College of Stomatology of Wuhan University was the first dental school who introduced the Kavo head-simulator for clinical teaching.In this study,students of stomatology were investigated before and after head-simulator training and questionares were made.The results have showed that head-simulator teaching system not only raises the activity of student,consolidates the basic knowledge,develops the clinical thinking ability,but also increases the ability of clinical operational skill before clinical training.

Objective To investigate the effects of isoflurane anesthesia on proliferation and differentiation of neural stem cells (NSCs) in the dentate gyrus of neonatal rats.Methods Ten SD rats,aged 7 days,weighing 16-20 g,were randomly divided into 2 groups ( n =5 each):control group (group C) and isoflurane group (group 1) .The rats in group Ⅰ inhaled 2.5％ isoflurane for 3 min for induction and then anesthesia was maintained with 1.5 ％ isoflurane for 4 h,while the rats in group C only breathed the room air for 4 h.Bromodeoxyuridine (BrdU) 100 mg/kg was injected intraperitoneally right before the induction and after the end of administration to label NSCs and their progeny in the dentate gyrus.At 24 h after the 2nd administration of BrdU,double immunofluorescence for BrdU and NeuroD (a marker of neuroblasts and immature neurons) was used to assess NSC proliferation and neuronal differentiation.Results Compared with group C,the number of BrdU+ cells in group Ⅰ was significantly decreased,whereas the fraction of NeuroD+/BrdU+ differentiated cells was increased in the dentate gyrus( P ＜ 0.05 or 0.01 ).Conclusion Isoflurane anesthesia suppresses the proliferation of NSCs and induces neuronal differentiation of NSCs in the dentate gyrus of neonatal rats.

Objectives To investigate the prevalence of left ventricular hypertrophy (LVH) and risk factors in children with chronic kidney disease (CKD). Methods The biochemical indices, blood pressure and left ventricular mass index (LVMI) in pa-tients with CKD were retrospectively analyzed. The risk factors of LVH were analyzed using Logistic regression. Results In 125 CKD patients, 32.00%were at 4th stage and 68.00%were at 5th stage. The estimate glomerular ifltration rate (eGFR) and hemo-globin (Hb) level were signiifcantly higher in CKD patients at 4th stage than in those at 5th stage. The intact parathyroid hormone (iPTH), serum phosphorus and LVMI were signiifcantly lower in CKD patients at 4th stage than in those at 5th stage (P<0.01). LVH was detected in 33.60%CKD patients. The eGFR and Hb level were signiifcantly lower in CKD patients with LVH than in those without LVH. The iPTH, serum phosphorus, systolic blood pressure and diastolic blood pressure were signiifcantly higher in CKD patients with LVH than in those without LVH (P<0.05). Logistic regression analysis indicated that only hypertension, hyperphosphatemia, moderate and severe anemia were the risk factors of LVH. Conclusion Control of hypertension, hyperphos-phatemia and anemia is the key to prevent LVH in CKD patients.

ObjectiveTo seek a convenient method for improving the clinical therapeutic effect on knee osteoarthritis.Method One hundred and eighty-six patients were randomly allocated to three groups, 62 cases each. The observation group received needle-sticking warm needling moxibustion plus articular cavity injection of sodium hyaluronate; control group 1, needle-sticking warm needling moxibustion; control group 2, articular cavity injection of sodium hyaluronate. Needle-sticking warm needling moxibustion was given once daily, five treatments followed by two days of rest, seven days as a course, for a total of five courses. Articular cavity injection of sodium hyaluronate was administered once a week, for a total of five injections. The therapeutic effects were compared after treatment.ResultA large part of the symptoms and signs resolved, and the cure rate, the cure and marked efficacy rate and the efficacy rate were 40.3%, 87.1% and 95.2%, respectively, in the observation group. Part of thesymptoms and signs resolved in control groups 1 and 2. The cure rate, the cure and marked efficacy rate and the efficacy rate were 24.2%,51.6%and 80.6%, respectively, in control groups 1 and 21.0%, 54.8% and 82.3%, respectively, in control groups 2. There were statistically significant differences in the cure rate (P<0.05), the cure and marked efficacy rate (P<0.01) and the efficacy rate (P<0.05) among the three groups. The therapeutic effect was significantly better in the observation group than in control groups 1 and 2.Conclusion Needle-sticking warm needling moxibustion plus articular cavity injection of sodium hyaluronate is clinically a better way to treat knee osteoarthritis.

