1.Microleakage of Resin- Modified Glass Ionomer Cement and Compomer Restorations in vitro

ObjectiveTo evaluate the marginal integrity of restoration in two types of mate- rials, or resin- modified glass ionomer cement and polyacid- modified composite resin (compomer). MethodsRestorations of four kinds of material (GC Fuji Ⅱ LC, VitremerTM, Dyract compomer and F2000 compomer) were placed in the facial Class V cavity preparations in forty noncarious human molar teeth. The teeth were randomly assigned to 4 experimental groups of 10 teeth each. After thermal cy- cling( × 20,5 ~55℃ ), the teeth were immersed in 1% basic fuchsin dye for 24 hours at room tempera- ture. Staining along the teeth restoration interface was recorded. ResultsThe data indicated signifi- cant differences between all the restorative materials for both occlusal and gingival scores ( P=0.026, P = 0. 000)respectively. Further analysis revealed there were statistically significant differences between GC Fuji Ⅱ LC and others on occlusal margins versus Dyract compomer and others on gingival margins. Conclusion Factors such as dental conditioning, rates of water absorption and thermal coefficient were related to microleakage. Resin - modified glass ionomer cement showed less microleakage than polyacid- modified composite resin tested.
2.Biological Characteristics of the Rhizobia Isolated from Acacia confusa Grew in the Karst Environment

The paper is about the systematic studies of biological characteristics of 15 stains rhizobia isolated purified from Acacia confusa grew in Guangxi karst environment.The results showed that there were typical characteristics of rhizobia.there were negative reaction about use of 3-ketolactose and beef extract peptone nutrient agar medium,and positive reaction about use of starch and citrate medium,and produce acid in reaction of BTB and litmus milk medium,(NH4)2HPO4 was used as nitrogen sources and both four monosaccharides and three disaccharides could be utilized as carbon sources in 15 strains rhizobia isolated Acacia confuse.Among the 15 strains for the tests,11 strains could deoxidize the nitrate of medium into nitrite,14 strains could grow well on NaCl solution concentration 3.0 %~4.0 %,14 strains could grow at 39℃,13 strains may grow on highest pH12 and 4 strains on lowest pH4 cultrue medium.15 strains can grow in 10% and 11 strains in 10%~30% of CaCO3 solution concentration.
3.Experimental study on the ileum tissue proteins differentially expressed in Pi-qi deficiency syndrome rats.

OBJECTIVETo identify the ileum tissue proteins differentially expressed in Pi-qi deficiency syndrome rats using proteomic approach.
METHODSThirty-seven rats were randomly divided into 3 groups, i. e., the normal control group (Group 1), the Pi-qi deficiency syndrome model group (Group 2), and the reserpine group (Group 3). The Pi-qi deficiency syndrome model was established using excessive exerting combined with irregular diet, and peritoneally injecting Reserpine Injection (1 mg/mL) respectively. The ileum tissues were separated after identified fuzzy method. The differentially expressed proteins were separated with two dimensional electrophoresis (2-DE), analyzed by Mass Spectrometry, and identified by MASCOT Software.
RESULTSThree proteins were differentially expressed in Group 2 and nine proteins were differentially expressed in Group 3 (P < 0.05).
CONCLUSIONPi-qi deficiency syndrome was closely related with decreased expression of albumin as well as increased expressions of trypsin and glucose-regulated protein 78.

5.Effect of three TCM methods for activating blood circulation on early stage apoptosis in rats with chronic atrophic gastritis complicated precancerous lesion.

OBJECTIVETo comparatively study the effects of three TCM methods for activating blood circulation, i.e. in combined with resolving stasis (A), regulating qi (B) and supplementing qi (C), respectively, on early stage cell apoptosis in precancerous lesion of rats with chronic atrophic gastritis (CAG).
METHODSRat model of CAG with precancerous lesion was duplicated by insertion of spring in pylorus and gastric perfusion of high-salt hot paste; and the impact of treatment on cell apoptosis was determined using Annexin V/PI double labeled flow cytometry.
RESULTSAfter being intervened for 12 weeks, the early stage cell apoptosis rate in the natural recovery group was significantly higher than that in the control group (P <0.01); while it lowered more significantly in the three groups receiving TCM therapeutic methods for activating blood circulation, showing significant difference compared with the natural recovery group (P <0.01).
CONCLUSIONThree therapies of activating blood circulation all show inhibitory action on the early stage apoptosis of precancerous lesion in CAG rats, which is possibly one of their action mechanisms for improving or reversing the precancerous lesion.

6.Expression of GBD gene of Streptococcus mutans glucan binding protein A in mammalian cells.

