1.Construction of pEGFP-VEGF recombinant plasmid and its expression in osteoblast

Objective:To construct a vector containing human vascular endothelial growth factor,VEGF 121 (VEGF 121 ) with green fluorescence protein as the reporter gene,and then to observe its expression in osteoblast.Methods:VEGF 121 gene was amplified from foetus brain library by PCR,then ligated with pEGFP-C1 to form recombinant plasmid pEGFP-VEGF and transfected into osteoblast by liposome.The green fluorescence protein expression was observed by fluorescence microscopy and the VEGF expression was last detected by immunocytochemistry and ELISA.Results:VEGF 121 gene fragment was amplified by PCR and ligated to pEGFP-C1,then transfected into osteoblast successfully.The strong expression of green fluorecensce protein was observed by fluorecensce microscopy,and the expression of VEGF protein was detected by immunocytochemistry and ELISA.Conclusion:Recombinant plasmid pEGFP-VEGF is effectivly expressed after being transfected into osteoblast in vitro,which provides theoretical support for further study with its simple and reliable detecting methods.

4.Pulmonary sequestration: a literature review of 2625 cases

Total 2625 cases of pulmonary sequestration were collected from literatures of Chinese National Knowledge Infrastructure from 1998 to 2008. The age, gender, clinical symptoms, chest CT scan,chest X-ray findings, lesion location, supplying artery, venous reflux, and preoperative misdiagnosis of 2625 cases were retrospectively reviewed. The male: female ratio of all cases was 1.58: 1. The main clinical symptoms of pulmonary sequestration were cough, sputum, fever, hemoptysis and chest pain, but 13.36% patients were asymptomatic. Chest CT scan showed pulmonary massive lesions (49.00%), pulmonary cystic lesions (28.57%), pulmonary cystic-solid lesions ( 11.57% ), and flaky shadows (7.96%). Pulmonary sequestration mainly located in the lower lobe; 66. 43% located in the left posterior basal segments, and 20. 16% located in the right posterior basal segments. Pulmonary sequestrations were divided into three types, intralobar sequestration (83. 84% ), extralobar sequestration ( 16.03% ), and mixed sequestration (0.13% ). The arterial supply was mainly from branches of thoracic aorta (76. 55% ) and abdominal aorta (18.47%). The clinical manifestations of pulmonary sequestration were various, so the preoperative misdiagnosis rate was as high as (59 ± 20) %. We should take full advantage of clinical symptoms, imaging findings and location characteristics to improve the preoperative diagnosis rate.
5.Application of gene expression profile in tumor radiotherapy

Gene expression profile technology is characterized by high automation, high accuracy and large information volume, which provides a new way for the development of individualized therapies and novel therapeutic targets. Some researches have identified sets of discriminating genes by using gene expression profile technology that can be used for the predietion of response to radiotherapy in human cancers. Some studies also have confirmed the therapeutic tagerts of radiation sensitizer.
6.Test analysis on tumor marker CA19-9,CEA,CA242,CA125 and AFP in assessing efficacy of chemotherapy in gastric cancer

Objective To investigate the test analysis of tumor marker CA19‐9 ,CEA ,CA242 ,CA125 and AFP for jointly assess‐ing the chemotherapeutic effect of gastric cancer .Methods Totally 160 patients with gastric cancer in our hospital from January 2010 to December 2013 were selected as the research subjects .The expressions of related tumor markers were detected before treat‐ment and at 3 months after treatment .The relationship between the TNM staging and the tumor markers ,changes of tumor markers before and after treatment ,and changes of tumor markers in the patients with recurrence and metastasis ,and patients without recur‐rence and metastasis were analyzed .Results Different TNM staging had significant correlation with tumor markers associated posi‐tive expression rate .The positive expression rate of related tumor markers in the patients with stage Ⅲ and stage Ⅳ were signifi‐cantly higher than that in the patients with stage Ⅰ - Ⅱ .The positive expression rates of CA19‐9 ,CA242 ,CA125 and AFP after chemotherapy were significantly decreased compared with before treatment (P<0 .05) ,while the CEA level had no statistically sig‐nificant difference before and after treatment (P>0 .05) .After treatment ,the CEA ,CA19‐9 ,CA242 ,CA125 and AFP positive ex‐pression rates were declined ,moreover showing statistical differences compared with pre‐treatment (P<0 .05) .The CEA ,CA19‐9 , CA242 ,CA124 and AFP levels in the patients with recurrence and metastasis were significantly higher than those in the patients without recurrence and metastasis (P<0 .05) .Conclusion CA19‐9 ,CEA ,CA242 ,CA125 and AFP have a significant correlation with tumor TNM staging ,their expression level and positive expression rates have certain correlation with the prognosis in the pa‐tients with gastric cancer ,which is worthy of focus in clinic .
7.Cognitions and behavior of cough etiquette of community residents

