1.Research progress in notch signaling pathway in breast cancer stem cells

Breast cancer stem cells (BCSC) are group of cells exhibiting self-renewal and multi-directional differentiation potentials. These cells have an important role in the occurrence, development, metastasis, and recurrence of breast cancer. In normal circumstances, the ability of mammary stem cells to differentiate and undergo self-renewal is governed by related signaling pathways. After this mechanism is destroyed, breast stem cells undergo abnormal differentiation, forming breast cancer stem cells that unlimitedly proliferate to develop into breast cancer. As research on BCSC increasingly deepens, regulation of BCSC by notch signaling and its crosstalk with several signaling pathways have drawn a great deal of attention in this field. This paper reports the signaling pathways of breast cancer stem cells and latest studies on this field to better understand the essential role of notch signaling pathway in the occurrence and development of breast cancer and corresponding clinical targeted therapy.
2.Different Expression of CD_(21) on Peripheral Blood B Lymphocytes in Children in Different Age Periods

Objective To investigate the clinical significance of the expression of CD_(21) on peripheral blood B lymphocytes in different age periods.Methods All the cases was divided into newborn group,infant group and children group.The expression of CD_(21) on B lymphocytes were analyzed by the flow cytometry.Results 1.The number of CD_(21) on B lymphocytes of newborn group were obviously lo-(wer) than those in other two groups;and with growth of a child,the numer tended to grow up.2.Mean fluorescence intension(MFI),the expression of CD_(21) on B lymphocyte,also had the characteristic of increase with the age.Conclusions With the growing up of a child,the expression of CD_(21) on B lymphocyte has the tendency of up-regulation;which conforms to the feature of immune system development in childhood.Because of the higher expression of CD_(21) on B lymphocyte in certain age,it has the sensitivity to Epstein-Barr virus infection for children.

4.Relationship between expression of PGP9 .5 and clinicopathological fea-tures of cervical carcinoma, and in vitro study of underlying mechanism

AIM: To detect the protein expression of protein gene product (PGP9.5) in cervical carcinoma samples, and to explore its relationship with clinicopathological features and prognosis of cervical carcinoma patients.The potential value of PGP9.5-siRNA in the treatment of cervical cancer was preliminarily investigated in vitro.METHODS:The clinical data of cervical cancer patients ( n=180 ) who received surgical treatment in Department of Gynecology of Tianjin Fifth Central Hospital and Tianjin Central Hospital of Gynecology and Obstetrics from January 2008 to June 2015 were retrospectively analyzed.Immunohistochemical staining for PGP9.5 in all pathological specimens from cervical cancer biopsy was performed.The patients were divided into high expression of PGP9.5 group and low expression of PGP9.5 group.The relationship between PGP9.5 expression and clinicopathological parameters, such as age, HPV infection, path-ological grade, tumor diameter, lymph node metastasis, depth of invasion and clinical stage, were analyzed.The overall survival was analyzed by Kaplan-Meier method and the log-rank test.The PGP9.5-siRNA, NC-siRNA, PGP9.5 eukaryotic expression plasmid and empty vector were transfected into the SiHa cells, and the effects of PGP9.5 expression on the abili-
ties of colony formation and cell invasion were determined by Western blot, colony formation assay and Transwell experi-ment.RESULTS:The relationships between the expression level of PGP9.5 and patients’ pathological characteristics in-cluding grade, tumor size, lymph node metastasis, depth of invasion, vascular involvement and clinical stage were statisti-cally significant ( P<0.05) .The overall survival rates of 3 and 5 years in PGP9.5 high expression group were significantly lower than those in PGP9.5 low expression group ( P<0.05 ) .Compared with control group, the protein expression of PGP9.5, the number of colony formation and the number of invasive cells in si-PGP9.5 group were significantly decreased. The protein expression of PGP9.5, the number of colony formation and the number of invasive cells in PGP9.5 group were significantly increased.CONCLUSION:Over-expression of PGP9.5 protein indicates poor prognosis of cervical cancer, which may be a good predictor for the prognosis of cervical cancer patients.Inhibition of PGP9.5 expression may be an ef-fective way of gene therapy for cervical cancer.
5.The evaluation of rosco disk diffusion on fluconazole susceptibility test of yeast-like fungi

Objective The practical evaluation of Rosco Disk Diffusion method on the clinical antifungal susceptibility test for yeast-like fungi. Methods The Fluconazole susceptibility test was detected by Rosco Disk Diffusion method and NCCLS M27-A broth macrodilution method with 76 yeast-like fungus strains isolated from clinical specimens. Three standard strains were used as the quality control. Results Coincidence rate of the two methods was 90.8%,The sensitive strains detected by one method didn't show resistance in another detection method, and the resistant strains detected by one method yet didn't show sensivity in another.Conclusion Rosco disk diffusion method can be used in clinical detection instead of NCCLS M27-A broth macrodilution method.
6.THE CHEMORADIOTHERAPY OF HIGH RISK VALVAR CARCINOMA

