1.Optimization of taxol extraction from Taxus baccata L.

Objective To optimize and establish the extractive technology of taxol from Taxus baccatal L.. Methods HPLC was applied to determine the content of taxol. The orthogonal test was adopted to examine the effects of the 4 factors consisting of ethanol concentration,ethanol volume,extractive times and extractive time. Results Ethanol concentration and ethanol volume were essential factors influencing the extraction of taxol. High concentration and amount of ethanol facilitated the extraction. Extractive times and extractive time were also important. By single-factorial effect,2 h and 2 times in each extraction of taxol were the best. Conclusion The optimum extractive condition is T9 (A3B3C2D1) and T8 (A3B2C1D3).
2.Effect of compound qingqin liquid on the expression levels of ang II and COX-2 mRNA transcription and protein expression in the renal tissue of uric acid nephropathy rats: an experimental study.

OBJECTIVETo investigate the effect of Compound Qingqin Liquid (CQL) on the expression level of angiotensin II (Ang II) and COX-2 mRNA transcription and protein expression in the renal tissue of rats with uric acid nephropathy.
METHODSSD rats were randomly divided into the blank control group, the model group, the positive drug group, the high, moderate, and low dose CQL group according to number randomization principle. The model was established by gastrogavage of adenine, accompanied with yeast feeding. Distilled water was given by gastrogavage to rats in the blank control group and the model group. Allopurinol at the daily dose of 9.33 mg/kg was given by gastrogavage to rats of the positive control group. CQL at the daily dose of 3.77 g/kg, 1.89 g/kg, and 0.09 g/kg was respectively given by gastrogavage to rats in the high, moderate, and low dose CQL groups. All treatment lasted for 6 weeks. Rats were randomly divided at week 4 (3 in the blank control group, and 6 in the rest groups), and the rest rats were killed at week 6. The renal tissue was extracted. The expression level of Ang II and COX-2 mRNA transcription were detected by RT-PCR. The expression level of Ang II was detected by ELISA. The expression level of COX-2 protein was detected by Western blot and immunohistochemical assay.
RESULTSCompared with the blank control group, except the mRNA expression of Ang II at week 4, the mRNA and protein expression of Ang II and COX-2 obviously increased at week 4 and 6 in the model group (P < 0.01, P < 0.05). The COX-2 protein expression at week 4 was obviously lower in the high and moderate dose CQL groups than in the model group and the low dose CQL group (P < 0.05); the average integral of optical density value was obviously lower in the positive control group than in the model group. Except the mRNA expression of Ang II in the high dose CQL group at week 6, the mRNA and protein expression of Ang II obviously decreased in the positive control group and each dose CQL group (P < 0.01, P < 0.05). Of them, the effects were better in the high and moderate dose CQL groups than in the positive control group and the low dose CQL group (P < 0.05, P < 0.01). Besides, the mRNA expression of COX-2, the average integral of optical density value were obviously lower in the positive control group and each dose CQL group than in the model group (P < 0.05). The protein expression of COX-2 was obviously lower in the high and moderate dose CQL groups than in the model group (P < 0.05). Of them, the mRNA expression of COX-2 was better in the moderate dose CQL group than in the positive control group (P < 0.05); the protein expression of COX-2 was better in the high dose CQL group than in the low dose CQL group (P < 0.05).
CONCLUSIONCQL was capable of lowering the expression level of Ang II, COX-2 mRNA transcription and protein expression, thus suppressing the inflammatory pathological injury of the renal tissue.

3.Study on the risk factors and its interaction on Parkinson disease.

OBJECTIVETo explore the risk factors of Parkinson disease (PD), interaction between family history of PD and other risk factors, as well as the relative strength of genetic factors over the vulnerability of PD.
METHODSOne 1:1 matched case-control study including 157 pairs of cases and controls was conducted in Qilu Hospital of Shandong University.
RESULTSConditional logistic regression analysis showed that family history of PD, mental labor, insecticide, alcohol drinking and history of depression all had positive relationship, while smoking had a negative relationship with PD. The AP (AB)s of family history of PD and insecticide, alcohol drinking, history of depression were 55.2%, 34.0%, 41.4% and the RERIs were 8.96, 3.31, 7.85 respectively. The heritability of PD patients' first degree relatives was 36.86% +/- 5.76%, and second degree relatives was 20.66% +/- 6.81%.
CONCLUSIONFamily history of PD had an additive model synergism on PD, coexisting with other risk factors. Genetic factors had a smaller action on PD than environmental factors.

4.Mechanism of colon cancer cell apoptosis induced by telocinobufagin: role of oxidative stress and apoptosis pathway.

