Vascular endothelial growth factor(VEGF) is one of the best characterized angiogenic regulators which has many effects in promoting endotheliocyte proliferation and differentiation,increasing the microvascular permeability,inducing the angiogenesis,triggering the growth,survival and migration of tumor cells by combining its specificity receptor (VEGFR).Dysregulation of VEGF expression and signaling pathways therefore plays a pivotal role in the pathogenesis and clinical features of hematologic malignancies,direct and indirect targeting of VEGF and its receptors therefore may provide a potent novel therapeutic approach to overcome bone marrow angiogenesis and multidrug resis-tance thereby improve patient outcome.Recent years,a novel VEGF blockade system using RNA interference attracts more and more people's attention.The small interfering RNA (siRNA) targeting VEGF/VEGFR can completely inhibits the expression of VEGF and induce the silence of corresponding genes.

4.An Effect Evaluation on the Intervention Mode of AIDS High -risk Behaviors

Objective To explore and assess the intervention mode consist of governments,professional organizations and volunteers for AIDS high -risk behaviors.Methods Commercial sex workers,gays,venereal outpatients and other target population were chosen to be under intervention measures by AIDS high -risk behaviors intervention team consist of government,CDC and volunteers.Then awareness rate,use rate of condom and care rate of infectious were calculated. Results A team of 313 volunteers were set up.The cadres,students and owners of public places were trained and it reached 42 000 person training times in 2012.The awareness rate of AIDS -prevention knowledge among commercial sex workers increased from 76.67% to 91.41% and the use of condom increased from 66.67% to 89.74%.The care rate of infections /patients increased from 0 to 100%.Conclusion This model plays an important role in the AIDS prevention and treatment.It is a good working mechanism to build the intervention mode consist of governments,professional organizations and volunteers.
5.Effect of herbal cake-partitioned moxibustion on MEK1/2 and ERK1/2 expressions of gastric tissues in rats with spleen deficiency syndrome

Objective:To observe the effect of herbal cake-partitioned moxibustion on the expressions of mitogen-activated protein kinase (MEK1/2) and extracellular regulatory protein kinase (ERK1/2) in gastric tissues of rats with spleen deficiency syndrome, and to explore the possible mechanisms of herbal cake-partitioned moxibustion in treating spleen deficiency syndrome. Methods:Sixty Sprague-Dawley (SD) rats were randomly divided into a blank control group (group A), a model group (group B), a ranitidine group (group C), and a herbal cake-partitioned moxibustion group (group D) by random digit, 15 rats in each group. Rat models of spleen deficiency syndrome were made by intragastric administration of 4℃ 200% concentrated Da Huang (Radix et Rhizoma Rhei). After successful modeling, the rats in group C were treated with 25 mg/(kg·bw) ranitidine by intragastric adminstration and rats in group D were treated with herbal cake-partitioned moxibustion at Zusanli (ST 36) and Zhongwan (CV 12), for 8 d. Excepted for rats in group A, all the other rats were treated with indomethacin at 5 mg/(kg·bw) at 8:00 a.m. on the second day after finishing all the intervention and sacrificed 7 h later to isolate the stomach. Histopathological changes of the gastric tissues were observed under light microscope after hematoxylin-eosin (HE) staining. The protein expressions of MEK1/2 and ERK1/2 in the gastric tissues were detected by immunohistochemistry. Results:After intervention, the gastric mucosal injury in group B was significantly severer than that in group A, with large breakage and ablating; the damage of gastric mucosa was decreased in group C compared with group B; the gastric mucosal surface remained relatively complete, and the status of breakage and ablating was significantly improved. After intervention, compared with group A, the protein expressions of MEK1/2 and ERK1/2 in gastric tissues of the other groups were significantly higher (P<0.01). Compared with group B, the protein expressions of MEK1/2 and ERK1/2 in group C and D were significantly higher (allP<0.01). Compared with group C, the protein expressions of MEK1/2 and ERK1/2 in group D were significantly higher (P<0.01). Conclusion: Herbal cake-partitioned moxibustion promotes the repair of gastric mucosa in rats with spleen deficiency syndrome, via improving protein expressions of MEK1/2 and ERK1/2 in gastric tissues, as well as activating MEK/ERK signaling pathway.
6.Effects of trichostatin A on human lung cancer cell strains A549.

