1.Clinical significance of changes of lipid in elderly patients with hepatic schis-tosomiasis

Objective To explore the clinical significance of lipid levels including total cholesterol TC triglyceride TG high?density lipoprotein HDL?C low density lipoprotein LDL?C and apolipoprotein APOAⅠand APOB of elder?ly patients with hepatic schistosomiasis. Methods A total of 280 hospitalized elderly patients with hepatic schistosomiasis 198 cases of chronic liver fibrosis and 82 cases of hepatocirrhosis were chosen as study objects and their clinical data were collected and analyzed retrospectively. Meanwhile the lipid levels between the patients with liver fibrosis and hepatocirrhosis and those among the patients with ABC degrees of Child?pugh grading of liver function were compared. Results Among the 280 patients the abnormality rates of the lipid levels were 34.8% 69/198 and 100% 82/82 in the liver fibrosis group and he?patocirrhosis group respectively and the difference between them were statistically significant χ2=5.74 P 0.05 . The levels of TC HDL?C LDL?C APOAⅠof the patients in the latter group were significantly lower than those in the former group all P 0.05 . The levels of TC TG HDL?C APOAⅠ APOB of the patients with C degree liver function were significantly lower than those of the patients with A degree liver function and the levels of TC TG HDL?C of the former were also lower than those of the patients with B degree liver function all P 0.05 . Conclusions The lipid levels of the elderly patients with he?patic schistosomiasis reduce obviously in the course of hepatocirrhosis and it is correlated with the damage level of the liver. Lipid and apolipoprotein detections have certain values on the illness judgment and prognosis assessment.
2.Actions of ginsenosides on sleep architecture and cortical electroencephalogram power spectrum in rats

Objective To study the effects of ginsenosides (GS) on spontaneous sleep architecture and Cortical EEG power spectrum. Methods 24 adult SD rats were randomly divided into the control, GS 10 and 100mg/kg groups ( n = 8). Rats were instrumented with sleep-wake recording electrodes. After recovery from surgical operation,rats were orally administered GS 10 and 100mg/kg or distilled water once per day for 6 days. On GS administration day 1 and 6,Polygraphic signs of undisturbed sleep-wake activities were recorded for 12 h after GS administration. Results On GS administration day 1 ,only 100mg/kg GS increased significantly total sleep and the non-rapid eye movement ( NREM ) sleep but decreased wakefulness [(9.40 ± 0.88 ) h, ( 8.00 ± 1. 21 ) h,(2.46 ±0.81)h s (7.55 ±1.59)h,(6.36±1.54)h,(4.38 ±1.62)h,(P<0.01, P<0.05, P<0.01),respectively] ;Low and high dose GS enhanced δ-wave power of NREM sleep and wakefulness (P< 0.05 ) but reduced θ-wave power of wakefulness (P<0.01) and-wave power during NREM, REM sleep and wakefulness (P < 0.01 ),moreover,Low and high dose GS lowered θ-wave power of REM and NREM stage(P<0.05 ) ,respectively. After 6days of GS administration, Low and high dose GS increased markedly total sleep(P<0.05 ) and NREM sleep(P<0.05 ) but decreased wakefulness (P <0.05 ) and sleep-wake cycles (P < 0.05, P < 0.01 ); moreover, Low and high dose GS enhanced δ-wave power during NREM sleep and wakefulness (P < 0. 05 ) but reduced θ-wave power of wakefulness(P < 0.05 ) and -wave power during NREM, NEM sleep and wakefulness (P < 0. 05 ), 10mg/kg GS also lowered θ-wave power of NREM sleep (P<0.01). Conclusion These results demonstrate that GS can regulate spontaneous sleep architecture in time dependent manner,as well as cortical EEG power spectrum in rats.
3.Relationship between contrast-enhanced ultrasound of renal cortical blood perfusion and SCr, BUN in rabbits with acute renal failure