To explore the cognitions and behavior of cough etiquette of 556 residents in Longhua Community and self-compare the changes of cognitions and behavior before and after health education and training.The results showed that awareness rate of cough etiquette was low before training (senior group:25.4%,children:11.9%,young white-collar:36.1%,immigrants:14.3%,P < 0.05) and increased significantly (senior group:97.2%,children:99.2%,young white-collar:100.0%,immigrants:98.6%,P < 0.05).Except for no difference in compliance aspects of immigrants (P > 0.05),the other three groups improved after training.And the difference was statistically significant before and after training (P<0.05).
8.Correlations of serum Hcy,Cys C levels and type 2 diabetic macrovascular complications and its clinical sig-nificance

Objective To explore the relation between serum Hcy,Cys C levels and type2 diabetic macro-vascular complications and its clinical significance.Methods The serum levels of Hcy and Cys C were measured with immunoturbidimetry in 80 cases of type 2 diabetic patients with macrovascular complications and 80 cases of type 2 diabetes without macrovascular complications,and in 80 healthy subjects which was seleted as control.Results Serum levels of Hcy and Cys C of type 2 diabetic patients with macrovascular complications were (26.42 ± 5.75)μmol/L and (2.30 ±0.40)μmol/L,which were higher than those of patients without macrovascular complica-tions for (15.31 ±3.13)μmol/L and (1.51 ±0.27)μmol/L(t =15.17,14.64,all P <0.05);Moreover,which were also higher than those in the healthy control group for (8.86 ±1.62)μmol/L and (0.85 ±0.14)μmol/L(t =26.29, 30.56,all P <0.05).The serum levels of Hcy and Cys C of type 2 diabetic patients with carotid arteries increased were (32.53 ±6.46)μmol/L and (2.83 ±0.52)μmol/L,which were higher than those of patients without carotid arteries for (18.46 ±3.63)μmol/L and (1.65 ±0.30)μmol/L(t =11.64,12.06,all P <0.05).In patients of type 2 diabetic with macrovascular complications,Hcy was positively correlated with serum Cys C(r =0.617,P <0.05). Conclusion sCD40L and YKL -40 may have important role in the pathogenesis of type 2 diabetic patients with ma-crovascular complications,which can be used as a valuable reference target in the diagnosis and monitoring of vascular complications in elderly type 2 diabetes,and it may be one of the important independent risk factors of patients with type 2 diabetes.
9.Readiness Rate of Military Medical Equipment and Analysis on Affecting Factors

Objective To study the calculating model of military medical equipment readiness rate and the main affecting factors in order to make a valuable reference to the decision -making of medical organization for the further medical support. Methods Based on analysis of the problems in calculating method of military medical equipment readiness rate at present, a new improved calculating model and the analysis method relating to assigning mission was put forward by combining the technical requirements of GJB5338-2005 and the actual of medical equipment management. The factors affecting readiness rate in process of manufacturing, operation and maintenance support were analyzed. Results A new improved calculating model of military medical equipment readiness rate was established and the evaluating method was made by adjusting those correlated parameters in the calculation formulas. Conclusion The solution of the model is simple and convenient and the analyzing results show that the readiness rate is close to the reality. It is a practical method for achieving quality state information of medical equipment, so it can meet the requirements of military medical equipment organization in peacetime.
10.Characteristic of children’s EEG complexity at different ages and in different states

Objective: To study characteristics of children's EEG complexity at different ages and in different physiological states.Methods: The continuous 24 hour EEG recordings were obtained from 16 electrodes in 45 essential healthy children between the ages of 0 to 15 years. EEG complexity was analyzed by non linear measure in 7 states: awake with eyes opened, awake with eyes closed, NREM (nonrapid eye movements) sleep including stages ⅠandⅡ (light sleep), ⅢandⅣ(deep sleep) and REM (rapid eye movements) sleep. Meanwhile, the correlation was analyzed between complexity and ages.Results: (1)The global EEG complexity in state of being awake with eyes opened was greater than that with eyes closed;that in wakefulness state was greater than in sleep state. The EEG complexity gradually decreased with the increase of deep sleep in NREM sleep state.The complexity in REM sleep state was greater than that in deep sleep state, but lower than in wakefulness state. (2) The global EEG complexity was positively related to ages in state of being awake with eyes opened, state of being awake with eyes closed, light sleep, and not related to ages in deep-sleep state and REM sleep state.(3)In every brain area EEG complexity was positively related to ages in state of being awake with eyes closed .In paracentral region EEG complexity was positively related to ages in states of being awake with eyes opened and light sleep.Conclusion: The EEG complexity was used to study the brain dynamical characteristics in different physiology states and the relationship between encephalic electric activity and brain development. It can be used as an objective index to evaluate the function and development of brain.
