1.Application of gene-knocking out mice in the research of atherosclerosis

ApoE and LDL-R knocking out mouse may spontaneously develope atherosclerosis and so is applied in the research of atherosclerosis. These mouse model, the model generated by gene targeting, like IL-1, C-reactive protein, scavenger receptor, IgG Fc,C1q and CD44, play important role in research of inflammatory factor and immune factor on atherosclerosis.
2.Analysis of coronary flow reserve of myocardial bridging in the left anterior descending by coronary flow imaging

Objective To evaluate the accuracy of coronary flow velocity reserve(CFVR) measured by color Doppler coronary flow imaging(CFI) in the patients with myocardial bridging(MB) of the left anterior descending (LAD) coronary artery and to analyze coronary hemodynamic changes and clinical significance of the LAD MB. Methods Consecutive 110 patients with suspected or known coronary disease were examinated by coronary angiography(CA), coronary intravascular ultrasound(IVUS) and intracoronary Doppler(ICD). Of these patients, 11 cases were diagnosed as MB of LAD. The LAD flow were measured by CFI at baseline and hyperemic conditions (intravenous administration of adenosine 140 μg·kg-·min-)and CFVR were calculated. Compared with the measurements of ICD, the accuracy of CFI was analyzed. Nine normal cases served as control group. Results The prevalence of MB was 10% in this study. Seven eases presented with stable angina. MB located in the middle segment of LAD in 7 cases and in the middle-distal segment of LAD in 2 cases. The ratio of LAD stenosis was greater than 80% in 2 cases, one of them simultaneously combined with circumflex stenosis(90 %). CA showed no significant stenosis in 9 cases (MB group) in the LAD and IVUS showed no plaque. Flow velocities were measured by CFI in the middle-distal LAD in nine patients without significant stenosis. CFVR calculated by average peak velocity well correlated with those measured by ICD (regression equation: Y=0.87 X+0.39; correlation coefficient: r=0.81,P<0.01). The coronary flow velocities in hyperemic conditions were significantly lower in MB group than in control group. CFVR were also significantly lower in MB group than in control group. Conclusions CFI is an accurate method for assessment of CFVR. Decreased CFVR of patients with MB may be one of the causes of angina pectoris.
3.Relationship of retinal lesions and postoperative visual acuity of cataract phacoemulsification

AlM: To study the relationship of retinal associated lesions and postoperative visual acuity of cataract phacoemulsification.
METHODS:From February 2013 to February 2014, 120 cases with given cataract phacoemulsification were divided into observation group and control group. Cases in observation group were with of retinal associated lesions, those in control group were without retinal associated lesions. The best corrected visual acuity, ision acuity, visual evoked potential, intraocular pressure were compared between two groups.
RESULTS:Two weeks after surgery, visual acuity in observation group was higher than those of before therapy. Visual acuity recovery rate of observation group was significantly slower than that of control group; best corrected vision, visual acuity in observation group was significantly lower than that in control group;observation group's amplitude (12. 01±4. 50ü V) was significantly lower than control group, incubation period (114. 29±11. 32ms) was significantly higher than control group; After 6, 12, 24h, intraocular pressure (23. 64 ± 4. 28, 24. 12 ± 5. 13 and 18. 28± 3. 22mmHg) were significantly higher than control group.
CONCLUSlON: Retinal associated lesions can lead to visual evoked potential change, elevated lOP, affect postoperative visual recovery level.
6. Effect of gene silencing of Golgi a-mannosidase . on the invasion and migration of human gastric carcinoma BGC-823 cells

