1.Effect of Qidan Granule on PMC Derived Peptide Content and Structure of Hippocampal CA1 Region in Microwave Radiated Rats.

OBJECTIVETo explore the protection of high intensity microwave radiation on hypothalamo-pituitary-adrenal axis (HPAA) activity and hippocampal CA1 structure in rats and the protectiveeffect of Qindan Granule (QG) on radiation injured rats.
METHODSTotally 48 Wistar rats were randomlydivided into 8 groups, i.e., the normal control group, post-radiation day 1, 7, and 10 groups, 7 and 10days prevention groups, day 7 and 10 treatment groups, 6 in each group. Rats in prevention groups wererespectively administered with QG liquid (1 mL/100 g, 4. 75 g crude drugs) for 7 days and 10 days bygastrogavage and then microwave radiation. Then preventive effect for radiation injury was statisticallycalculated with the normal control group and the post-radiation day 1 group. Rats in treatment groupswere firstly irradiated, and then administered with QG liquid (1 mL/100 g, 4.75 g crude drugs). Finally preventive effect for radiation injury was statistically calculated with the normal control group, post-radiation day 7 and 10 groups. Contents of corticotrophin releasing hormone (CRH), beta endorphin (beta-EP), adrenocorticotropic hormone (ACTH), and heat shock protein 70 (HSP70) were detected. Morphological changes and structure of hippocampal CA1 region were observed under light microscope.
RESULTSCompared with the normal control group, contents of CRH and beta-EP significantly decreased in each radiation group. Serum contents of ACTH and beta-EP significantly increased in post-radiation day 1 and 7 groups (P < 0.05). Compared with radiation groups, beta-EP content in serum and pituitary significantly increased, and serum ACTH content significantly decreased in prevention groups (P < 0.05). Pituitary contents of CRH and beta-EP significantly increased in prevention groups. Serum contents of ACTH, beta-EP, and HSP70 were significantly lower in day 7 treatment group than post-radiation day 7 group (P < 0.05). Morphological results showed that pyramidal neurons in the hippocampal CA1 region arranged in disorder, with swollen cells, shrunken and condensed nucleus, dark dyeing cytoplasm, unclear structure. Vessels in partial regions were dilated with static blood; tissues were swollen and sparse. In prevention and treatment groups pathological damage of hippocampal CA1 region was obviously attenuated; neurons were arranged more regularly; swollen, pycnotic, or deleted neuron number were decreased; vascular dilatation and congestion was lessened.
CONCLUSIONQG could affect HPAA function and activity of high intensity microwave radiated rats, showing certain preventive and therapeutic effects of microwave radiated rats by adjusting synthesis and release of partial bioactive peptides and hormones in HPAA, improving pathological injury in hippocampal CA1 region.

3.Effect of blocking RAGE on macrophage infiltration of diabetic wound healing

Objective·To investigate the influence of blocking receptor for advanced glycation end products (RAGE) on macrophages infiltration in diabetic wound healing. Methods·Ninety-six male C57BL/6J mice (8-week-old) were divided into diabetic group (n=72) and normal group (n=24) randomly. Diabetic mice were induced by streptozocin multiple intraperitoneal injection. One full-thickness excisional wound (diameter of 9 mm) was created by a sterilized punch. Diabetic mice were divided into 3 groups in which different topical treatments were applied to the wounds. Anti-RAGE antibody were applied in group R, rabbit IgG applied in group I, normal saline applied in group C. Normal mice were applied with saline topically (group N). All treatments were repeated on day 3 and day 7 after wounded. The wound healing conditions were observed. The wound and surrounding tissues from animals in each group were excised on day 1, 3, and 7 after wounded. Immunohistochemistry was utilized to investigate the changes of macrophages infiltration in quantity. Macrophages were also analyzed with respect to morphology by transmission electron microscopy (TEM). Results·① The wound closure ratio of group R was higher than those of group C and group I on day 14 after being wounded (P=0.000). ② On day 1, the numbers of macrophages in group R and group N were both bigger than those of group C and group I, but smaller on day 14 (P=0.000). ③ The morphological characters of macrophages also existed great differences under TEM. Conclusion·Number and morphology of macrophages are both abnormal in diabetic wound in a RAGE pathway depending manner. Based on macrophages, it suggests that impaired healing of diabetic wound is closely related to RAGE pathway.
4.Design of remote diabetes management system based on dynamic blood glucose monitoring

