Objective To design Wound Area Measurement System based on the digital image processing.Methods A reference object whose area is known to the wound is set and the digital image processing is utilized to measure the wound area.Results The system was used in the experiment of animal model,and the anticipated results were achieved.Conclusion The system is fit for measuring the wound area in the studies of wound repair.
2.Overexpression of Penicillium expansum lipase gene in Pichia pastoris.

The alkaline lipase gene of Penicillium expansum (PEL) was coloned into the yeast integrative plasmid pPIC3.5K, which was then transformed into His4 mutant yeast GS115. Recombinant Pichia strains were obtained by minimal olive oil-methanol plates screening and confirmed by PCR. The expression producus of PEL gene was analysis by SDS-PAGE and olive oil plate, the result indicated that PEL gene was functionally overexpressed in Pichia pastoris and up to 95% of the secreted protein. Recombinant lipase had a molecular mass of 28kD, showing a range similar to that of PEL, could hydrolyze olive oil and formed clear halos in the olive oil plates. Four different strategies (different media, pH, glycerol and methanol concentration) were applied to optimize the cultivation conditions, the activity of lipase was up to 260 u/mL under the optimal cultivation conditions. It is pointed out that the absence of the expensive biotin and yeast nitrogen base in the medium increased the lipase production. The possible reason of this result is absence of yeast nitrogen base increased the medium pH during cultivation, and PEL shows a higher stability at this condition. The lipase activity of the supernatant from the culture grown at pH 7 was higher than the one from the culture in the same medium at pH 6.0 is due to the pH stability of PEL too. The results also showed that the methanol and glycerol concentration had a marked effect on the production of lipase.

3.Protective effect and nursing intervention of magnesium magnesium oxalate on liver function after hepatectomy

Objective To study the protective effect of nursing intervention and magnesium oxalate on liver function after hepatectomy. Methods 64 cases of liver resection patients in our hospital were randomly divided into the control group and the experimental group, with 32 patients in each group. In the control group, patients were given 150mg magnesium magnesium sulfate for 7 days after the operation. On this basis treatment of the control group, the experimental group was carried out targeted nursing intervention, payed attention to the patient's psychological state, patiently explain to the patients, did well preoperative preparation, and closely observed the correlation of liver function. Clinical indicators of the two groups of patients were compared and analyzed. Results 7 days after operation, the level of ALT in the experimental group was (50.20±23.21) U/L, and the level of AST was (25.40±9.03) U/L. The level of ALT in the control group was (95.43±43.90) U/L, and the level of AST was (38.33 ± 9.19) U/L, there were statistical significance in two groups (P<0.05). There were no obvious adverse reactions in two groups, and there was no statistical significance (P<0.05). The level of serum ALT in the experimental group decreased to normal range, and the number of patients was significantly higher than that of the control group with statistical significance (P<0.05). Conclusion The effect of Magnesium Oxalate and nursing intervention on liver function after hepatectomy is ideal. It can significantly improve the liver function of patients with high safety and clinical significance.
4.Effect of 1, 2-dichloroethane on blood brain barrier.

OBJECTIVETo investigate the effect of 1, 2-dichloroethane (1, 2-DCE) on blood brain barrier.
METHODSAcute toxic encephalopathy model was copied with the consecutive static inhalation of 1, 2-DCE. The water content of brain tissue was measured, and the blood brain barrier permeability was detected with lanthanum nitrate. The brain microvascular endothelial cells and neuroglial cells were cultured in vitro, which were administrated with 1, 2-DCE. The cell morphologic structures were observed under light microscope and electron microscope.
RESULTS(1) The extracellular edema was most found in the cerebral tissue and the leakage of lanthanum particles through the barrier were found with the lanthanum tracking method. (2) The water content in cerebral cortex in the moderate and high dose groups was significantly higher than that in the control group and became severer with the increases of the intoxicated time. The water content in cerebral medulla was significantly increased only at 6 hours after the intoxication. (3) The normal morphological structure of brain microvascular endothelial cells and neuroglial cells could be injured by 1, 2-DCE, and the injury to neuroglial cells caused by 1, 2-DCE occurred earlier and severer than that to brain microvascular endothelial cells.
CONCLUSION1, 2-DCE can damage blood brain barrier and induce cerebral edema.

