1.Inheritance of academic idea and experience about using traditional Chinese medicine from JIN Shi-yuan.

Professor Jin Shi-yuan has been worked in traditional Chinese medicine (TCM) over 70 years. He made prominent contributions in identification, processing, dispensing of TCM and reasonable use proprietary Chinese medicine. In over 70 years, he has mastered herbal medicine and traditional Chinese Medicine. It is also professor JIN's academic characteristic. Professor JIN's practical experiences were summarized according to the current situation about clinical medication, change of species of Juhong and Chenpi has been different from species of medical history. The quality is lower than before. Medicinal parts of Danggui, Gancao, Huangqin and Wuyao has been changed. So the actions of these herbal medicines have been changed also. Fresh herbal Qianchangpu has disappeared but it should be used clinically. Medical history, change of species, change of medicinal part, and change of preparing process in professor JIN's academic idea were be summarized periodically. The result is hoped to be referred by administration, manufacture, medical treatment of TCM.

4.Expressions and clinical significances of Hepcidin, BMP6 and HJV in gastric cancer

Objective To study the expressions of Hepcidin,bone morphogenetic protein 6 (BMP6) and hemojuvelin (HJV) in gastric cancer,and to explore their relationships with clinical pathological characteristics.Methods Hepcidin,BMP6 and HJV were detected by immunohistochemistry in 62 cases of gastric cancer patients,and the relationships among them and clinical pathological features were analyzed.Results Compared with normal gastric mucosa tissues,Hepcidin was highly expressed in gastric cancer tissues (13.3% vs.56.5%,x2 =8.99,P < 0.01),while the differences of the expression of BMP6 and HJV in the two groups were not statistically significant (60.0% vs.40.3 %,x2 =1.89,P > 0.05;93.3% vs.83.9%,x2 =0.88,P > 0.05).The expression of Hepcidin was related to T stage (x2 =6.98,P =0.02),but it was not related to age,sex,lymph node metastasis,distant metastasis and anemia.The expressions of BMP6 and HJV were not related to T stage,age,sex,lymph node metastasis,distant metastasis and anemia.Hepcidin was not related to the expressions of BMP6 and HJV (r=0.13,P>0.05;r=0.15,P>0.05).Conclusion Hepcidin is highly expressed in gastric cancer tissues,which is related to the T staging of gastric cancer,and can be used as an objective indicator of the biological behavior of gastric cancer.There were no differences in the expression of BMP6 and HJV between normal gastric mucosa and gastric cancer tissues.
5.Inhibition of invasive and chemotactic abilities of SKOV3 cells bv human epithelial growth receptor-2 small interfering RNA

Objective To investigate the effects of RNA interference(RNAi)targeting human epithelial growth receptor-2(HER-2)gene on the invasive and chemotactic ability of SKOV3 cells. Methods Glyeeraldehyde-3-phosphate dehydrogenase(GAPDH)was used as the positive control. Lipofectamine 2000 mediated transient transfection was conducted to transmit the siRNA into SKOV3 cells. Three pairs of specifically targeted(HER-2 siRNA Ⅰ,HER-2 siRNA Ⅱ,HER-2 siRNA Ⅲ)sequence were selected in the coding region of HER-2 mRNA.Transfection of HER-2 siRNA was conducted with lipofeetamine 2000 in ovarian carcinoma cell line SKOV3.The HER-2 gene expression was assessed by real- time PCR and western blot assays.Changes of invasive and chemotaetie capacity of SKOV3 cells were measured by polycarbonates coated with or without matrigal.Results Western blot results showed that the expression of GAPDH protein was decreased in specifically transfected cells and with the increase of siRNA dose,the expression of GAPDH protein was decreased.GAPDH protein gray value in control group, different doses(0.5,1.0,1.5,2.0 ?g)GAPDH siRNA interference groups were 0.6855?0.0259,0.5698 ?0.0275,0.4542?0.0296,0.3341?0.0178 and 0.1816+0.0180,respectively.There was a significant difference in each group(F=198.126,P
6.Relationship between level of serum erythropoietin and chemotherapy in elderly patients with cancer

