1.Reversal of drug resistance of trastuzumab-resistant human breast cancer cells by recombinant adenovirus carrying PTEN gene

Objective:To study the role of adenoviral-mediated PTEN gene transfection in reversing drug resistance of trastuzumab-resistant human breast cancer.Methods:Recombinant adenoviruses carrying PTEN gene(AdPTEN) were constructed and transfected into trastuzumab-resistant human breast cancer cell line BT474.Proliferation and apoptosis of BT474 cells treated with AdPTEN and trastuzumab were measured by MTT assay and FCM.Time course of BT474 cells apoptosis induced by AdPTEN was analyzed by detection of DNA fragmentation.Western blotting was employed to measure phosphorylated-Akt expression levels in AdPTEN treated BT474 cells.The nude mice model of BT474 cells was employed to observe the effects of AdPTEN and trastuzumab on trastuzumab-resistant human breast cancer in vivo.The expression of PTEN gene,cancer cells apoptosis and ultrastructural changes were evaluated after the mice were sacrificed.Results:We successfully constructed recombinant AdPTEN;and the titer of the recombinant adenovirus was about 4.2?1011TCID50/ml.PTEN gene expression was identified by PCR,RT-PCR and Western blotting assay.Combinatorial AdPTEN and trastuzumab significantly suppressed the proliferation of BT474 cells and induced the apoptosis.The percentage of apopotosis cells treated with AdPTEN was(20.7?5.82)%,companied by cells cycle arrest in G1 phrase(P
4.Clinical research of Lipo PGE_1 in the treatment of diabetic peripheral neuropathy

Objective: To observe the therapeutical effects of Lipo PGE_ 1 in the treatment of diabetic peripheral neuropathy. Methods: 42 cases of type 2 diabetes patients with diabetic peripheral neuropathy were recruited and were divided into two groups at random:therapy group treated with Lipo PGE_ l and control group treated with Vitamine B_ 1 and Vitamine B_ l2 . Results: After a treatment of 20 days,symptoms such as pain,numbness and hypoesthesia in cases of therapy group had been improved at different degrees.Examination of Neuroelectrophysiology indicated an increase of sensory nerves conduction velocity and motor nerve conduction velocity.The effects of therapy group outweighed that of control group. Conclusion: Lipo PGE_ 1 can significantly relief the symptoms of diabetic peripheral neuropathy and increase nerve conduction velocity.The mechanism of action may involve improvement of blood-supply of nerve by vasodilatation and inhibition of platelet aggregation that caused by PGE_ l .
5.Progress on bone marrow mesenchymal stem cells transplantation for spinal cord injury.

Bone marrow mesenchymal cells (BMSCs) are regarded as donor cells in cell transplantation therapies for spinal cord injury (SCI) for they have the ability of favourable proliferation and multi-directional differentiation, and are easily isolated and culturd and have less immunological reaction. It has been confirmed that subarachnoid space injection is the most ideal delivery technique of BMSCs. Bone marrow mesenchymal stem cells transplantation is safe and its reconditioning role is certain for SCI in early clinical application. The mechanism of BMSCs promoting functional recovery after SCI is probably concerned with vicarious function, nerve trophism, immunosuppression and promoting axonal regeneration by BMSCs.

6.Eliciting thinking from the competition in basic medical design of experimentation for cultivating excellent doctors

The purpose of the National Competition in Basic Medicine Innovation Forum & Ex-perimental Design in colleges is to boost Excellent-doctor Education Training Plan, cultivating students' scientific research ability and other comprehensive capacity. In practice, the originality and the scientific nature of the selected topic were emphasized through the provision of scientific research funding. The teachers changed their traditional instructional concept and teaching models. They guided the students to select the scientific subjects and took various measures to change students ' learning mode, resulting in gradually pro-moting the students' scientific research level and comprehensive ability. The results showed that these novel strategies are feasible and effective, which may provide a new way for improvement of the excellent doctor program.
8.Radiographic evaluation of the effect that local injection of platelet-rich plasma enhances bone consolidation

Objective In this study, we tested the ability of platelet-rich plasma to enhance bone consolidation during distraction osteogenesis in a rabbit model of leg lengthening. Methods Twenty New Zealand white rabbits, which were randomly divided into two groups, the PRP treated group and the control group (n=10). Osteotomy will be made under the tibiofibular junction of left tibia, and 1cm tibia shaft will be removed through a second osteotomy. The tibia will be shorted for 1cm and then fixed by Orthfix unilateral lengthener with 4 pins. Lengthening will start 7 days after the osteotomy surgery, at a rate of 1.0 mm/day, in two steps, for 10 days. 500ul PRP were respectively injected into the distraction gaps at the end of the 10-day lengthening and at 10 days before termination. No management in the control group. The animals were terminated at day 20 following final lengthening. Serial radiographs will be taken at the day of surgery, day 12, day 17, day 27, day 37, using a C-arm radiography system. The excised bone specimens were scanned by X-ray and Micro-CT, and the bone mineral content(BMC) and the bone mineral density(BMD) of the regenerates were calculated and compared. Results The regenerate of PRP treated group was significantly better than the control group. The X-ray figures showed that the regenerate was significantly more than the control group. The figures of micro-CT 3D reconstruction showed that the growth and remodel of the regenerate in PRP treated group was better than the control group. The localization of some enzyme in adult worms of Clonorchis sinensis was studied by histochemical method in this paper. Acid phosphatase was detectcd mainly in digestive duct, subtegument and tne walls of testis, uterus and ovary; acetylcholinestcrase was found in oral sucker, ventral sucker, pharynx; and ATPase was found to exist in oral sucker, ventral sucker, pharynx and musclxe layer in the subtegument. The eggs of C. sinensis possessed the above three enzymes (Figs.l-5).