Objective: To investigate the effect of gene silencing of Golgi a-mannosidase II(GM II) by RNA interference on the invasion and migration of human gastric carcinoma BGC-823 cells. Methods: Four targeted locations were designed and four plasmid vectors expressing short hairpin RNA (shRNA) for gene silencing of GM II were constructed, and the negative control plasmid vector was also constructed. The plasmids were transfected into human gastric carcinoma BGC-823 cells by Lipofectamine 2000. The expression levels of GMIImRNA and protein were detected by RT-PCR and Western blotting, respectively. The best plasmid for gene silencing of GMIIwas selected. The invasive and migration capabilities of BGC-823 cells induced by gene silencing of GMIIwere detected by Transwell invasion assay and scratch-wound assay, respectively. Results: Plasmid vector pGPU6/GFP/Neo-1303 was the best one for the gene silencing of GM .. The number of cells invading through the matrigel in the plasmid vector pGPU6/GFP/Neo-1303-transfected group was much lower than those in the blank control group and the pGPU6/GFP/Neo-shNC-transfected group (P < 0.05). After the cells were cultured without serum for 24 h, the invasive and migration capabilities of the cells transfected with pGPU6/GFP/Neo-1303 were significantly weakened than those of the blank control group and the pGPU6/GFP/Neo-shNC- transfected group (P < 0.05). Conclusion: The expressions of GMIImRNA and protein in human gastric carcinoma BGC-823 cells can be silenced by RNA interference, which results in the inhibition of invasive and migration capabilities of BGC-823 cells. These results suggest that GMIImight be a novel target for the treatment of human gastric carcinoma. © 2012 by Tumor.
7. Research progress in pharmacological activities of ginsenoside Rb1

OBJECTIVE: To review the pharmacological effects of ginsenoside Rb1 research progress. METHODS: According to domestic and foreign research reports of ginsenoside Rb1 in recent years, the pharmacological effects of Rb1 research progress. RESULTS: This article reviewed pharmacological effects and pharmacokinetic research of ginsenosides Rb1 from the central nervous system, cardiovascular system, immune system, and anti-tumor, anti-hepatic warm ischemia-reperfusion, lowering blood sugar and other aspects. CONCLUSION: Ginsenoside Rb1 is an important chemical material with a lot of pharmacological activities, studying its pharmacological effects can provide relevant information for further its development and utilization.
8. Cartilage-derived morphogenetic protein growth factor 1-induced rat adipose-derived stem cells in repairing knee joint defect in rabbits

Objective:To repair knee joint defects in rabbits with rat adipose-derived stem cells induced by cartilage-derived morphogenetic protein growth factor 1 (CDMP1) ,so as to assess the feasibility of using heterogeneity cells as the seed cells for cartilage tissue engineering. Methods: The second generation ADSCs were seeded on scaffold, cultured for another two weeks in presence of CDMP1(50 μg/L),and identified by immunohistochemistory method. Bilateral rabbit knee joint defect model was established. The left side defect was embedded with ADSCs-scaffold composite (experimental group); the right side was embedded only with the scaffold(control group). Nine rabbits were killed in each group 8,16,24, and 48 weeks after embedding and the tissues were made into slices for safranine O and haematoxylin eosin staining. Results: In the experimental group the defects were filled with white semi-transparent tissues 8 weeks after embedding, with clear boundary to the surrounding cartilage; 16 weeks after embedding, the boundary of defect was further improved but still could be seen; 24 weeks after embedding, the repair outcomes were satisfactory, with the newly-generated chondrocytes having a nearly normal morphology (sphere shape,cartilage lacuna),and safranine O and haematoxylin eosin staining results were both positive; and 48 weeks after embedding, the boundary of the repair region could be clearly seen,and the repair effects were not as satisfactory as those of after 24 weeks. In the control group the boundary between the repairing area and the normal circumjacent area was visible at all 4 time points,with clear boundary and granulation tissues; the newly generated cells took a spindle shape and were negative for H-E and safranine O staining. Conclusion: The knee joint defects of rabbits can be satisfactorily repaired by using CDMP1-induced ADSCs seeded on spongy bone scaffold of cattle, which provides a theoretical basis for using heterogeneity cells as the seed cells for cartilage tissue engineering.
