In the face of escalating problems with pathogen control, the development of proper formulations of existing antibiotics is as important as the development of novel antibiotics. Daptomycin is a lipopeptide antibiotic with potent activity against Gram-positive bacteria. Currently, only injectable solution of daptomycin has been approved for clinical use. In the present study, the formulation of PEGylated liposomal daptomycin (PLD) was prepared and optimized, and its efficacy against methicillin-resistant Staphylococcus aureus (MRSA252) strains was investigated. The obtained PLD had a mean vesicle diameter of (111.5 +/- 15.4) nm and a mean percent drug loading of (5.81 +/- 0.19) % with high storage stability. Potent activity of PLD against MRSA was demonstrated in vitro with a more sustained effect than that of conventional liposomal daptomycin and daptomycin solution. In addition, intravenous administration of a single dose (equal to human use) of PLD significantly increased the survival of mice in a MRSA252 systemic infection model compared with other formulations. Drug distribution in the lung was significantly enhanced following administration of PLD, and no measurable tissue lesions or pathological changes were detected during PLD treatment. Taken together, PEGylated liposomes loaded with daptomycin may represent a promising approach to reduce MRSA252 infections, especially those involving bloodstream dissemination, such as hematogenous pulmonary infection.

0.05).Conclusion GH improves FAH of children with GHD.Height at the initiation of puberty is the most significant determining factor for the long-term efficacy.Hence,it is important that the diagnosis should be made and treatment be initiated as early as possible to afford children with GHD the opportunity to make up much of their height deficit before puberty.Adequate dosage of GH should be used for the children taking initial treatment at puberty to attain satisfactory FAH.

Objective To develop an accurate, reliable method without the need of initialization in animal PET modeling for estimation of the tracer kinetic parameters based on the artificial immune network. Methods The hepatic and left ventricular time activity curves (TACs) were obtained by drawing ROIs of liver tissue and left ventricle on dynamic 18F-FDG PET imaging of small mice.Meanwhile, the blood TAC was analyzed by sampling the tail vein blood at different time points after injection. The artificial immune network for parametric optimization of pharmacokinetics (PKAIN) was adapted to estimate the model parameters and the metabolic rate of glucose (Ki) was calculated. Results TACs of liver, left ventricle and tail vein blood were obtained.Based on the artificial immune network,Ki in 3 mice was estimated as 0.0024,0.0417 and 0.0047,respectively.The average weighted residual sum of squares of the output model generated by PKAIN was less than 0.0745 with a maximum standard deviation of 0.0084,which indicated that the proposed PKAIN method can provide accurate and reliable parametric estimation. Conclusion The PKAIN method could provide accurate and reliable tracer kinetic modeling in animal PET imaging without the need of initialization of model parameters.

Objective To discuss the effects of different drying methods on composition and antioxidative activities of the volatile oil fromCymbopogon citrates; To optimize the best drying method for Cymbopogon citrates. MethodsCymbopogon citrates was dried by drying in the sun, drying in the shade and oven drying at 40℃. Volatile oil was extracted by steam distillation. Chemical constituents in the volatile oil were analyzed by GC-MS and the antioxidative activities were determined by ferric reducing antioxidant power (FRAP method). Results Extraction rate of the volatile oil fromCymbopogon citratesunder the environment of freshness, sun drying, shade drying and oven drying at 40℃ were 0.25%, 1.21%, 1.19% and 1.17%, respectively; after dried by different methods, main constituents and antioxidative activities of the volatile oil fromCymbopogon citrates were basically same. Conclusion Different drying methods have little influence on composition and antioxidative activities of the volatile oil fromCymbopogon citrates. Oven drying at 40℃ was the best way to dryCymbopogon citrates.

AIM: To construct a lentiviral vector harboring RNAi sequence targeting human annexin A2 (ANX A2) and to investigate the functions of ANX A2 in antiphospholipid antibody (APL)-induced tissue factor (TF) expression in monocytes. METHODS: Four different short hairpin RNAs (shRNA) targeting ANX A2 gene were constructed and cloned into the pGCSIL-GFP vector. After identification with PCR and sequencing, the effective interference sequence was further determined by Western blotting analysis. The recombinant lentivirus LV-RNAi-ANX A2 harvested from 293T cells was transferred into THP-1 cells and the ANX A2 mRNA and protein expression on THP-1 cells were examined. The transferred-THP-1 cells were stimulated by APL/β_2GPI compound, and the TF mRNA and TF activity was assayed. RESULTS: The RNAi sequences targeting the human ANX A2 gene were successfully inserted into the lentiviral vector and the high-performance RNA interference sequence was sieved out. The recombinant lentivirus was harvested from 293T cells with a viral titer of 3×10~(12) TU/L. THP-1 cells infected with LV-RNAi-ANX A2 showed almost lockout of ANX A2 both at mRNA and protein levels. The TF expression was also significantly decreased in the transferred-THP-1 cells induced by APL/β_2GPI compound. CONCLUSION: The lentiviral vector constructed in the present study blocks the ANX A2 expression in THP-1 cells and further inhibits the TF expression induced by APL/β_2GPI compound, which indicates that ANX A2 do play an important role in APL-induced TF expression on monocytes.

Microbiology 2008;0(12):-
A strain, Pseudomonas sp. X-2-45, with high and stable lipolytical activity was screened by continuously subculturing a lipase-producing bacterium P. sp. LP-1 in culture medium containing Jatropha oil as a sole carbon source. Its hydrolytic activity was 29.79 U/mL, which was increased by 288% as compared to that of parent strain. Furthermore, the growth and lipase synthesis of X-2-45, its catalytic ability to hydrolyze vegetable oils, as well as ester synthesis between fatty acids and organic alcohols were studied. Results showed that rates of bacterial growth and lipase synthesis were significantly raised. Bacterial biomass and lipase activity reached the highest level after 30 h of incubation. Moreover, growth stationary period was prolonged and lipase produced exhibited good stability in culture media during incubation period. Hydro- lytic activity of P. sp. X-2-45 lipase toward Jatropha oil was increased by 378% as compared to parent strain, suggesting that acclimation to Jatropha oil was an effective approach for improving substrate selectivity oflipase. Finally, results of ester synthesis catalyzed by P. sp. X-2-45 lipase indicated that this lipase could catalyze esterification reactions between lauric acid and n-butanol, n-octanol, 1-dodecanol or glycerol, palmitic acid or stearic acid and methanol, n-octanol, 1-dodecanol or glycerol, oleic acid and methanol, n-butanol, n-octanol, 1-dodecanol or glycerol.
