1.Separation and Content Comparison of Vitexin from Anemarrhenae Rhizoma Before and After Salt Processing

Objective To study the chemical components of Anemarrhenae Rhizoma;To establish a method to determine the content of vitexin;To compare the content difference of vitexin from Anemarrhenae Rhizoma before and after salt processing. Methods 70% ethanol extract of Anemarrhenae Rhizoma was separated and purified by silica gel column chromatography technique, and compound was identified by physicochemical properties and spectral data. HPLC was used with Ecosil column (4.6 mm×150 mm, 5μm), mobile phase of acetonitrile-0.2%acetic acid solution (14.5∶85.5), velocity of 1.0 mL/min, and determine wavelength of 340 nm. Results The compound was vitexin, a good linearity (r=0.999 8) in the range of 0.039 8-1.99μg. The average recovery rate was 98.02%, RSD=0.21%. Vitexin content was 0.008 5% in crude Anemarrhenae Rhizoma, 0.008 1% in processed products. Conclusion This is the first time separation of vitexin from Anemarrhenae Rhizoma. The method for content determination of vitexin is accurate, specific, and highly sensitive in the present experiment. Content of vitexin decreases slightly in processed products of Anemarrhenae Rhizoma compared with crude Anemarrhenae Rhizoma.
2.Pioglitazone ameliorating radiation fibrosis in rat hearts

Objective To explore whether pioglitazone can ameliorate radiation-induced fibrosis in rat heart.Methods A total of 46 Sprague-Dawley rats were divided into six groups (control group, pioglitazone (Pio) 10 mg·kg-1 ·d-1 group, Pio 20 mg·kg-1 ·d-1 group, 18 Gy irradiation + placebo group;18 Gy irradiation + Pio 10 mg·kg-1 ·d-1 , and 18 Gy irradiation + Pio 20 mg·kg-1 ·d-1 group).Experimental animals were exposed to radiation at the chest, then administered Pio or placebo for one month.At 3 months later, the rats were killed and their heart tissues were collected for Masson staining, Western blot analysis and real-time polymerase chain reaction assay (real-time PCR).Results Masson staining revealed significant myocardial fibrosis in rats exposed to radiation, while these changes were reduced when the rats were given Pio.Western blot analysis showed that the PPAR-γ protein expression in the heart tissue of irradiated rats were higher than in the non-irradiated group (F =12.435, P < 0.05).Real-time PCR assay showed that PPAR-γ mRNA expression in the irradiation + Pio 20 mg· kg-1· d-1 group was higher than that in the Pio 20 mg· kg-1 ·d-1 group (F =2.333, P < 0.05).The expression of TGF-β1 protein in the irradiation + placebo group were higher than those in the other five groups (F =17.578 ,P <0.05), and the CDK5 protein expression had a high level in the three irradiated groups while only the irradiation + placebo group was statistically higher than controls (F =3.651, P < 0.05).Conclusions Pioglitazone may ameliorate radiation fibrosis in the rat heart through its anti-fibrotic activity, perhaps via inhibiting Cdk5-mediated PPAR-γ phosphorylation.

5.Clinical Observation of Acupuncture plus Paroxetine for Depression Due to Liver-qi Stagnation

Objective To observe the clinical efficacy of acupuncture plus Paroxetine in treating depression due to liver-qi stagnation, for seeking a more effective treatment for this disease.Method Sixty patients with depression due to liver-qi stagnation were randomized into a treatment group and a control group, 30 cases in each group. The treatment group was intervened by acupuncture plus Paroxetine, while the control group was by Paroxetine alone, successively for 8 weeks. Hamilton Depression Scale (HAMD), Chinese medicine syndrome and sign score, and central neurotransmitters were observed before and after the treatment, for evaluating the therapeutic efficacy.Result The total effective rate was 96.6% in the treatment group versus 71.4% in the control group, and the treatment group was superior to the control group (P<0.05).Conclusion Acupuncture plus Paroxetine is an effective method in treating depression due to liver-qi stagnation.

Objective To study the effects of choline chloride on activities of myosin adenosine triphosphatase (mATPase) of muscle fibres and muscle atrophy in tail-suspended rats. 9.The serum homocysteine levels and dynamic changes in patients with different etiologic macrocytic anemia

Objective To examine the serum homocysteine,folic acid and Vitamin B12 levels in macrocytic anemia patients and observe their dynamic changes following therapy. Methods Homocysteine,folic acid and Vita-min B12 were analyzed by electro-chemiluminescence immunoassay. Complete blood cell count was analyzed by opti-cal method and resistance method. Results The homocysteine is significantly higher in nutritional megaloblastic a-nemia[ (71.26±27.84)μmoL/L ] than in drug-induced megaloblastic anemia[(11.44±5.06)μmol/L],in myel-odysplastic syndrome[ (9.51±4.13)μmol/L] and in the normal control group[(8.74±5.42)μmoL/L] (P<0.01). After treated with low-dosage folic acid and Vitamin B12,the patients with megaloblastic anemia presented slow declining but eventually normal homocysteine levels,compared with those received high-dosages. Conclusion Homocysteine can be used for differential diagnosis of macrocytic anemia. The duration of remaining of abnormal ho-mecysteine levels is related to the dosage of folic acid and Vitamin B12.
10.Clinical Analysis of Knee Osteoarthritis Treated Mainly by Acupuncture

Purpose: To observe the curative effects of different treatments on genual osteoarthritis.Methods: The 148 cases were randomly divided into 4 groups: acupuncture group; Chinese herbs plus spectrograp irradiation group; comprehensive treatments group and western medicine group. Results:The total effective rate in these groups were 78.9%, 72.7%, 92.3% and 78.9% respectively, and there was not a significant difference among the groups (P>0.05). The marked effective rate in comprehensive treatments group was higher than that in other three groups (P<0.05). Conclusion: Acupuncture and Chinese herbs plus sepectrograp irradiation had certain effect on gonitis. Comprehensive therapies could coordinate the actions of acupuncture, and Chinese herbs plus spectrograp irradiation, and raise the curative effect.