Objective To design a remote diabetes management system based on dynamic blood glucose monitoring to solve the problems of traditional diabetes patient management in glucose data manual collection,unidirectional information flow and out-hospital glucose management.Methods A remote diabetes management system was developed with Java EE distributed microservices-based architecture and front-end and back-end separation mode,which used Vue for the front-end user interface design and IntelliJ IDEA development tools supporting Java language for the backend.The system developed was composed of a doctor App,a patient App and a background management terminal.The doctor App had three modules for user management,data viewing and intervention follow-up,the patient App had three modules for data monitoring,event recording and doctor-patient communication and the background management terminal had three modules for user management,data management and device management.Results The system developed realized individualized intervention and long-term glucose control based on real-time data acquisition of patient glucose fluctuation,diet,medication,exercise and other conditions.Conclusion The system developed can be used for the centralized online and offline management for in-hospital and out-hospital diabetes patients,which reduces the incidence of adverse events due to high or low blood glucose and improves the efficiency of medical staffs.[Chinese Medical Equipment Journal,2023,44(9):42-48]

Objective To analysis the effects on the growth of rats by repeated restraint in dermal exposure test. Methods SD rats in the restraint group was bound for 6 hours per day for 91 days according the way by dermal exposure, while SD rats in the control group didn't receive the treatment. Clinical signs, body weight and food consumption changes were observed for 91 days. When the study was terminated, hematology, clinical biochemistry, urinalyses, gross necropsy, and histopathology were carried out. Statistical methods such as the generalized estimating equation were used to compare the differences between two groups. Results The statistical results of generalized estimating equation showed that there was an interaction between the group and test time for male and female rats in body weight changes (P<0.05), and the body weight of male rats in the restraint group was lower than the control group (P<0.05) . Further analysis showed that for male rats there was significant difference between groups since the forth week (P<0.05), and the interaction was found between groups and test time (P<0.05) . For female rats, the interaction was found since the eighth week between the group and test time (P <0.05) .There was no significant differences in other parameters between two groups (P>0.05) . Conclusion Repeated restraint during dermal exposure affected the body weight gain of rats, and the sensitivity of male rats was higher than that of female rats.
7.Investigation of prediction and prevention status for pressure ulcers in comprehensive hospitals

Objective To analyze the status of prediction and prevention for pressure ulcers in comprehensive hospitals in order to get suggestions for pressure ulcers.Methods Using consensus investigating times,tools,ways and procedure flows and judgment criteria,and organizing 457 nurses who trained and passed examination to do a cross-sectional investigation for condition of prediction and prevention for pressure ulcers in 12 comprehensive acute care hospitals.Data were calculated by SPSS 16.0 statistic software.Results Totals of 39 952 cases run into the database,the prevalence of pressure ulcers was 1.579% and the incidence of hospital-acquired pressure ulcers was 0.628 % (251/39 952).Out of patients with Braden scale ≤ 16 points totally 4 710 cases,the first assessment rate was 50.235% and reassessment rate was 56.645%.The first assessment rate of 380 patients found out pressure ulcers on admission was 87.38% and reassessment rate was 90.842%.The reassessment rate of 251 patients with hospital-acquired pressure ulcers was 95.219%.The first assessment rate of 1 094 critical care patients on admission was 94% and reassessment rate was 69.104%.The application rate of support surfaces or devices on beds and chairs for 4 710 patients with risk of pressure ulcers was 61.805%and 3.652%,respectively.The regular changing position rate on beds and chairs for 4 710 patients with risk of pressure ulcers was 75.138% and 7.834% respectively.The difference of Braden scores by clinical nurses and trained wound care team nurses was significant [(14.01 ± 3.54) vs (3.29 ± 3.54) ;t =3.221,P < 0.01].Pressure ulcer risk patients use decompression pad status and timing change positions comparative differences were statistically significant (P < 0.01).Conclusions The results of prediction and prevention for pressure ulcers in twelve comprehensive hospitals did not meet the guidelines' aim.It is important to set up the organized frame and train different professional nurses for preventing pressure ulcers,and enhance clinical nurses' recognition to prevent pressure ulcers,check and guide them regularly.
8.Transplantation of autologous peripheral blood stem cells for the treatment of lower limb arteriosclerosis obliterans.

