1.Polymorphism of CDI4 gene promoter but not Toll-like receptor 4 gene is associated with colorectal cancer in Chinese patients

Objective To investigate an association between Toll-like receptor(TLR)4 Asp299Gly or CD14 C-260T polymorphisms and colorectal cancer in Chinese patients.Methods By a method of polymerase chain reaction-based restriction fragment length polymorphism(PCR-RFLP), TLR4 Asp299Gly and CD14 C-260T polymorphisms in unrelated 110 patients with colorectal cancer and 160 healthy controls in Chinese Han race population were genotyped.Results Significant differences of CD14 genotypes were found between healthy controls and the patients with colorectal cancer.The fre- quency of the CC genotype in healthy controls was extremely lower than that in the group of colorectal cancer(15.6% vs.31.8%,P=0.0027,OR=0.3968,95%CI:0.2209-0.7129)while the frequency of the CT genotype in healthy controls was significantly higher than that in the group of colorectal cancer (48.1% vs.30.9%,P=0.0056,OR=2.074,95%CI:1.246-3.452).No TLR4 Asp299Gly mutant genotype was detected in any patients and healthy controls in Chinese Han population.Conclusion The polymorphism of CD14 C-260T but not TLR4 Asp299Gly is associated with colorectal cancer in Chinese patients.
2.Risk Factors of Incision Infection after Liver Transplantation

OBJECTIVE To study and find reasonable methods of nursing care of incision through analyzing the risk factors in liver transplant patients.METHODS A total of 416 liver transplantation operations in our hospital from Sep 2004 to May 2006 were evaluated.The risk factors that resulted in the incision infection were considered.RESULTS The incision infection rate was decreased from 13.9% to 4.3% after improvement.CONCLUSIONS Nursing care is of great significance in preventing the incision infection in liver transplant patients.
3.Health management and metabolic syndrome in 207 employees

Objective To analyze the effects of health management of metabolic syndrome on the control of related risk factors. Methods A total of 430 patients diagnosed with metabolic syndrome in 2008completed questionnaire and before randomly assigned to the health management group ( n = 207 ) and the control group (n=223). One year later, all the participants were investigated for the risk factors of food intake, cigarette smoking, alcohol consumption, staying-up and emotion, and the changes of body mass index ( BMI), blood pressure ( BP), triglyceride (TG), high-density lipoprotein cholesterol (HDL) and fasting blood glucose (FBG). Student's t test and x2 test were used for data analysis. Results For the health management group, the incidence of high-salt, high-fat and high-sugar intake, overtake of food, lack of physical exercises,staying-up and alcohol drinking was significantly decreased at 1 year (P < 0. 05 ), when compared with baseline and the control group ( both P < 0. 05 ). For the health management group, BMI,systolic blood pressure (SBP),TG and FBG was improved at 1 year (P<0. 05). In comparison with the control group, BMI, SBP, diastolic blood pressure (DBP),TG and FBG of the management group were significantly improved (P < 0. 05 ). Conclusions Patients with metabolic syndrome may have better treatment compliance after receiving systematized and individualized health management.

