1.Colposcopy image database design and application of retrieval algorithm.

Due to the insufficiency in colposcopy image database collection in china, a novel image database is developed and it will be the basis of a computer-aided-diagnosis system for colposcopy. An improved search algorithm of color is designed based on the current content-based image retrieval algorithms for the characteristics of colposcopy images. This algorithm is authenticated by more than one hundred of clinical pictures with primary satisfactory result.

2.Research on stomach remaining-floating sustained-release preparation of compound demethylcantharidin

AIM:To make compound demethycantharidin(including Radix astragali and Rhizoma bletillae) into stomach remaining-floating sustained-release preparation. METHODS:Radix astragal and Rhizoma betillae pulverized to superfine powders were combined with hydrophilic gel to manufacture stomach remaining-floating sustained-release tablet(floating tablet). RESULTS:The tablet started to drift in 3 min in the artificial gastric juice and float lasted for more than 8 h. By isotopic tracering,the floating tablets given on an empty stomach were retarded for 2-3 h and on large meal for 4-5 h in combination with ordinary tablet only retarded for 1 h. A pharmacopoeia dissolution method was set up to determine the release. 50% of tablets were disintegration in 5 h,70% in 8 h. The t 1/2 was 5.1 h(2-4 h for ordinary tablets),and the MRT was 8.3 h(3.3 h for ordinary tablets),and the remaining-floating sustained-release tablets showed a relative bioavailability of 230.71%. CONCLUSION:Pretreatment of raw herbs from compound demethylcantharidin and selection of the excipients will help to produce the herb-containing floating tablet.

4.Effects of Chitosan Flocculation Clarification Process and Alcohol Precipitation Process on Water Extract of Codonopsis Radix

Objective To explore the effects of chitosan flocculation clarification process and alcohol precipitation process on the principal chemical constituents of water extract of Codonopsis Radix. Methods The influence of two purification processes on water extract of Codonopsis Radix was investigated through lobetyolin contents, extract yield, and relative apparent content of each component in HPLC fingerprint as evaluation indexes. Results Chitosan flocculation clarification process showed a significantly higher extract yield of water extract compared with alcohol precipitation process, and it has a markedly better retention effect for strong polarity constituents; alcohol precipitation process exhibit a little better retention effect for lobetyolin and a better retention effect for weak polarity constituents. Conclusion The above two processes have some regularity in the influence on the main chemical constituents in the water extract of Codonopsis Radix, which can provide some guidance for the reasonable choice of the purification process for water extracts of Codonopsis Radix, and other TCM water extracts.
5.Establishment of 43-plex SNP Typing System and Its Forensic Application

Objective To evaluate the application of 43-plex SNP typing system in forensic science. Methods The typing of 43 SNP loci in 123 unrelated Han individuals from East China was detected by MALDI-TOF-MS. The application value of 43-plex SNP typing system was assessed according to the foren-sic parameters of population genetics. Results All the 43 SNP loci of 123 individuals showed no signifi-cant departure from Hardy-Weinberg equilibrium (P>0.05). Excepted rs1355366, rs2270529, rs10776839 and rs938283, there were 39 SNP loci had minor allele frequencies (MAF), which were greater than 0.25. Among the 25 loci MAFs, 24 ranged from 0.4 to 0.5, while 3 were close to 0.4. The DP, CDP, PIC, Ho, PEtrio and PEduo of the 43 SNP loci were 0.2901-0.6544, 1-9.8×10-11, 0.1708-0.5000, 0.1557-0.5935, 0.0854-0.2500 and 0.0146-0.1250, respectively. The CPEtrio and CPEduo were 0.999986 and 0.9924361, respectively. Conclusion The 43-plex SNP typing system in present study shows a high polymorphism, which can be an effective supplement and verification for traditional STR genetic markers. It also can be used with other commercial kits for the forensic paternity testing and individual identification.
6.Effects of low concentrations of di-(2-ethylhexyl) and mono-(2-ethylhexyl) phthalate on steroidogenesis pathways and apoptosis in the murine leydig tumor cell line MLTC-1.

The aim of this study was to evaluate the effects of low concentrations of DEHP and MEHP on steroidogenesis in a murine Leydig tumor cell line (MLTC-1) in vitro. The result of flow cytometry analysis revealed that the proportion of apoptotic cells was significantly increased after the exposure to DEHP. All three genes (P450scc, P450c17, and 3βHSD) under study showed an increased expression following exposure to DEHP or MEHP, although some insignificant inhibitory effects appeared in the 10 μmol/L treatment group as compared with the controls. It was also found that compared with the controls. It was also found that DEHP or MEHP stimulated INSL3 mRNA and protein especially in the 0.001 μmol/L treatment group. Testosterone secretions were stimulated after the exposure to DEHP or MEHP. Alternations of steroidogenic enzymes and INSL3 in MLTC-1 cells might be involved in the biphasic effects of DEHP/MEHP on androgen production.

7.Influence of Shenqing Recipe on morphology and quantity of colonic interstitial cells of Cajal in trinitrobenzene sulfonic acid induced rat colitis.