OBJECTIVETo evaluate the clinical efficacy of mobilized autologous peripheral blood stem cells (PBSC) transplantation in a 48 years old patient with lower limb arteriosclerosis obliterans (ASO).
METHODSrhG-CSF 600 micro g/d for 5 days to mobilize stem cells. On the fifth day, PBSC were collected with a Version 4 blood-cells separator. Three hours late, the PBSC were intramuscularly injected into the ischemic areas of the two lower limbs (3 x 10(9) cells per limb). The clinical and laboratory findings were monitored every week for 3 months. Forty-four days after the implantation, left lower limb with severe ASO was given an additional implantation of the same number of cells as the first time.
RESULTSThe peripheral blood CD(34)(+) cells were increased from 0.18% to 0.75% after 5 days of rhG-CSF mobilization. Three months after the first stem cell transplantation, severe pain lameness, local cool-feeling and ulcer were improved, and ABI increased from 0.49, 0.69 to 0.50, 0.85, the amplitude of blood flow and laser Doppler blood perfusion were also significantly improved (P < 0.01). At the same time, digital subtraction angiographic scores for new collateral vessel formation were showed as + 3(rich). No related complication or adverse effect were observed during the 3-month observation.
CONCLUSIONTransplantation of mobilized autologous PBSC might be a simple, safe, and effective method for the treatment of ASO.

9.Preparation of antibody against mouse UPF1 and the expression of UPF1 in adipose cell differentiation

Objective To prepare polyclonal antibodies against mouse UPF1 protein and to investigate the expression of UPF1 protein during adipocyte differentiation. Methods UPF1 protein expression vector was constructed to prepare and purify rabbit UPF1 antibody. The differentation of 3T3-L1 cells was induced and the expression of UPF1 was detected by CoIP. Results 1)High specific mUPF1 polyclonal antibody was prepared and the titer of this anti-body reached 640 000;2)The expression of UPF1 protein did not change during adipogenesis;3)In the process of adipocyte differentiation,interaction of UPF1 and UPF2 was increased. Conclusions 1)The polyclonal antibodies prepared by using 550 amino acids at the C terminal of mUPF1 protein could effectively recognize intact mUPF1 pro-tein;2)The interaction of UPF1 protein with UPF2 protein during adipocyte differentiation is enhanced.

OBJECTIVETo investigate the prevalence of domestic violence (DV) in Hunan.
METHODSUsing a multi-stage sampling strategy, 9451 households involving 32 720 persons in urban, rural and industrial areas in Hunan, China were studied. Multiform clue investigation and face-to-face interviews were combined to investigate the prevalence of DV.
RESULTSA lifetime prevalence of DV was reported by 1533 households (16.2%). A total of 1098 households (11.6%) reported at least one incident of DV in the previous year. Both lifetime and 12-month prevalence of DV varied significantly by geographic setting (P < 0.01). The lifetime prevalence abuse rates were: spousal 10.2%, child abuse 7.8%, and elder 1.5%. With regard to household structure, the lifetime prevalence of DV was highest among those remarried families (21.0%), followed by married couples with one child and extended families with several generations living together (20.1% and 20.0%, respectively). The highest rate of spousal abuse was found among remarried families (14.7%), while child and elder abuse was most prevalent among extended families (12.4% and 4.1%, respectively).
CONCLUSIONSThe findings suggested that although the prevalence of DV in Hunan was modest compared to Western countries, it remained a serious public health problem affecting over 1 in 10 households. Furthermore, the prevalence of various types of DV varied by geographic setting and family structure, suggesting that diverse geographic setting and family constellations carried different risk and protective features.