Objective To evaluate the renal cortical blood perfusion changes in rabbits with acute renal failure (ARF) with gray scale contrast-enhanced ultrasound, and to explore the relationship between these changes and the blood creatinine (SCr), as well as the blood urea nitrogen (BUN). Methods Rabbit ARF models were established with 50% glycerin injected into the rabbits' thighs. Gray scale contrast-enhanced ultrasound was performed on the day before injection (T_0) and 1, 4, 8, 12 days (T_1, T_4, T_8, T_(12)) after injection. The renal cortex perfusion time-intensity curve (TIC) was analyzed, including parameters like arrival time (AT), time to peak intensity (TTP), amplitude of peak intensity (A) and slope rate of TIC (β) of renal cortex. Meanwhile the SCr and BUN were measured, the correlation between SCr, BUN and parameters were analyzed. Results Compared with the value of T_0, the value of TTP, A, β after injection (T_1, T_4, T_8) were statistically different, respectively (P<0.05), but the differences among T_1, T_4 and T_8 were various. No linear correlation between above parameters and SCr, BUN was found. Conclusion The renal cortical blood perfusion changes can be early observed with gray scale contrast-enhanced ultrasound, but there is no linear correlation between the changes of parameters and SCr, BUN.
4.Effectiveness of surgical treatment for fibrous dysplasia in proximal femur with severe varns deformity

Objective To evaluate and analyse the effectiveness of surgical treatment for fibrous dysplasia in proximal femur with severe varus deformity.Methods A retrospective study was performed in 21 patients (24 femora)of fibrous dysplasia who were treated in our hospital between August 2000 and May 2009.All patients had severe femoral varus deformity.The four-step procedures were performed orderlv as valgus osteotomy,lesion curettage,impacting of massive bone allograft,and fixation by femoral intramedullary nail.There were 6 patients with monostotic disease,15 with polyostotic diseases,including 12 males and 9 females with a mean age of 22.7 years(range,14-39 years).The average neck-shaft angle and femoral varus angle was 75°(range 55°-105°)and 30°(range,18°-45°),respectively.The average length of thigh shortened 3.4 cm(range,2.0-4.5 cm)compared with the contralateral thigh.Results All of the 21 patients were followed up from 21 months to 7 years with an average period of 3 years and 4 months.There were 30 osteotomy sites in 24 femurs,28 osteotomy sites showed bone union after 3-6 months from surgery.Two distal location of the double-level osteotomy showed nonunion,which received bone grafting again and got bone union after 3 months finally.The femoral mechanical alignments of the 21 patients had been recriticd.The average neck-shaft angle was 118°(range,95°-135°)postoperatively,the femoral varus angle disappeared.The average extremity lengthening was 2.8 cm(range,1.8-3.6 cm)postoperatively.There were no infection,recurrent fracture and progression of deformity.The visual analogue scales(VAS)score of 17 patients decreased to zero postoperatively from 7-10 preoperatively,and that of the other 4 patients decreased to 3-4 postoperatively from 8-10 preoperatively.The result of Harris hip functional score was excellent in 12 cases,good in 6,and fair in 3.Conclusion The valgus osteotomy can rectify varus deformity effectively.The reconstract nail of the fumer can support the stability of femur.Impacting of massive bone allograft can improve the capacity of the femur.
5.Limb salvage strategies for malignant scapular bone tumors in region S2

