Objective To set up a simple method to measure the SR Ca2+ release function in skeletal muscle strips. Methods The time from peak tension to 75 % relaxation(TR75) during intermittent tetanic contraction(fatigue contraction) of isolated musculus soleus strips was analyzed. It was prolonged rapidly, and then shortened slowly. The ratio of maximal TR75 to initial TR75(R-TR75) indicated a balance between SR Ca2+ release and uptake. If the SR Ca2+-ATPase(SERCA) activity and inhibition extent of SERCA in fatigue contraction were identical,R-TR75 should be an index for SR Ca2+ release function. Results Higher stimulation voltage or 5 mmol/L caffeine perfusion which increased the SR Ca2+ release channel open probability induced an increase of R-TR75 from 2.5 in control to 3.0.On the contrary, inhibition of SR Ca2+ release with 5 mmol/L magnesium sulfate perfusion significantly decreased R-TR75.Inhibition of SR Ca2+ release by metabolites during fatigue contraction was recovered slowly. So R-TR75 at the second fatigued contraction was reduced after 5 or 10 min of recovery, but no significant difference after 60 min of recovery.During intermittent tetanic contraction R-TR75 was increased to be 2.9 times of normal in unloaded soleus. Conclusion The above results suggest that R-TR75 during muscle strip fatigue contraction without any change of SERCA activity can indirectly reflected SR Ca2+ release function. In 2-week unloaded soleus, the enhanced R-TR75 indicates an increase in SR Ca2+ release function.

ObjectiveTo probe into the clinical efficiency of reconstruction to person who had composite tissue defect of the floor of mouth and the lower part of face with the free forearm flap and free iliac bone in the same term.MethodsBefore the surgery, the surgeon should carefully evaluate the scope and shape of defects in 11 cases who had the composite tissue defects of the floor of mouth and the lower part of face, to design the individual forearm flap, so that it matches with the defect region. During surgery, the first resumption of defects using titanium plates forming the basic shape of mandible and the occlusal relationship, then used to re-sawing to take modeling of the iliac bone, transplanted free iliac bone and fixed after the inside of the titanium plate, thereby restoring the continuity of mandible missing.ResultsFree forearm flap and free iliac bone all survived in 11 cases, the success rate of 100%. The patients were followed up for 6 to 12 months, although the shape of restoration areas were different levels of fat, but eating, swallowing and other oral function had been well improved, 7/11 could enter normal diet, 4/11 to enter liquid diet, At the same time, the patients in social activities can communicate in a language daily, sensory function of skin flap and facial appearance has been satisfactory recovery.ConclusionConformal free forearm flap and free iliac bone is an ideal way to reconstruct the composite tissue defects of the floor of mouth and the lower part of face in the same period, not only safe, practical prognostic effect, and also can significantly improve the patients quality of life, which is worthy of reference and use.

OBJECTIVETo screen and isolate secondary metabolite biosynthesis-related gene for establishing the foundation of functional gene research, we construct a cDNA library of Glycyrrhiza uralensis.
METHODTotal RNA was isolated from G. uralensis using the method of lithium chloride sedimentation. Double strand cDNA was joined into pBlueScript II vector. The number of clones, recombinant rate and length of insert fragments were determined.
RESULTThe capacity of the original library was 1.15 x 10(7) with a recombinant rate of 98.2% and the inserted cDNA fragments ranged from 0.5 to 4.8 kb. 126 ESTs through random sequencing were obtained. The most homological proteins came from leguminous plants, including Arabidopsis thaliana, Oryza sativa, and so on. Most of the proteins were related to genes linking cell matabolism, resistance, growth retardation and dormancy.
CONCLUSIONThe library has enough capacity, high recombinant rate and long insert fragment for the further study.
