1.Detection of the Siberian Tick-borne Encephalitis Virus in the Xinjiang Uygur Autonomous Region, northwestern China.

Until the recent emergence/re-emergence of human-pathogenic viruses in ticks, tick-borne viruses have been neglected as causative agents of human disease (particularly in China). To gain insight into the diversity of tick-borne viruses in Xinjiang Uygur Autonomous Region (northwestern China), we conducted illumina deep sequencing-based screening for virus-derived small RNAs in field-collected Ixodes persulcatus ticks. We found 32, 631 unique virus-matched reads. In particular, 77 reads mapped to the tick-borne group within the genus of Flavivirus, and covered 3.8%-2.4% viral genomes. In addition, 32 unique reads were specific to the Siberian subtype of tick-borne encephalitis viruses (TBEV-Sib) which have never been reported in Chinese TBE loci. We confirmed the potential existence of TBEV-Sib by amplification (using reverse transcription-polymerase chain reaction) of genomic fragments from the envelope gene or 3' genomic terminus from the pools of examined ticks. Both sequences demonstrated high homology to TBEV-Sib strains attached geographically to southern Siberia with nucleotide identity of 97.2%-95.5% and aminoacid identity of 99.4%-98.3%, respectively. In conclusion, we report, for the first time, detection of TBEV-Sib in the natural TBE loci of China. These novel data may provide genetic information for further isolation and epidemiologic investigation of TBEV-Sib.

3.Isolation of tick-borne encephalitis virus Far-eastern subtype and Siberian subtype in the China-Kazakhstan border area in Xinjiang

To investigate the genotype and the biological characteristics of Tick-borne encephalitis virus (TBEV) in Charles Hilary endemic foci in the China-Kazakhstan border area in Xinjiang,ticks were collected by flagging during May to June in 2012 and 2014,and were stored in liquid nitrogen.TBEV strains were isolated from tick samples by inoculating BALB/c mice and BHK-21 cells.The FE gene fragments of TBEV-Far and the S gene fragments of TBEV-Sib were detected by RT-PCR from infected mice brain tissue and BHK cells,and then subjected to sequence alignment.Totally 16 TBEV strains were isolated from Ixodes persulcatus and Dermuceuter silvarum,among 13 strains were Far eastern subtype,three strains were Siberian subtype.It was first time that the TBEV-Sib was isolated in China.The Charles Hilary TBE natural foci were in the China-Kazakhstan border area,and both TBEV-Far and TBEV-Sib co-circulated.
4.Determination of cocaine and benzoylecgonine in guinea pig's hair after a single dose administration by LC-MS/MS.

A sensitive LC-MS/MS method to determine cocaine and its major metabolite benzoylecgonine in guinea pig' s hair has been established. About 20 mg of decontaminated hair sample was hydrolyzed with 0. 1 mol x L(-1) HCl at 50 degrees C overnight, in the presence of cocaine-d3 and benzoylecgonine-d8 used as internal standards, and then extracted with dichlormethane. The analysis was performed by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Positive electrospray ionization (ESI +) and multiple reactions monitoring (MRM) mode were used. The limit of detection (LOD) for cocaine and benzoylecgonine was 1 pg x mg(-1). The calibration curves of extracted standards were linear over the range from 5 pg x mg(-1) to 250 pg x mg(-1) (r2 > or = 0.9997). The method was validated and applied to the analysis of guinea pig's hair after a single dose administration of cocaine hydrochloride. Cocaine and benzoylecgonine were not only detected, but also quantified in guinea pigs hair.

5.Comparison of clozapine in nail and hair of psychiatric patients determined with LC-MS/MS.

As a keratinized material, nail recently has attracting researchers' attention in the pharmaceuticals analysis. There are comparatively limited studies concerning nail's xenobiotic determination and its mechanism. This article reported the development of a sensitive, specific and reproducible LC-MS/MS method, which could be as a foundation of other studies on drug determination in nail. It can also be regarded as the first report on organic drug in mainland China. Sixteen nail samples from volunteers, who were ingested clozapine for more than nine months, are confirmed positive after being analyzed by the method. It is found that contents of clozapine in the patients' nails are above the nanogram level. Besides, a comparative study of clozapine concentration in nails and hair was made, with a result that there exists a correlation between the two materials in terms of clozapine concentration.

6.Construction of antisense telomerase hTERT and its effect on K562 cells.