OBJECTIVETo investigate the effects of telocinobufagin on viability and apoptosis of colorectal cancer (CRC) cells and explore the mechanism of telocinobufagin-induced apoptosis.
METHODSMTT assay was performed to detect the viability of CRC cells exposed to telocinobufagin. Nuclear staining with Hoechst 33342 and flow cytometry were used to analyze the cell death of CRC cells. Expressions of proteins related with cell apoptosis and oxidative stress were determined with Western blotting.
RESULTSTelocinobufagin decreased the viability of CRC cells in a time- and dose-dependent manner. The presence of karyopycnosis and apoptotic bodies together with the results of flow cytometry suggested that telocinobufagin induced cell apoptosis to cause cell death. Western blotting showed that telocinobufagin exposure of the cells resulted in upregulated p53 and Bax protein expressions and promoted cleavage of caspase 9 and PARP. Telocinobufagin induced phosphorylation of Bad and PARP cleavage, and suppressed phosphorylation of IKBα and TAK1 and expression of survivin in the cells.
CONCLUSIONTelocinobufagin can decrease the viability of CRC cells by inducing cell apoptosis, which involves p53-mediated Bax activation and inhibition of the IAP pathway.

5.Chemical constituents of Cyanotis arachnoidea.

AIMTo investigate the chemical constituents of Cyanotis arachnoidea.
METHODSBy using chromatographic methods for separation and combination with spectral analysis, their chemical structures were determined.
RESULTSSix compounds were identified as ajugasterone C-20, 22-acetonide (1), 20-hydroxyecdysone-20, 22-acetonide (2), 22-oxo-ajugasterone C (3), 22-oxo-20-hydroxyecdysone (4), beta-sitosterol (5), daucosterol (6).
CONCLUSIONCompound 3 is a new compound, 4 was a new natural compound.

6.Involvement of TLR4/NLRP3 inflammasome in contrast medium-induced inflammation and injury in renal tubular epithelial cells

AIM: To investigate whether Toll-like receptor 4 ( TLR4 ) and Nod-like receptor protein 3 (NLRP3) inflammasome were involved in contrast medium (CM)-induced inflammation and injury in renal tubular epithe-lial cells.METHODS: Iopromide was used to injure NRK-52E cells in the study.The cell viability was measured by CCK-8 assay.The protein levels of TLR4, NLRP3, apoptosis-associated speckle-like protein (ASC), caspase-1 and cleaved caspase-3 were determined by Western blot .The releases of interleukin ( IL )-1βand IL-18 were detected by ELISA .The apoptotic rate was evaluated by Hoechst staining , and mitochondrial membrane potential ( MMP) was analyzed by JC-1 staining.siRNA was transfected into the NRK-52E cells to silence NLRP3 expression.RESULTS:CM decreased the viability of NRK-52E cells (P<0.05).CM also elevated the protein levels of cleaved caspase-3, TLR4, NLRP3, IL-1βand IL-18 (P<0.05).Silencing NLRP3 attenuated CM-induced releases of inflammatory cytokines .Moreover, treat-ment with TLR4 inhibitor TAK-242 or knockdown of NLRP3 by siRNA transfection both attenuated cell apoptosis and loss of MMP caused by CM .CONCLUSION:TLR4/NLRP3 inflammasome takes part in the pathogenesis of CM-induced acute kidney injury , and mediates CM-induced injury and inflammation in renal tubular epithelial cells .
7.Effects of bradykinin on the proliferation of rabbit corneal endothelial cells and the expression of tight junction-related proteins ZO-1 and ZONAB

Objective To investigate the effects of bradykinin (BK) on the proliferation of rabbit corneal endothehal ceils (RCECs) and the expression of tight junction-related proteins zonula occludens-1 (ZO-1) and zonula occludens-1-associated nucleic-acid-binding protein (ZONAB),and to explore the underlying mechanisms of BK on cell proliferation in corneal endothelium.Methods RCECs at logarithmic growth phase were treated with different concentrations of BK (0.01,0.1,1,10 μmol · L-1) BK group,with the controls left untreated.Morphological changes of cells in each group were examined under phase-contrast microscope,and MTT assays were used to detect cell proliferation at 24 h,48 h,72 h,96 h after BK treatment.And,at 72 h,the expression levels of ZO-1 and ZONAB protein were determined by Western blot.Results After 72 h of treatment,the cells in each group were fused into pieces and closely linked into a monolayer;but after 96 h,the growth of the cells was restricted,with the intercellular space become larger and the cells exfoliated.Compared with the control group,BK induced a significant increase of absorbance value and cell viability,and the differences were statistically significant (all P < 0.001),and the promoting effects showed a concentration-dependent manner,and 1 μmol · L-1 BK demonstrated the strongest regulative effect (P < 0.001).Western blot results showed that BK upregulated the expression of ZO-1 and ZONAB protein in a concentration-dependent manner.Conclusion BK can stimulate the proliferation of RCECs in a time-and concentration-dependent manner,and the mechanisms are probably associated with ZO-1/ZONAB-mediated signaling pathway.
8.Dosimetric differences between volumetric modulated arc therapy plans with different arc numbers for cervical cancer