OBJECTIVETo evaluate the expression of recombinant plasmid pcDNA3.1/GBD of glucan binding protein of Streptococcus mutans in mammalian cells COS-7.
METHODSEukaryotic plasmid carrying encoding gene of GBD of Streptococcus mutans gbpA was constructed and the plasmid was introduced into COS-7 cells by Lipofectamine reagent. The transient expressed protein in COS-7 cells was detected by immunochemistry technique.
RESULTSThe positive expression was detected in plasma of the cells which were transfected with recombinant plasmid pcDNA3.1/GBD. The cells which were transfected with pcDNA3.1 were negative.
CONCLUSIONGBD can translate and express in COS-7 cells after transfected with recombinant plasmid pcDNA3.1/GBD. The expressed protein locates in the plasma and the protein is able to combine with anti-GbpA antibody. The expressed protein has the antigenicity and is a candidate gene vaccine.

7.In vitro expression of hemophilia B gene mediated by lentivirus.

OBJECTIVETo construct a three plasmids lentiviral vector containing canine coagulation factor IX (cFIX) gene with ubiquinone promoter (PUB) and observe the expression of cFIX gene.
METHODSLentivirus was generated by transient three-plasmid transfection, namely, the VSV-G envelope expression cassette, the delta NRF packaging plasmid and the PTK 164 plasmid. Viral particles were used to infect the target cell, third passage mesenchymal stem cells (MSCs) and 293T cell respectively at MOI 3: 1. The cFIX activity was detected in cultured cells with one-stage clotting assay.
RESULTSThe MSCs were obtained in vitro. The lentivirus infected MSCs and 293T cells all expressed the active factor IX with the activity of (26.30 +/- 2.10)% and (19.70 +/- 1.53)%, respectively, which are significantly higher than that of control (1.00 +/- 0.05)%.
CONCLUSIONSThe lentiviral vector of three plasmids with ubiquinone promoter (PUB) was constructed and can transfect the MSCs and 293T cells.

8.Characterization of Notch gene involved in genetic regulatory networks in dental pulp stem cells.

OBJECTIVETo investigate the characterization of Notch gene involved in genetic regulatory networks in dental pulp stem cells.
METHODSThe pulp tissue was separated from mouse teeth and digested by collagenase type I. Single-cell suspensions of dental pulp were seeded into 6-well plates with alpha modification of Eagle's medium supplemented with ES cell qualified Fetal Bovine Serum. Colony-forming efficiency was assessed in 14ds culture. Transcripts for Notch were detected by reverse transcription-PCR by using total RNA isolated from cells.
RESULTSThere were clonogenic cells in dental pulp cell and the incidence of colony-forming cells derived from mouse dental pulp cells was 1.6-2.5 colonies/10(4) plate. Mouse-specific Notch mRNA expressed in colony-forming cells.
CONCLUSIONNotch mRNA expressing in colony-forming cells provided a more detailed understanding of mouse dental pulp stem cell biology.

9.Re-growth of induced-osseous tissue encapsulated in muscular fasciae for prefabrication of an osteo-musculo-cutaneous flap.

OBJECTIVETo improve the survival of mesenchymal stem cells (MSCs) for prefabrication of an osteo-musculo-cutaneous flap.
METHODSIn a mouse model, the compound of MSCs and p(3HB-co-3HH) were embeded in the latissimus dorsi muscle as an experimental group and the muscle pocket of the buttock as the control. The examinations of the HE staining, hybridization in situ of osteonectin mRNA and Von kossa staining were used to evaluate the results.
RESULTSThe expression of osteonectin mRNA and the Von kossa staining showed that the latissimus dorsi muscle group was superior to the control in 2 weeks and 4 weeks after the surgery in vivo.
CONCLUSIONThe results indicate that the above-mentioned technique may be a good alternative for the prefabrication of the osteo-musculo-cutaneous flap.

10.Comparative proteomics analysis of the ileum of Pi-Yang deficiency model rats.

OBJECTIVEBased on proteomics technology, Pi-yang deficiency syndrome (PYDS) correlated differential proteins were screened, thus providing powerful experiment reliance for exploring the essence of PYDS.
METHODSTotally 36 SD rats of SPF grade were randomly divided into the normal control group (n = 16) and the PYDS group (n = 20). The PYDS model rats were induced by improper diet, overstrain, and administration of yang impairing bitter cold herbs. The total proteins of the ileum were separated and extracted from rats in the PYDS group and the normal control group. The differential protein dots were identified using Delyder 2D 6.5 image analysis software by two-dimensional gel electrophoresis (2-DE) technology. The finger print map of corresponding peptide qualities was obtained by applying MALDI TOF/TOF. The differential proteins were identified using Mascot search library.
RESULTSJudged by statistics and fuzzy mathematics, Pi-yang deficiency rat model was successfully established. Eight proteins with differential expressions involving cell skeleton, energy metabolism, and signal transduction, and so on were obtained. Of them, there were 4 up-regulated proteins, i.e., desmin, cytokeratin8 (CK8), pyruvate kinase (PK), and ezrin. Four down-regulated proteins were glyceraldehyde-3-phosphate dehydrogenase (GAPDH), cytokeratin19 (CK19), cytokeratin1 (CK1), and actin.
CONCLUSIONThe pathogenesis of PYDS might be slowed energy metabolism rate, reduced energy production, changed structure of ileal villin, and weakened absorbing and digestive functions.
