0 vs 10.3+/-7.7; P<0.05) in the postconditioned group were significantly reduced compared with those in the control group. Blood lactate contents in the postconditioned group was significantly lower that those in the control group 1, 3, 6, 9, 12 and 20 hrs after surgery. The postconditioned group showed more improved hemodynamics and respiratory function than the control group.
CONCLUSIONSIschemic postconditioning may provide clinical benefits with respects to myocardial and pulmonary protections in children undergoing repair for tetralogy of Fallot.

The incidence of deacclimatization to high altitude syndrome (DAHAS) prevailed up to 80% in highland troops, and 100% in manual workers, and severe DAHAS could significantly affects patients' health, work and life. So it is imperative to develop effective prevention and treatment measures for DAHAS. The present review analyzes effective prophylactic and therapeutic measures against DAHAS, implemented at our hospital.