Objective To investigate and discuss the limb salvage strategies of malignant bone tumors in region S2 of shoulder girdle.Methods The data of 17 patients(13 males and 4 females)were retrospectivelv analyzed.All of them had malignant scapular tumor at least in region S2,and underwent limb salvage between January 2005 and January 2009.They aged from 14 to 55 years old(mean,32.6 years).The histologic types of them were chondrosarcoma in 6 patients,osteosarcoma in 4,fibrosareoma in 1,myeloma in 1,Ewing's sarcoma in 1,rhabdomyosarcoma in 1,non-Hodgkin lymphoma in 1,malignant giant cell tumor of bone in 1 and recurrent chondrosarcoma in 1.The tumors were staged according to Enneking surgical staging system:IB in 4 and IIB in 13.According to the MSTS classification system,the region S2 and lateral 1/3 of region S1 were effected in 11 cases,the region S2 and large part of region S1 were effected in 6 cases.The rotator cuff was involved to some extent in all patients,and the articular capsule were involved in 8 cases.Neo-adjuvant chemotherapy was given to 14 patients.The surgical strategies were depended on the range of tumor and preservation of articular capsule.The scapular prosthetic replacement was done in 8 cases with a relatively large range of tumor or resection of articular capsule,otherwise the scapular allograft replacement was done.Among the 11 cases with region S2 and lateral 1/3 of region S1 effected,the scapular allograft replacement was done in 9 cases with articular capsule preserved and the prosthetic replacement was done in 2 cases with articular capsule resected.And the prosthetic replacement was done in the other 6 cases.The articular capsule and partial rotator cuff were preserved and reconstructed preferentially.Results All patients were followed up 16-62 months (mean,35.1 months).Postoperative complications were noted in 9 patients, including mild allograft rejection and mild allograft resorption in 1 patient,allograft rejection in 1,prosthetic acromion exposure in 3,allograft resorption in 4.Local recurrence were noted in 1 patients.Three patients were dead because of extensive metastasis.The average function scores were 74.1%(range,53.3%-93.3%).Conclusion During the limb salvage surgery.it's very important to guarantee the en bloc resection of the tumor.In the meanwhile,the en bloc resection and functional reconstruction should be well balanced.
6.Association between osteocalcin Hind Ⅲ genetic polymorphism and body mass index variation Investigation of 390 premenopausal women in Nanchang region

BACKGROUND: Body mass index(BMI)is a commonly used phenotype for obesity,which is determined by multiple genetic factors.OBJECTIVE: To investigate whether osteocalcin(also known as bone Gla protein,BGP)Hind Ⅲ genetic polymorphism is associated with BMI variation.METHODS: A total of 390 premenopausal women from a local population of Nanchang City were selected.Body weight and height were measured.All participants were genotyped at the BGP Hind Ⅲ locus using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).RESULTS AND CONCLUSION: The BGP genotype frequencies of HH,Hh and hh were 0.077,0.408 and 0.515,respectively.The distribution of BGP Hind Ⅲ genotypes was consistent with Hardy-Weinberg equilibrium(P > 0.05).The BGP Hind Ⅲ were significantly associated with BMI(P=0.002),which could explain about 5.47% of BMI variation.On average,BMI of individuals with HH genotype was the highest[(22.81±0.73)kg/m2],individuals with Hh genotype was intermediate[(21.50±0.53)kg/m2],while individuals with hh genotype was the lowest[(20.23±0.63)kg/m2].Therefore,carriers of HH and Hh genotypes had,respectively,approximately 12.75% and 6.28% higher BMI than carriers of the hh genotype.To our best knowledge,this is the first study reporting the association of BGP Hind Ⅲ genetic polymorphism and BMI in healthy premenopausal women.
7.Inhibitation of effective bcl-2 siRNA on apoptosis of human leukemia-60 cells

Objective To study the effect of bel-2 siRNA on apoptosis of HL-60 cells.Methods bcl-2 siRNA was synthesized in vitro transcription with silencer siRNA construction kit.The synthesized siRNA was transfected into HL-60 cells with Amine siPORT transfection.We used MTT flow cytometer and hoechst 33258 flourescence stainning t0 evaluate cell proliferation and apoptosis. Results.Bcl-2 siRNA could partially inhibit the growth of HL-60 cells.After incubated with bcl-2 siRNAl for 48 hours,HL-60 cells exhibited morphologic characteristic of apoptosis including chromatin condensation,crescents formation and nuclear fragmentation.Conclusion Effective bcl-2 siRNA can induce apoptosis and inhibit cell proliferation.
8.Applying Fuzheng Huayu Gantang comprehensive therapeutic program for treatment of post-hepatitis B liver cirrhosis complicated with glyco-metabolic abnormality.