Objective To study the relationship between serum erythropoietin(Epo)level and chemotherapy-induced hemoglobin(Hb)level in elderly patients with cancer. Methods One hundred senile patients(aged≥60 years)with cancer and one hundred and seventy non-senile patients(aged<60 years)with cancer were selected as aged group and control group,respectively.The serum Epo level was detected by enzyme-linked immunosorbent assay(ELISA). Results (1)The average levels of the serum Epo in aged group and control group were(22.0±15.1)U/L and(30.4±21.8)U/L,respectively(t=1.2988,P>0.05).A negative correlation was found between the levels of Epo and Hb in elder cancer patients(r=0.3700,P<0.001).(2)The levels of serum Epo were(14.7±10.6)U/L,(20.2±9.0)U/L and(42.3±24.8)U/L in 55 cases with normal Hb level,22 cases with mild anemia and 23 cases with moderate to severe anemia,respectively(F=11.6886,P<0.01).(3)The levels of Epo were(20.2±10.8)U/L,(45.2±39.1)U/L and(25.8±15.9)U/L before chemotherapy,after 4 treatment cycles and 2 treatment cycles respectively(F=4.5477,P<0.01).The levels of Hb were(111.0±20.5)g/L,(96.8±16.6)g/L and(102.1±19.3)g/L before chemotherapy,after 4 treatment cycles and 2 treatment cycles respectively(F=4.0071,P<0.01).Conelusions There is no statistical difference in serum Epo level between senile patients with cancer and non-senile patients with cancer.but serum Epo levelis higher in patients with anemia than without anemia.There is a negative correlation between the levels of Epo and Hb.Chemotherapy can decrease the level of Hb and increase the serum 1evel of Epo in part of patients.The changes are correlated with the conditions of patients before chemotherapy and the time of chemotherapy.

8.Cloning and prokaryotic expression analysis of squalene synthase 2 (SQS2) from Salvia miltiorrhiza f. alba.

According to the designed specific primers of gene fragment based on the Salvia miltiorrhiza transcriptome data, a full-length cDNA sequence of SQS2 from S. miltiorrhiza f. alba was cloned by the method of reverse transcription polymerase chain reaction (RT-PCR). The SmSQS2 cDNA sequence was obtained, this sequence is named SmSQS2 and its GenBank registration number is KM244731. The full length of SmSQS2 cDNA was 1245 bp, encoding 414 amino acids including 5'UTR 115 bp and 3'UTR 237 bp. Sequence alignment and phylogenetic analysis demonstrated that SmSQS2 had relative close relationship to the SQS2 of S. miltiorrhiza. The induction of E. coli [pET28-SQS2] in different temperature, induction time, IPTG concentrations and density of inducing host bacterium (A600) were performed, Shaking the culture at 30 degrees C until the A600 is approximately 0.6 and add IPTG to final concentration of 0.2 mmol x L(-1), and then the optimal expression of SmSQS2 recombinant protein were accumulated after the induction time of 20 h. The research provided important base for the study of sterol and terpene biosynthesis of SQS2 in S. miltiorrhiza f. alba.

9.The effect of beta-amyloid precursor protein peptide on the expressions of PDK, PKB, p-CREB in the neurons of hippocampal gyrus in rats after cardiopulmonary resuscitation