5.Correlation between compassion fatigue and workplace violence in emergency department nurses

Objective:To investigate the workplace violence and compassion fatigue of nurses in emergency department and to explore the relationship between the two.Methods:The general information questionnaire, workplace violence scale and professional quality of life scale were used to investigate 957 emergency department nurses of 28 Level II hospitals and above.Results:The scores of each dimension of the professional quality of life scale for nurses in the emergency department were: compassion satisfaction score was 29.91±7.82, the burnout score was 26.63±5.66, and the second trauma score was 23.17±5.94. The total score of compassion fatigue is 49.80±10.42. The incidence of workplace violence was 77.6%. Workplace violence was negatively correlated with compassion satisfaction ( r=-0.250, P<0.01) , and positively correlated with burnout, secondary trauma, and total compassion fatigue ( r=0.349、0.340、0.384, P<0.01) . Whether there is only non-physical violence in the compassion satisfaction, burnout, secondary trauma, compassion fatigue total score is not statistically significant. Conclusion:Compassion fatigue is more serious in emergency department nurses, and the incidence of workplace violence is higher. Workplace violence has a positive effect on compassion fatigue. Nursing managers should actively prevent workplace violence and improve the working environment, thus reducing empathy fatigue.

Objective To evaluate the use of gated SPECT in patients with hypertrophic obstructive cardiomyopathy (HOCM) and the effects of percutaneous transluminal septal myocardial ablation (PTSMA) on myocardial perfusion.Methods 99 Tcm-methoxyisobutylisonitrile (MIBI) and 18F-fluorodeoxyglucose (FDG) images were performed in 31 HOCM patients before PFSMA and in 15 patients 3-7 d after PTSMA.The images in different left ventricular segments were analysed by using scores.Results In 99Tcm-MIBI images, uptake decreased at the septal regions in 12 HOCM patients (80.0%, 12/15) after PTSMA, 18F-FDG images also showed decreased uptake at the septal regions in 5 HOCM patients (33.3%, 5/15) after PTSMA.Conclusion 99Tcm-MIBI images might be an important method to evaluate PTSMA results, and 18 F-FDG images showed important value as reference.
8.An analysis on HIV rapid test demanding among commercial sex workers

Objective To learn the awareness and related factors of HIV rapid test among commercial sex workers(CSWs)in order to improve the HIV rapid test service. Methods The cross-sectional survey was conducted among CSWs in WenzhouCity and Quzhou City which were divided into high-grade, middle-grade and low-grade. Face to face questionnaire surveywas conducted in the research investigation. The investigation content includes social demographic characteristics, rapiddetection of desire, sexual behavior and HIV testing services. EpiData 3.0 database was used for data entry. Statisticalsoftware SPSS21.0 was used in chi-square and logistic regression analysis of multiple factors. Results A total of 401 CSWswere observed, including 136 people in high-grade, 182 people in middle-grade and 83 people in low-grade. Twohundred and two people and 200 people were investigated respectively in Wenzhou and Quzhou City. The rate of willing toaccept the rapid HIV testing was 56.36%(226) . Analysis by logistic analysis, 30 years old of above(OR=3.312, 95%CI:1.427-7.691), commercial sexual behavior without using condom or casual usage in last 6 month(OR=4.560, 95%CI:2.348-8.855), more than 10 commercial sexual partners(OR=8.767, 95%CI: 2.993-25.677), and having HIV rapidtesting in last year(OR=7.784, 95%CI: 3.108-19.496) were influencing factors for willing to accept HIV rapid testing.Conclusion In CSW HIV/AIDS intervention, adult, few numbers of sexual partners need to be focused. Improving HIVrapid test service, condom using service, and promoting risk sexual awareness may improve the HIV rapid test acceptancewillingness and behavior.
9.Effect of seedling and fertilizering on yield and quality of cultivated Changium smyrnioides.

OBJECTIVETo study the effects of seedling and fertilizer on yield and quality of cultivated Changium smyrnioides and build a basis for its GAP.
METHODExperiments were conducted to study the effects of seedling quality and different fertilizering measures on yield and quality of cultivated C. smyrnioides.
RESULTThe seedling and fertilizering depth affected the yield of roots obviously. Organic fertilizer combined with nitrogen and phosphorus could increase the amylose content obviously.
CONCLUSIONIn cultivation, the seedling (root length > 5.0 cm, root diameter > 0.3 cm, fresh root weight > 0.3 g) is optimal. The ideal combination is nitrogen, phosphorus and organic fertilizer. The suggested fertilizering depth ranged from 10 to 15 cm.

10.Rapid improvement of lipase production in Penicillium expansum by genome shuffling.

In the present study, the genome shuffling was used to improve lipase production of Penicillium expansum. A lipase producing mutant strain-Penicillium expansum FS8486 and a wild type of Aspergillus Tamarii FS-132 isolated from soil of a volcano in Xinjiang were used as the parental strains. After two rounds of genome shuffling, several elite daughter strains were screened. The lipase activity in one of the daughter strains was increased 317% over the starting strain FS8486. Comparisons of the morphology, RAPD (Random Amplification of Polymorphic DNA) polymorphism and the fatty acid compositions between the daughter and the parental strains suggested that the filial generation were generated by genome shuffling. In this study, the genome shuffling used successfully first time in eukaryotic microorganism and increases the production of the desired metabolite in short time, the study will be useful to spread the genome shuffling in eukaryotic microbial breeding.