OBJECTIVETo study the clinical effects of Fuzheng Huayu Gantang comprehensive therapeutic program (FHGP) on post-hepatitis B liver cirrhosis associated with glyco-metabolic abnormality (LCGA).
METHODSThe patients with LCGA enrolled in the randomized controlled clinical trial were assigned to 2 groups, the treated group (68 cases) and the control group (74 cases), they were treated respectively by FHGP and conventional TCM and Western medicine therapeutic program for 3 months. Indexes including fasting plasma glucose (FPG), 2 h postprandial plasma glucose (2 h PG), fasting insulin (FINS) were detected, homeostasis model assessment insulin resistance (HOMA-IR) was calculated, and the score of syndrome was recorded before and after treatment. Then the effects on syndrome and glyco-metabolic abnormality were evaluated through statistical analysis.
RESULTSLevel of 2 h PG after treatment was lowered in both groups (P < 0.01), but significant difference was found in the pre-treatment to post-treatment decrement of FPG and HOMA-IR between the two groups (P < 0.05). The syndrome improving rate and the total effective rate on glyco-metabolic abnormality in the treated group were significantly better than those in the control group respectively (85.3% vs 64.9% , P < 0.01; 80.9% vs 62.2%, P < 0.05).
CONCLUSIONFHGP has the capability to improve the syndrome and glyco-metabolic abnormality of patients with LCGA.

9.Role of mitochondrial calcium uniporter in cardioprotection induced by ischemic postconditioning in isolated rat heart.

OBJECTIVETo investigate the role of mitochondrial calcium uniporter in cardioprotection elicited by ischemic postconditioning (Postcond).
METHODSMale Sprague-Dawley rats were used for Langendorff isolated heart perfusion. The hearts subjected to global ischemia for 30 min followed by 120 min of reperfusion. Left ventricular developed pressure (LVDP), maximal rise/fall rate of left ventricular pressure (± dP/dtmax) were measured. The level of lactate dehydrogenase (LDH) in the coronary effluent was measured spectrophotometrically, the content of formazan of myocardium was also measured at the end of reperfusion.
RESULTCompared to I/R group, Postcond had an significant increase in the mechanical function of the left ventricle, with LDH release reduced and the content of formazan increased. Spermine, the opener of mitochondrial calcium uniporter, deteriorated the mechanical function of left ventricle and decreased the formazan content, and increased LDH release. Ruthenium red, the inhibitor of mitochondrial calcium uniporter, increased the mechanical function of the left ventricle, decreased the LDH release, but the content of formazan was not increased.
CONCLUSIONThe inhibition of mitochondrial calcium uniporter is involved in the mechanisms of ischemic postconditioning.

10.Therapeutic bloodletting at Jing-well points combine hypothermia attenuated acute cerebral edema after traumatic brain injury in rats.

OBJECTIVETo investigate the influence of therapeutic bloodletting at Jing-well points and hypothermia on acute cerebral edema after traumatic brain injury (TBI) in rats.
METHODSSeventy-five SD rats were randomly divided into sham-operation group (Sham), TBI group (TBI), bloodletting group (BL), mild-induced hypothermia group (MIH), and bloodletting plus MIH group (BL + MIH) (n = 15). The model of TBI was established by electric controlled cortical impactor (eCCI). The rats of BL group were bloodletting at Jing-well points immediately after injury, twice daily. While the MIH group was settled on a hypothermia blanket promptly after TBI for 6 hours, so that the temperature dropped to 32 degrees. Each of measurement was performed after 48 hours. Magnetic resonance imaging (MRI) was used to evaluate the dynamic impairment of cerebral edema after TBI (n = 3). In addition, mNSS score, measurements of wet and dry brain weight, and Evans Blue assay were performed to investigate the neurologic deficit, cerebral water content (n = 8), and blood-brain barrier permeability (BBB), (n = 4), respectively.
RESULTSMRI analysis showed that the cerebral edema, hematoma and midline shifting of rats in TBI group was more serious than other treatment group. Meanwhile compared with TBI group, the mNSS scores of every treatment group were meaningfully lower (all P < 0.05). Furthermore, treatment with BL+ MIH group was superior to the separated BL and MIH group (all P < 0.01). In addition, brain water content of each intervention group reduced to varying degrees (all P < 0.05), especially that of MIH group and BL + MIH group (P <0.01). BBB permeability of each treatment group was also significantly improved (all P < 0.01), and the improvement in MIH group and BL + MIH group was much better than the BL alone group (P < 0.05, P < 0.01).
CONCLUSIONOur major finding is that bloodletting at Jing-well points and MIH can reduce cerebral edema and BBB dysfunction and exert neuroprotective effects after TBI. The results suggest that the combination of BL and MIH is more effective than other treatment being used alone.