Objective To explore the effects of beta-amyloid precursor protein (APP17) peptide on the changes in the expressions of phosphoinositide 3-kinase(PI3K), protein kinase B(PKB) and phosphorylation of cAMP response element binding protein (p-CREB) in the neurons of hippocampal gyms in rats after cardiopulmonary resuscitation. Method Twenty-one Wistar rats were randomly divided into three groups, namely the sham-operated control group, the resuscitation group and resuscitation with APP17 peptide-treated group. The rat model of asphyxial cardiac arrest was made by clamping the endotracheal tube and the standard external cardiopulmonary resuscitation ( CPR) was performed until the restoration of spontaneous circulation ( ROSC) observed.ROSC was defined by the appearance of normal QRS waves of electrocardiogram and mean artery pressure ( MAP)≥60 mmHg for more than 10 minutes. Rats of resuscitation group and control group received intravenous 0.9%NaCl, and the rats of the APP17 peptide group were treated with APP17 peptide(10μg·300 g~(-1), i. v.) after ROSC. Rats were sacrificed by decapitation after reperfusion 2 hours and then the cerebral hippocampal gyrus was immediately separated to detect PI3K, PKB and p-CREB by immunohistochemistry ( IHC) and Western-blot analysis. Statistical comparisons were made by one-way analysis of variance (ANOVA) . Results IHC showed that there was no significant difference in PDK positive cells between resuscitation group and control group (2.75 ±1.80 vs. 2.53 ± 1.53, P > 0.05) . The PDK obviously more increased in the APP17 peptide group than in resuscitation group(5.85 ± 2.83 vs. 2.75 ± 1.80, P < 0.01) .The counts of PKB and p-CREB positive cells were obviously lower in resuscitation group than those in control group (2.45 ± 1.36 vs. 5.22 ± 2.50, P < 0.05);(2.41 ± 1.11 vs. 8.31 ±3.02, P < 0.01 ). The PKB and p-CREB positive cells were significantly higher in the APP17 peptide group than in resuscitation group (9.66±4.32 vs. 2.45 ±1.36, P < 0.01);(14.18 ± 3.96 vs.2.41 ± 1.11, P <0.01). The PKB and p-CREB protein levels in resuscitation group were lower than those in the control group. Conclusions The expressions of PKB and p-CREB in neurons of hippocampus gyrus 2 hours ROSC, and APP17 peptide could restorer the expression of PDK, PKB and p- CREB and thereby protect the neurons of hippocampus gyrus from the injury of CPR.
10.The therapeutic effects of combination of γ-aminobutyric acid, sodium dimercaptopropane sultanate and vitamin B6 in large doses on liver and heart in rats with acute tetramine intoxication

Objective To observe the therapeutic effects of the co-administration of γ-aminobutyric acid (CABA), sodium dimercaptopropane sulfonate (Na-DMPS) and vitamin B6 in large doses on liver and heart of rats with acute tetramine intoxication, and compare their separate effects of either GABA or Na-DMPS alone with those of the triad combination. Method Thirty rats were randomized into control group (n = 6), tetramine intoxication without treatment group (n = 6), tetramine intoxication treated with GABA group (n = 6), tetramine intoxication treated with Na-DMPS group (n = 6) and tetramine intoxication treated with triad combination (GABA + Na-DMPS + vitamin B6, GNDV n = 6) group. Samples of blood, liver tissue and heart tissue of rats with acute tetramine intoxication were collected immediately two hours after medication with different drugs. Serum alanine aminotrasferase (ALT), aspartate aminotransferase (AST), creatine kinase (CK) and creatine kinase isoenzyme (CK-MB) were measured, and the pathological changes of liver tissue and heart tissue were observed under microscope. Results The symptoms of poisoning were apparently relieved and the latency for convulsion/muscular twitch were obviously delayed in poisoned rats treated with GABA, Na-DMPS and GNDV separately. Furthermore, combination group showed the latent period delayed longer than either GABA or Na-DMPS groups The GABA, Na-DMPS and GNDV significantly lowered the serum levels of ALT, AST, CK and CK-MB in rats with tetramine intoxication, and those serum levels of enzymes were lower in GNDV group than those in either GABA group or Na-DMPS group. However, there were no difference in those serum enzymes between GABA group and Na-DMPS group. Moreover, the intoxicated rats treated with combination treatment had the slightest pathological changes in liver and heart (GNDV < GABA or Na-DMPS). Conclusions The co-administration of γ-aminobutyric acid, sodium demercaptopropane sulfonate and vitamin B6 in large doses for the treatment of tetramine intoxication is a method of choice.