5.A1AT alleviates pancreas exocrine cells damage to transplanted islets in mice

Objective To investigate the effect of alpha 1-antitrypsin (A1AT) concerning in reducing the injury of transplanted islets by pancreas exocrine cells and promoting proliferation of the pancreas B cells.Method The pancreases of mice were digested with collagenase,islets were isolated artificially,and pancreatic exocrine cells were collected.In purified islet group (n =6),100 islets were seeded into a 6 well culture plate.In experimental group(n =6),100 islets were co-cultured with equal volume of pancreas exocrine cells,and 0.5 mg/mL A1AT was added into a 6-well culture plate.In control group(n =6),100 islets were co-cultured with equal volume of pancreas exocrine cells.After 48 h,insulin content of islets in each well and trypsin concentration in the supernatant of each well were measured.The islets were cultured in low sugar and high sugar 1640 medium,then glucose stimulated insulin secretion (GSIS) test was carried out.In vivo,8-9-week old male BALB/C mice were induced with STZ (190 mg/kg body weight,i.p) to establish the diabetic model and randomly divided into two groups.In experimental(n =10) and control(n =10) groups,250 islets and the equal volume of pancreatic exocrine cells were transplanted into different regions of left kidney subcapsule,resepctively.The experimental group was injected with A1AT (83 mg/kg,qd,i.p) for 28 days after operation,and the control group was injected with the same amount of normal saline (qd,i.p) for 28 days.Both two groups were given EDU (5 μg/g,qd,i.p) for 28 days.The blood glucose level was monitored continually.Nephrectomies were performed after 28 days.The expression of anti-amylase antibodies in the renal subcapsule was detected by immunohistochemical staining,and the proliferation of islet beta cells was examined using immunofluorescence staining.Result Insulin levels and insulin stimulation index in the control group were decreased as compared with those in the purified islet group; those in the experimental group were higher than in the control group,but lower than in the purified islet group.Trypsin concentration in the control group was increased as compared with the purified islet group,that in the experimental group was lower than the control group,but higher than in the purified islet group (all P<0.01).After islets transplantation,the blood glucose levels in control and experimental groups were normal,but those in the control group recovered later than in the experimental group (P<0.01).At 3rd day after nephrectomy,the blood glucose levels were >21 mmol/L in both two groups.A large number of anti-amylase antibody-positive cells were found in the renal subcapsule in the control group while little seen in the experimental group after 28 days.The immunofluorescence showed that the insulin +/EDU + B cells in the experimental group were more than those in the control group.Conclusion Conclusion Co-culture of islets and pancreatic exocrine cells with A1AT can prevent islet cells from damage caused by trypsin.A1AT could inhibit the secretion of pancreatic amylase from pancreatic acinar cells and promote proliferation of islet beta cells.
6.Clinical evaluation of low vision and central foveal thickness in highly myopic cataract eyes after phacoemulsification

measured by OCT 1 and 3mo after surgery. Chi-square test was used to compare preoperative and postoperative BCVA. Pearsion’s correlation was used to evaluate relationship between postoperative BCVA and central foveal thickness.RESULTS:The ratio of BCVA<0. 05 was 30%,14%, 11%and 7% respectively for per-operation, 1wk, 1 and 3mo post - operation. After surgery, the central foveal thickness was significantly increased in group with BCVA<0. 3 comparing to group with BCVA≥0. 3. Three month post-operation, central foveal thickness was significantly decreased in both groups comparing to that in 1mo post-operation (P<0. 01). There has significant correlation between 3mo postoperative BCVA and central foveal thickness (r=-0.716, P<0.05).CONCLUSlON: ln this study, BCVA is improved after 3mo follow up. There has significant correlation between postoperative BCVA and central foveal thickness.

Objective This study aimed to explore the personal experiences and inward wodd of AIDS patients as parents in suburban areas of western China.Methods The phenomenological approach wag used in this study.The data was collected by in-depth interview.13 AIDS patients as parents who calne from suburban areas of western China were interviewed.The interview data were read repeatedly,coded,classified,analyzed and extracted.Results Six major categories emerged from the data:complex emotional fluctuations;confidentiality of the illness:isolation from social life;worsening economic situation;top priority to their children:deftciency of spiritual resorts.Conclusions AIDS patients as parents suffer considerable stress from psychology,economy and social life.It is needed that strengthening psychological support to them,broadening financial accesses for them,understanding and respecting them to improve their living conditions.
8.Effect of acyl-CoA carbrolase antisense oligonucleotide on non-alcoholic fatty liver cell model