OBJECTIVESTo investigate whether antisense human telomerase reverse transcriptase (hTERT) could inhibit the activity of telomerase and the proliferation of K562 cells.
METHODSThe antisense plasmid was constructed by reverse insertion of hTERT PCR product into plasmid pLNCX-neo. Then the constructed plasmid was introduced into K562 cells by liposomes-mediated DNA transfection. The inhibition effects of telomerase on the proliferation of K562 cells were analyzed by MTT and colony formation assay, the telomerase activity of K562 cells by TRAP-PCR ELISA methods.
RESULTSThe growth rate of antisense hTERT transfected K562 cells was significantly lower than those of the controls, and the colony formation capacity of the transfected cells decreased significantly (P < 0.01), the colony number is (100.33 +/- 7.57)/10(3) cells, (92.67 +/- 5.86)/10(3) cells and (50.33 +/- 6.11)/10(3) cells for control K562 cells, K562 neo cells and antisense hTERT transfected HL60 cells, respectively. The telomerase activity of antisense hTERT transfected K562 cells was significantly inhibited.
CONCLUSIONThe expression of an antisense sequence to the mRNA sequence of telomerase protein subunit can inhibit the activity of telomerase, slow the cell growth and inhibit the capacity of colony formation of K562 cells.

7.Identification of Culex Flavivirus by deep sequencing approach in Xinjiang, China

Objective To investigate the diversity of mosquito-borne viruses in Xinjiang , China, and to identify mosquitos-borne viruses of medical importance rapidly .Methods The virus-derived RNAs in mosquitos captured in wild were screened and confirmed by using Illumina deep sequencing approach and reverse transcription PCR , respectively .The alignment analysis was performed by using gene sequences from GenBank.Results One hundred and forty-four Culex Flavivirus ( CxFV, Flavivirus genus, Flaviviridae) specific sequences were identified .The overlapping reads were assembled into 7 uncontinuous viral genomic contigs.The gaps between the contigs were further filled by RT-PCR products, which resulted in reconstruc-tion of viral genomic 5′and 3′terminus (687 nt and 411 nt).Phylogenetic analysis showed that the newly identified CxFV belonged to America/Asian genotype , which specifically clustered into a clade with other CxFV strains from China mainland ,sharing 98.2%-99.5%homologies in nucleotide sequences and 99.5%in amino acids sequences among them .Conclusion Illumina deep sequencing approach was successfully applied to arthropod-borne virus surveillance .The recently emerged Culex Flavivirus was detected for the first time in Xinjiang, China.
8.Simultaneous analyses of cocaine and its metabolite benzoylecgonine in urine by LC-MS/MS.

OBJECTIVE:
To establish a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the simultaneous analysis of cocaine (COC) and its metabolite benzoylecgonine(BZE) in urine samples.
METHODS:
A single dose of COC (10 mg/kg) was injected intraperitoneally into guinea pigs and urine samples were collected for 7 days. The urine samples were extracted by auto solid phase extraction (SPE), separated by the Allure PFP propyl column with a mobile phase consisting of methanol and 20 mmol/L ammonium acetate buffer [0.1% formic acid (80:20, V/V)], and then analyzed by LC-MS/MS. Multiple reaction monitoring (MRM) mode was used to analyze COC (m/z 304.2-->182.3, m/z 304.2-->150.1) and BZE (m/z 290.2-->168.3, m/z 290.2-->105.0).
RESULTS:
COC and BZE showed a fairly good linearity over the range of 2.0-100 ng/mL (r=0.9995). The detection limit was 0.5 ng/mL. The recovery rate was greater than 90% and the deviation of intra- and inter-day precision was less than 6%. BZE was the major target detected in urine samples, and its detection window was longer than COC.
CONCLUSION
This newly developed method shows high sensitivity and selectivity, and is suitable for the simultaneous analysis of cocaine and benzoylecgonine in urine samples.

9.Prevalence of Anaplasma spp. infection in a desert landscape region of Heshuo, Xinjiang

Objective To study the prevalence ot Anaptasmosts among human,domestic sheep and tick population in Heshuo area,southern Xinjiang and to investigate the diversity of Anaplasma species.Methods Ticks were captured from wild field and blood samples were collected from healthy residents and their domestic sheep.Indirect fluorescent assay was carried out to determine the presence of Anaplasma specific IgG antibodies in blood sample of human and goats,respectively.Nested PCR and sequence alignment of Anaplasma partial 16S rDNA were used to investigate the diversity of Anaplasma species.Results 43.31% (55/127) of human beings and 27.50% (55/200) of the goats were found positive forAnaplasma specific IgG.In total,367 ticks were captured,including 3 genus and 4 species,which mainly consisting of dominate Hyalomma (H.)asiaticum (47.41%) and Rhipicephalus (R.)pumilio (37.60%).5.00% (18/360) of the questing ticks and 4.49% (7/156) of the goat blood samples were found to have had 16S rDNA,representing Anaplasma sp.by nested PCR,but none was found from human beings.Results from sequencial alignment revealed that the positive amplicons were identified to be Anaplasma phagocytophilum (99.2% GU046565,99.5% GU064897 and 99.5% AB196721)and Anaplasma central (99.2%GU064903).Conclusion Human and zoonotic anaplasmosis,which were probably transmitted by H.asiaticum and R.pumilio,were co-circulating in the desert landscape of Heshuo area,Xinjiang.
