Zonulolytic effects of alpha-chymotrypsin and trypsin were examined on rabbits' eyes using an apparatus which enables one to press the enucleated eyeballs. The time interval between the applications of enzymes and the protrusion of the lens was measured. The results showed that the trypsin was 4 to 5 times more effective than alpha-chymotrypsin.

4.Efficacy of procedure of the dissociation of the monkey renal cell by transfusion of trypsin

Trypsinization method in our modification was harvested high yields of dissociated cells. The total of cells is obtained 19.47 X 108 per pair of monkey kidney weighted 10 gram. Whereas rate of available cell reach 83.9% and rate of single cell and small group (<5 cells) collected was about 90%.

5.In vitro screening of natural compounds with antitumor activity by trypsin.

In order to investigate the feasibility of in vitro screening the antitumor activity of natural compounds by trypsin, porcine trypsin was used to for screening test, which is marked by inhibition of enzyme activity. Four compounds, namely daidzin, genistin, matrine and oxymatrine, were selected as test subjects. The natural antitumor drug camptothecin was used as the control. The inhibitory effect was detected by two experimental methods: direct detection of trypsin activity inhibition and hydrolysis of bovine serum albumin by trypsin. The results showed the inhibitory effects of the four natural compounds on trypsin, and the inhibition rates of the four natural compounds were significantly different. The enzyme activity assay showed that the inhibitory effect of matrine was better than that of oxymatrine, indicating that trypsin had a good screening resolution. The inhibitory effect was significantly increased with the increased ratio of sample to trypsin, suggesting the structure-activity correlation and dose-effect correlation of the screening methods. Altogether, the experimental method of screening antitumor activity of natural compounds by trypsin has good application values. Since porcine trypsin is similar to human trypsin in terms of molecular structure and performance, it is more applicable for screening of antitumor efficacy of natural pharmacodynamic compounds.

6.Uptake of Ga-67 by Cultured Cells: Transferrin-dependent and Transferrin-independent Mechanisms.

PURPOSE: We determined whether the uptake of Ga-67 by cultured cells occur by both transferrin (Tf)-dependent and independent mechanisms and the mechanism and magnitude of its uptake may vary as the degree of expression of the transformed phenotype. MATERIALS AND METHODS: Uptake of Ga-67 between the tansformed and untransformed cells was compared. Cells were incubated with Ga-67 in either the presence or absence of Tf and with complete medium containing Ga-67 after preincubating with anti-Tf receptor antibodies at 37oC in 8% CO2. Monolayers of cells were washed and trypsinized. Radioactivity and protein content of the samples were determined. RESULTS: Uptake of Ga-67 by cultured cells occurred both in Tf-bound and ionic form and was increased with radioactivity and time. The magnitude for the uptake of Tf-bound form was approximately 3 and 6-fold greater than ionic form. In the presence of Tf, uptake of Ga-67 was 2-fold greater in the transformed cells. Conversely, In the absence of Tf, it was 1.5-fold greater in the untransformed cells. Regardless of blocking the Tf receptor by anti-Tf receptor antibodies, a significant amount of intracellular Ga-67 uptake was found. CONCLUSION: Dual mechanisms exist for the uptake of Ga-67 by cultured cells. The primary important one was the Tf-dependent system. Tf-dependent and independent mechanisms and the magnitude operated oppositely in the transformed cells when compared to their untransformed counterpart.

7.The Effects of Alpha-Chymotrypsin and Trypsin on the Rabbit's Cornea.

The posterior surface of the rabbit's cornea was scraped by a spatula or incised by a cystotome, followed by injection of alpha-chymotrypsin or trypsin into the anterior chamber. The concentration of enzymes used were 5mg/cc and O.5mg/cc of alpha-chymotrypsin and 5.000 Anson unit/cc and 500unit/ cc of trypsin. Three minutes after the introduction of enzymes into the anterior chamber, irrigation of the anterior chamber with normal saline was performed. The cornea was examined macroscopically and biomicroscopica11y for about 3 weeks. Histological examination was done on the 3rd and the 11th post-operative days. The cornea was affected more intensively and extensively when the stroma was injured than when only the endothelium was injured. Alpha-chymotrypsin was more toxic to the cornea than trypsin and led sometimes to a corneal perforation.

8.Effects of Trypsin, Collagenase and Dimethyl Sulfoxide on Dissociation of Rat Heart Cells.

New born rat heart cells were dissociated using trypsin and/or collagenase to elucidate the dissociation efficiency of these two enzymes. And the effect of dimethyl sulfoxide during and immediately after cell dissociation was also investigated to clarify the so-called protective activity of dimethyl sulfoxide on cell performance. The results can be summarized as follows. 1. Cold trypsin 18 hours pretreatment followed by warm collagenase treatment resulted best cell viability and cell yield. 2. Single warm trypsin treatment gave the poorest result. 3. Dimethyl sulfoxide did not seem to play any protective role during or immediately after rat heart cell dissociation. It had very damaging effect on rat heart cells.

9.The Effect of Alpha-chymotrypsin, Trypsin and Hyaluronidase on the Viscosity of the Rabbit's Vitreous Humor.

Alpha-chymotrypsin, trypsin and hyaluronidase were added to the rabbit's vitreous humor which was filtered through the glass filter. The viscosity was measured by Ostwald type viscosimeter with O.5mm capillary bore size. The relative viscosity of normal vitreons humor was 1.085. The relative viscosities of vitreous humor, to which 1mg and 2mg of alpha-chymotrypsin and 100 H.U.M. and 200 H.U.M. of trypsin were added, showed no significant changes. On the other hand, the vitreous humor which had been treated by 100 T.R.U. of hyaluronidase showed a marked reduction in the viscosity. The viscosity of normal vitreous humor, which had been preserved at 20 degrees for 3 hours, was significantly reduced.

Proteomics is a powerful subject focusing on large-scale study of protein structures and functions. A complete enzymatic digestion of protein complexes is the key step in modern high-resolution and high-throughput mass spectrometry (MS)-based identification and quantification. To achieve MS analysis, both peptide sample pretreatment and data acquisition are prerequisite in proteomic studies. In this paper, we summarized both the enzymatic proprieties of three common proteolytic enzymes, Trypsin, Lys-C and Glu-C, the optimization of multi-enzyme combination and an advanced sample pretreatment in proteomics research.