Objective To observe the effect of acyl-CoA carbrolase (ACC) antisense oligonucleotide (ASODN) on steatosis of HL-7702 hepatocytes.Methods The specific phosphorothioate modified ACC targeting ASODN were designed and synthesized by a computer software.Normal HL-7702 hepatocytes were cultured in vitro using intravenous lipid emulsions,a large number of triglyceride was induced in hepatocytes within 48 h,and a model of fatty liver cells in vitro was successfully formed.ACC-ASODN was transfected into the hepatocyte model by liposome method.The blank group (HL-7702 cells),model group (fatty liver cells of model) and low and high concentrations of the two experimental groups (5,10 μmol · L-1 ACC-ASODN) were set up.ACC-ASODN was transfected into fatty liver cell model 24 h later.ACC activity was determined by radioisotope technology.The malonyl coenzyme A content was determined by HPLC.The carnitine palmitoyltransferase Ⅰ (CPT Ⅰ) activity was determined by radioisotope technique.The level of triacylglycerol(TG) was determined by automatic analyzer.Results The value of the liver cell ACC activity (U) in blank group,model group and experimental-L,-H groups were respectively 0.74,0.33,0.20,0.18.The value of the intracellular content (nmol · L-1) of malonyl CoA in above four groups were respectively 14.3,12.4,7.5,6.8.The value of CPT Ⅰ (U) in above four groups were respectively 1.33,1.47,3.85,3.95;the value of TG conten(mmol · L-1) in above four groups were respectively 0.20,0.52,0.37,0.28.Compared with blank group,the factors in model group had significant differences(all P < 0.05).Compared with model group,the factors in experimental-L,-H groups had significant differences (all P < 0.01).Conclusion ACC-ASODN acts on hepatocytes,and by inhibiting the activity of ACC,the content of TG is decreased,and the steatosis of hepatocytes is alleviated.
9.Treatment of femoral intertrochanteric instable fractures with cemented artificial bipolar arthroplasty for senile patients

Objective To discuss the feasibility and effects of cemented artificial bipolar arthroplasty for senile patients with femoral intertrochanteric instable fractures. Methods From January 2004 to June 2008,46 aged cases with femoral intertrochanteric instable fractures were cured with cemented artificial bipolar arthroplasty. According to improving-classfication of Evans,there were 19 cases with type Ⅲfractures and 27 cases with type Ⅳ fractures. Results All cases were successfully operated. The surgery time was 50-90 (68.00 ±20.43) minutes. Bleeding volume was 160-600 (210.00 ±40.26) ml. The time in hospital was 16-26 (19.30 ± 4.56) days. All cases were retrospectively reviewed for 6-39 (17.10 ± 4.33)months. Three cases died of other diseases, 43 cases were graded by the system of Harris. The Harris scale was 70-94 (86.70 ±5.16) scores,and the choiceness rate was 90.7%(39/43). No dislocation,loosening,subsidence and late infection occurred in all the implants. Conclusions Cemented artificial bipolar arthroplasty has good advantages of reduced laying up period and few complications. The short-term outcome is satisfactory.
10.Effect of suppressive oligodeoxynucleotides on the functional differentiation in CD4(+) Th1 lymphocytes in mice in vitro.

OBJECTIVE:
To explore the effect of suppressive oligodeoxynucleotides (Sup ODN) on the Th1 differentiation of CD4(+)T splenetic lymphocytes in mice.
METHODS:
The splenetic lymphocytes of BALB/c mice were separated, and then CD4(+) cells were purified with immune magnetic CD4(+) microbeads (positive selection). The purification was examined by fluorescence-activated cell sorter. CD4(+) cells, anti-CD3epsilon, anti-CD28, IL-12 and Sup ODN or control oligodeoxynucleotides (Con ODN) were co-incubated for 72 h. IFN-gamma and IL-4 in the supernatant were detected using enzyme-linked immunosorbent assay. The expression of T-bet mRNA in CD4(+) cells was tested by reverse transcription polymerase chain reaction.
RESULTS:
Sup ODN could significantly inhibit the release of INF-gamma and increase IL-4 production respectively (P<0.01). T-bet mRNA of CD4(+) lymphocytes was remarkably inhibited by Sup ODN as well (P<0.01).
CONCLUSION
In the presence of pro-Th1-cytokines, Sup ODN may affect the differentiation of CD4(+) T lymphocytes in vitro. Sup ODN can promote CD4(+) T cells to differentiate into Th2, and suppress them into Th1.