OBJECTIVETo explore the effect of trichostatin A (TSA) on human lung cancer cell strains A549.
METHODSA549 cells were exposed to TSA at different concentrations, then the growth-inhibiting effects of the cell line were detected with 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay; After the cells were exposed to TSA for 48 and 96 hours at 300 nmol/L, the change of the cell cycle and apoptosis of A549 were analyzed with flow cytometry. p21 protein and extracellular signal regulated kinase (ERK) expression were detected by Western blot.
RESULTSTSA inhibited the growth of A549 cells in time- and concentration-dependent manners. The proportion of apoptosis, G0/G1 and G2/M phase increased in accordance with raising of the TSA concentration. The expression of p21 protein was significantly up-regulated and the expression of phosphorylation ERK was significantly down-regulated after A549 cells were treated with TSA.
CONCLUSIONSHistone deacetylase inhibitor TSA can inhibit the proliferation of human lung cancer cell strains A549 and induce the cell cycle arrest and apoptosis in the A549 cells. This may be related to up-regulation of p21 protein expression and the down-regulation of phosphorylation ERK.

In many studies about biomedical research factors influence on the outcome variable,it has no influence or has a positive effect within a certain range.Exceeding a certain threshold value,the size of the effect and/or orientation will change,which called threshold effect.Whether there are threshold effects in the analysis of factors (x) on the outcome variable (y),it can be observed through a smooth curve fitting to see whether there is a piecewise linear relationship.And then using segmented regression model,LRT test and Bootstrap resampling method to analyze the threshold effect.Empower Stats software developed by American X & Y Solutions Inc has a threshold effect analysis module.You can input the threshold value at a given threshold segmentation simulated data.You may not input the threshold,but determined the optimal threshold analog data by the software automatically,and calculated the threshold confidence intervals.

OBJECTIVETo investigate the regular pattern of Cytomegalovirus (CMV)-specific T cells (CTL) immune reconstitution after human leukocyte antigen (HLA) matched sibling donor allogeneic bone marrow(BM) plus peripheral blood hematopoietic stem cell (PBSC) transplantation.
METHODSCTL from seventeen patients after transplantation was detected by flow cytometry, the IFN-γ secretion ability of CTL by enzyme-linked immunospot (ELISPOT) assay, and clonal analysis of TCR Vβ subfamily by gene scan assays. The relationship between CTL reconstitution and CMV infection was studied.
RESULTSBoth number and function of recipients CTL reached to normal control level at 30 d post-transplantation. The recipients achieved a high frequency CTL with IFN-γ response and restoration of T-cell receptor β (TCR Vβ) repertoire at one year post-transplantation. CTL with the central memory CD45RO(+)CD62L(+) cell phenotype expanded in PB when CMV was reactivated. The incidence of CMV reactivation was 35.83% (17.91% - 63.10%) after transplantation, and none of them developed CMV disease.
CONCLUSIONAfter HLA matched related donor transplantation using mixed grafts, immune recovery to CMV seems to be early and fast. The incidence of CMV infection and disease are low.

10.Predictive value of HATCH score on atrial fibrillation recurrence post radiofrequency catheter ablation.

OBJECTIVETo determine the predictive value of HATCH score on recurrence of atrial fibrillation (AF) after radiofrequency catheter ablation (RFCA).
METHODSThe data of 123 consecutive AF patients (74 paroxysmal and 49 persistent AF) who underwent RFCA between April 2009 and December 2010 in our department were retrospectively analyzed. Of theses patients, 65 (52.9%) patients had HATCH score = 0, 41 (33.3%) patients had HATCH score = 1, and 17 (13.8%) patients had HATCH score ≥ 2 (HATCH = 2 in 11 patients, HATCH = 3 in 5 patients, HATCH = 4 in 1 patient). The recurrence was defined as atrial tachyarrhythmia lasting more than 30 seconds after 3 months post RFCA. The patients were divided into recurrence group and no recurrence group. Relationship between HATCH score and recurrence was observed.
RESULTSThere were 43 cases in recurrence group and 80 cases in no recurrence group. After 12 months follow-up, HATCH score was significant higher in recurrence group than in non-recurrence group [(0.91 ± 0.94) score vs. (0.53 ± 0.80) score, P < 0.05]. The ratio of patients with HATCH ≥ 2 in recurrence group was higher than in non-recurrence group [23.3% (10/43) vs. 8.8% (7/80), P < 0.01]. The sensitivity and specificity of HATCH ≥ 2 to define the risk of recurrence was 25.0%, 92.4% respectively. Cumulative non-recurrence rate of patients with HATCH score ≥ 2 was lower than patients with HATCH score = 0 and 1 (P < 0.05).
CONCLUSIONHigher HATCH score is associated with increased risk of AF recurrence post RFCA.