Objective To investigate early nasogastric nutrition on patients with acute decompensated chronic heart failure,and observe its influence on patient's disease outcome.Methods From January 2012 to December 2013,100 cases with acute decompensated chronic heart failure were used as the research subjects,were randomly divided into experimental and control groups with 50 cases each group.Patients of two groups adopt expansion of blood vessels,strong heart,diuresis,and routine anti-infection treatment;Based on this scheme,experimental group adopt early nasogastric nutrition.The cardiac function index,the change of nutritional status and clinical curative effect and the result of follow-up within 1 year after discharge of two groups were compared.Results Left ventricular end-diastolic dimension (LVEDD) and left ventricular end-systolic dimension (LVESD) in two groups after treatment were decreased,and the descender of experimental group was superior to that of the control group with statistically significant difference (P ＜ 0.05).After treatment,left ventricular ejection fractions (LVEF) of two groups were improved,and the rise of experimental group was superior to that of control group with statistically significant difference (P ＜0.05).In the two groups after treatment,the serum brain natriuretic peptide (BNP) levels were significantly decreased,and the descender of the experimental group was more than that of control group with statistically significant difference (P ＜ 0.05).After treatment,body mass index (BMI),the level of serum prealbumin (PA),transferrin (TRF),and the total number of lymphocytes of two groups were increased significantly (P ＜ 0.05);and BMI,the level of serum PA and TRF,and the total number of lymphocytes of the experimental group were significantly better than those of the control group (P ＜ 0.05).The total effective rate of the experimental group was significantly higher than that of the control group (P ＜ 0.05).The exacerbation rates,readmission and mortality rate of experimental group follow-up within 1 year after discharge were significantly lower than that of the control group (P ＜ 0.05).Conclusions The early nasogastric nutrition on the patients with acute decompensated chronic heart failure was better able to improve patients' nutrition state,more conducive to patients with cardiac function recovery,improve the clinical curative effect,and reduce the exacerbation rates,and readmission and mortality rate.

ObjectiveTo investigate the effects of isoflurane anesthesia on the expression of brain-derived neurotrophic factor (BDNF) and phosphorylated extracellular signal-regulated kinase (p-ERK) in neonatal rat hippocampus.MethodsForty-eight SD rats of both sexes,aged 7 days,weighing 12-17 g,were randomly divided into 2 groups ( n =24 each):control group (group C) and isoflurane anesthesia group (group Ⅰ).In group Ⅰ,the rats were exposed to2.5％ isotlurane for 3 min and then 1.5％ isoflurane was inhaled for 4 h,while in group C the rats were exposed to air for4 h.Arterial blood samples were collected immediately after anesthesia for blood gas analysis and for determination of the blood glucose concentration.Five rats in each group were sacrificed at 0,6,24 and 48 h after anesthesia (T1-4) and hippocanpi were removed for determination of the expression of potassiumchloride cotransporter 2 (KCC2),potassium-chloride cotrmsporter 1 (NKCC1),BDNF and p-ERK by Western blot.NKCC1/KCC2 ratio was calculated.ResultsAcid-base imbalance,hypoxemia and glycopenia were not found immediately after anesthesia in both groups.Compared with group C,KCC2 expression was significantly down-regulated and NKCC1/KCC2 ratio was increased at T3 and T4,and the expression of BDNF and p-ERK was dewn-regnlated at T1 and T2 in group Ⅰ (P＜0.05).There was no significant difference in NKCCI expression at each time point between groups Ⅰ and C ( P ＞ 0.05 )、ConclusionIsoflurane anesthesia delays the neuronal development in neonatal rat hippocampus through down-regulating the expression of BDNF and p-ERK.

Objective To construct the eukaryotic vector that expressing hepatitis B virus (HBV) S and the single fragment of variety chain (ScFv) of monoclonal antiboy against cytotoxic Tlymphocyte-associated antigen-4 (CTLA-4) and to analyze the immunological activity of recombinant S-ScFv protein.Methods The oringially constructed pSect2/ScFv4F10 and pSect2/S were double enzyme digested by Sfi I and Hind Ⅲ,respectively.Then the HBV S gene was cloned into the pSect2/ScFv4F10 vector.The pSect2/ScFv4F10 and pSect2/S-ScFv4F10 were expressed in Chinese hamster ovary (CHO) cells,and the expressed proteins were verified through sodium dodecyl sulfatepolyacrylamide gelelectrophoresis(SDS-PAGE)andWesternblotting.Afterultrafiltration concentration and affinity chromatography,the biological affinity of the expressed ScFv4F10 and S-ScFv4F10 proteins were examined by competitive enzyme-linked immunosorbent assay (ELISA) and surface plasmon resonance (SPR) technology.The comparison between groups was done by One-way ANOVA.ResultsThe eukaryotic expression vector of pSect2/S-ScFv4F10was successfully constructed,and relative molecular mass of the expressed protein of S-ScFv4 F10 was about 52 000 that analyzed by SDS-PAGE and Western blotting.With the fixed concentration of 4F10-mAb against CTLA-4,the A570 value of the mixed reaction with purified CTLA-4 antigen gradually increased with the decrease of ScFc fusion protein proportion; when the molar ratio of ScFv,S-ScFv4F10∶4F10=2∶1,the competitiveinhibitionratesagainst 4F10conjugatedantigenwere72.6％and64.5％,respectively.The affinity constants of association kinetics for CTLA-4 mAb,ScFv4F10 and S-ScFv4F10 with CTLA-4 antigen were 7.29 × 108 mol/L,9.52 × 106 mol/L and 2.04 × 106 mol/L,respectively,and the dissociation constants of KD were 1.40 × 10-9 mol/L,1.05 × 10-7 mol/L and 4.91 × 10-7 mol/L,respectively.ConclusionsThe eukaryotic expression vector of pSect2/S-ScFv4F10is successfully constructed,and the recombinant protein of S-ScFv4 F10 has a fairly high affinity with CTLA-4 antigen.