4cm) or involved the urethra、 vagina、 clitoris or anus. Pathologic grouping was 25 squamous carcinomas 3 adenocarcinomas and 2 undifferentiated carcinomas. Radiotherapy was given by angled radiation with wedge block. Dose was 65 to 70 Gy/6-7W. RESULTS Thirty patients completed their inductive chemotherapy within 30 days for 86 courses. The response rate was 86. 7% (26/30). CR was 30. 16%. 18 of 30 patients were followed-up for over 5 years. Five years survival rate was 72.2 (13/18). Major acute complications were vuvitis (23%). Late adverse effects were 1 patient with ure-throstenotic uroschesis, 1 patient with pathologic transcervical fracture. None of patients had radiation rectitis and cystitis. CONCLUSION Inductive chemotherapy consisting of modified VBP regimen was active in reducing the vulvar tumor volume. Radiotherapy used angled radiation with wedged block to treat advance vulvar cancer was fa-ciliated. Chemoradiotherapy was found to be an effective treatment for locally advanced vulvar carcinoma, with acceptable morbidity even in an elderly population. The technique allowed maintenance of normal anatomical and physiologic function that would have been compromised by primary surgical approach. It yielded survival rates comparable to those achieved with ultraradical surgery in younger patients. Life quality was improved significant-ly.
7.Survey of Relationship between Symptoms and Dietary Factors in 54 Outpatients with Irritable Bowel Syndrome

Background:Irritable bowel syndrome(IBS)is a commonly seen functional intestinal disorder. Dietary factors are important factors affecting the abdominal symptoms of IBS patients. Aims:To investigate the effect of dietary factors on abdominal symptoms of IBS and the application of dietary adjustment strategy in patients lived in Xian,Shaanxi Province and the surrounding areas. Methods:Fifty-four outpatients with abdominal complaints who met the Rome Ⅲ criteria of IBS from Feb. 2015 to Jul. 2015 at the Second Affiliated Hospital of Xian Jiaotong University were enrolled. Twenty healthy volunteers were served as controls. All subjects completed a questionnaire containing issues mainly on IBS symptoms, eating habits,dietary factors causing abdominal symptoms,and the patients’coping styles. Results:Compared with healthy controls,more rice and less spicy food,fatty food,raw/ cold food,acidic beverages and alcohol drinking were observed in diet of IBS patients(P all < 0. 05). Abdominal symptoms of IBS patients could be caused or exacerbated predominantly by cold food(55. 6% ),fatty food(55. 6% ),raw food(48. 1% ),meat(48. 1% )and spicy food (40. 7% ). More than half of the IBS patients(63. 0% ) adopted an elimination diet to minimize their abdominal symptoms. Spicy food(59. 3% ),raw food(48. 1% ),cold food(44. 4% ),fatty food(44. 4% )and meat(37. 0% ) were the most frequently eliminated classes of food. Conclusions:Dietary factors can affect the abdominal symptoms of IBS in patients lived in Xian,Shaanxi Province and the surrounding areas and elimination diet based on foods they perceive as problematic is widely used. The classes of food and eating habits causing abdominal symptoms are quite different between domestic and overseas IBS patients.
8.Construction of mcpr1gene vector and expression of mcpr1 in escherichia coli

Objective: To construct mcpr1prokaryoti c expression vector and to express MCPR1 protein.Methods:PCR was used to obtain coding region of mcpr1. Construction of a high-level fusion protein expression vector pGEX-4T-mcpr1 was conducted by inserting the fra gment of coding region of mcpr1into a fusion protein expression vector pGEX -4T-1. Then the recombinant plasmid was transferred into E. colito prepar e the MCPR1/GST fusion protein. DNA sequencing and endonucleases digesting were used to check the coding region. Results:pGEX-4T- mcpr1 wa s constructed successfully and the coding region was inserted into the vector co rrectly. A new protein band of 36 000 was observed by SDS-PAGE analysis after i nduction by IPTG. The 36 000 protein amounted to 39 percent of the total prote in and existed mostly in precipitation of broken bacteria. Conclusion: MCPR1 protein can be expressed in E. coliexpression system and purif ied initially.
9.Refractive change of humans at sitting and supine position

Objective To investigate the spherical diopter and astigmatism change of humans at sitting and supine position.De- sign Prospective case series.Participants 96 eyes of 52 patients (spherical diopter from-2.50 D to-10.00 D,astigmatism diopter from -0.75 D to-4.50 D) were selected.Methods The subjects were examined with NIKON portable retinomax at sitting and supine posi- tion,respectively.Main Outcome Measures The spherical diopter,cylinder diopter and axis change were analyzed statistically.Re- sults Spherical diopter at supine position (-5.31?3.43 D) was a little higher than that at sitting position (-5.27?3.24 D) statistically(P= 0.25),and cylinder diopter at sitting position (-2.27?1.24 D) and at supine position (-2.35?1.19 D) was no statistically difference (P= 0.20).The axis of astigmatism changed from-16?to +18?.Axis change was within 2?in 52.1% eyes,6?-10?in 5.2%,over 10?in 3.1%. The change of axis rotation tended to counter-clockwise in the right eye and clockwise in the left eye.Conclusions Eye rotation at sit- ting and supine position may cause the astigmatism axis change.It may be one of the main factors affecting the results of LASIK.