Object To study the effects of the number of arcs in the treatment of cervical cancer with volumetric modulated arc therapy (VMAT) on the treatment plan, and to provide reference for the selection of the number of arcs in the clinical application.Methods CT images of 10 patients with cervical cancer were selected, it's the radiation therapist that delineated the target area and organs, the prescribed dose was 5040 cGy per 28 times to the target area, and single arc (Arc1), double arc (Arc2) and three (Arc3) plan were designed by RayStation planning system to compare the differences with the dosimetry characteristics of target and organs. The time required for planning optimization was recorded, and the difference between the monitor unit and the delivery time in the three plans was analyzed. A dose validation tool for PTW was used to verify the dose to analyze the influence of the number of arc on the passing rate of gamma verification.Results Single arc plan (Arc1), double arc plan (Arc2) and three arc plan (Arc3) for 10 cases of cervical cancer all proved to meet the clinical requirements. With the increase of arc number, the target area distribution got improved, V40, V45 and Dmean in the organs at risk of rectum, bladder, and femoral head gradually decreased in case of doses of 40 and 45 Gy, and V35, V40 and V45 in the normal tissue were lowered in case of the doses of 35, 40 and 45 Gy. With the increase of the arc number, the optimization time and the delivery time of the plan increased, while the difference between the monitor unit of the three plans was smaller. The passing rate of the three plans was more than 99.5%, and the difference was small.Conclusion Cervical cancer VMAT plan has the target dose distribution and the protection of organs at risk enhanced with the increase of the number of arcs, while the treatment time and cost increased. Therefore, it is necessary to take comprehensive considerations on the number of VMAT plan arcs.
9.Relationship between HBeAg seroconversion with genotypes and HBV specific CTL in patients with chronic hepatitis B treated with Adefovir dipivoxil

Objective To explore relationship between HBeAg seroconversion with HBV genotypes and HBV specific CTL in patients with chronic hepatitis B ( CHB) treated with Adefovir dipivoxil. Methods Seventy CHB patients had positive HBV DNA ( HBV DNA ≥ 1 X 104 copy/ml) ,45 cases had positive HBeAg,of whom 23 cages (51. 11% ) had genotype B, 22 cases (48. 89% ) had genotype C. ALT > 2 × upper limit of normal value(ULN) , human leukocyte antigen( HLA)-An positive,patients were treated with Adefovir dipivoxil ( commercial name is Mingzheng, Zhengda Tianjing Pharmaceutical Company), 10 mg, orally,once a day. After treatment for 12 months, observe relationship between HBeAg seroconversion with HBV genotypes and HBV specific CTL. Results After treatment with Adefovir dipivoxil for 12 months, HBV specific CTL(0. 68% ±0. 11% )was higher than that before treatment (0. 33% ±0. 11% ) , t =8. 36 P < 0.001, HBV DNA (3.01 ±0.2) log 10 copy/ml was lower than that before treatment (6.27 ±0.70) log10 copy/ml, t = 12. 63 P<0.001, HBV DNA turned negative ( <500 copy/ml) 43 cases (61.43%), in 45 cases with positive HBeAg, HBeAg turned negative in 13 cases (28.89%), 8 cases had HBeAg seroconversion( 17. 78% ) ,HBV specific CTL(0.86% ±0. 05% ) of patients with HBeAg seroconversion is higher than (0. 61% ±0. 07% )of patients without HBeAg seroconversion (37 cases, 82. 22% ) t =7. 88, P <0. 001. In 8 cases with HBeAg seroconversion, 7 cases had genotype B(30. 43% of genotype B) , 1 cases had genotype C(4. 55% of genotype C) ,x2 =5. 15,P <0. 05. Conclusion Adefovir dipivoxil can enhance HBV specific cellular immunity of CHB patients. After treatment, occurrence of HBeAg seroconversion is related to increase of HBV specific CTL level and may be related to genotypes.
10.Application of modified OSCE in evaluating core competencies of new nurses in neurosurgery standardized training

Objective:To explore the effectiveness of modified objective structured clinical examination (OSCE) for evaluating the core competencies of new nurses on standardized training in neurosurgery.Methods:Thirty-seven new nurses on standardized training in neurosurgery were evaluated using a modified OSCE protocol that was focused on core competencies required for clinical nursing practice on the basis of a clinical case library, which consisted of diagnostic assessment upon entrance, process teaching evaluation during training, and outcome assessment upon exit, at five OSCE stations (nursing assessment, skill practice, clinical thinking, health education, and humanistic care). SPSS 26.0 was used to perform the t-test and rank sum test. Results:The nurses highly recognized the effectiveness of modified OSCE and were largely satisfied with teaching. There were significant differences at entrance and exit in terms of the mean OSCE score ( t=24.21, P<0.001), independent learning ability ( t=10.29, P<0.001), and the scores of core competencies—good personal traits ( Z=5.24, P<0.001), clinical nursing ability ( Z=5.07, P<0.001), support and interpersonal communication ability ( Z=5.12, P<0.001), critical clinical thinking ability ( Z=5.17, P<0.001), professional construction and self-development ( Z=5.05, P<0.001), and the total score ( Z=5.27, P<0.001). Conclusions:The modified OSCE can comprehensively evaluate the core competencies of new nurses in neurosurgery standardized training, with good feasibility and effectiveness, which provides a reference for teaching management of standardized training programs.