OBJECTIVETo observe the influence of Shenqing Recipe (SQR), a kind of Traditional Chinese Medicine, on the morphology and quantity of colonic interstitial cells of Cajal (ICC) in trinitrobenzene sulfonic acid (TNBS)-induced rat colitis, and to investigate the possible mechanism of SQR in regulating intestinal dynamics.
METHODSSixty rats were randomly divided into normal control, model 1, model 2, mesalazine, and high-dose, and low-dose SQR groups with 10 rats in each group. TNBS (10 mg) dissolved in 50% ethanol was instilled into the lumen of the rat colon of the latter five groups to induce colitis. On the 4th day after administration of TNBS, each treatment group was administered one of the following formulations by enteroclysis gavage once a day for 7 days: 600 mg•kg⁻¹•d⁻¹ mesalazine, 2.4 g•kg⁻¹•d⁻¹ SQR, and 1.2 g•kg⁻¹•d⁻¹ SQR. Model 2 rats received normal saline solution. After 7 days colonic samples were collected. While the colonic samples of model 1 group were collected on the 3rd day after TNBS administered. Ultrastructure of ICC in the damaged colonic tissues was observed with transmission electron microscope. Expression of c-kit protein in colonic tissue was determined by immunohistochemical staining and Western blot.
RESULTSThe ultrastructure of colonic ICC in the rat model of TNBS-induced colitis showed a severe injury, and administration of SQR or mesalazine reduced the severity of injury. Similarly, the expression of c-kit protein of TNBS-induced colitis rat model was significantly decreased compared with the normal control group (P < 0.05). Treatment with SQR or mesalazine significantly increased the expression of c-kit protein compared with the administration of control formulations (P < 0.05), especially the high-dose SQR group.
CONCLUSIONSQR could alleviate and repair the injured ICC, and improve its quantity, which might be involved in regulating intestinal motility.

8.Clinical effect of rivaroxaban and enoxaparin on the prevention of lower deep vein thrombosis after artificial knee joint replacement

Objective To investigate the clinical effect of rivaroxaban and enoxaparin on the prevention of lower deep vein thrombosis after arti-ficial knee joint replacement.Methods A total of 120 patients who ex-perienced first artificial knee replacement surgery were divided into two groups.And 12 h after surgery , 60 patients in treatment group were gi-ven oral rivaroxaban 10 mg · d -1 , 60 patients in control group received subcutaneous enoxaparin heparin 4000 IU · d-1.The treatment of two groups all lasted for 14 d.The incidence of deep vein thrombosis , bleed-ing, complications and long -term efficacy of patients between groups were compared.Results There were no statistical difference between treatment group and control group in terms of activated partial thrombo-plastin time ( APTT ) , blood platelets count ( PLT ) and D -dimer ( all P>0.05 ) .The incidence rate of lower deep vein thrombosis in treatment group was 11.67%( 7/60 ) and control group 15.00%( 9/60 ) , they had no significant difference ( P>0.05 ).There were no statistical difference between two groups in postoperative drainage , total amount of blood transfusion and hemorrhagic index ( all P>0.05 ).The incidence rate of petechiae in treatment group was 23.33%(14/60), and 18.33(11/60) in control group (P>0.05).There were no sta-tistical difference between two groups in visual analogue scale ( VAS) pain scores and swollen knee at first three days and seven days after operation (all P>0.05).There were no statistical difference between two groups in knee mobility and keen society score ( KSS) function scores during the follow-up half a year after operation (all P>0.05).The knee mobility and KSS function scores during the follow-up one year after operation in treatment group were higher than that in control group ( all P <0.05 ).Conclusion Rivaroxaban and enoxaparin had equivalent efficacy on the prevention of venous thrombosis after artificial knee joint replacement surgery.However, the long-term clinical effect of rivaroxaban was better .
9.Pharmacokinetics of cyclosporine in type 1 diabetic rat model

Objective To investigate the pharmacokinetics of cyclosporine(CsA) in type 1 diabetes of rats.Methods Rats with type 1 diabetes were induced by intraperitoneal administration of 65 mg· kg-1 streptozotocin (STZ).Pharmacokinetics of CsA (10 mg· kg-1) was compared between diabetic and normal rats on the 5 weeks after STZ injection.Concentration of CsA in whole blood was measured by enzyme multiplied immunoassay technique.Results Type 1 diabetes rats were successfully induced as the fasting blood glucose levels exceeded 11.1 mmol · L-1 after STZ injection for 1 week.Beside glucose,total cholesterol (TC) and triacylglycerol (TG) in serum of diabetic rats were significantly higher than those in normal rats after STZ injection for 5 weeks.Unlike normal rats,the pharmacokinetics of CsA in diabetic rats exhibited a double peak after oral administration.The Tmax and Cmax were lower in diabetic rats than normal rats,although without statistical differences as follows:Tmaxwas (3.3±1.6),(3.2 ±2.5) h,Cmax was (579.0±208.5),(453.0 ± 104.8) ng· mL-1 in normal and diabetic rats.Compared with normal rats,AUC and t1/2 of CsA in diabetic rats decreased significantly,which were 0.51 and 0.70 times those of normal rats,respectively;AUC was (7343.2 ± 2333.7),(3729.7 ± 1106.6) h ·ng · mL-1,t1/2 was (8.5 ± 1.5),(6.0 ± 0.9) h in normal and diabetic rats.Conclusion The oral pharmacokinetics of CsA was significantly different in type 1 diabetic rat model compared to normal controls,which should be taken into consideration in clinical medication.
10.Dynamic investigation on the co-infection status of two pathogens in ticks from tourist point in Heilongjiang province

Objective To monitor the co-infection status of Borrelia burgdorferi sensu lato (R.b.s.1) and spotted fever group Rickettsia (SFGR) in tourist areas of Heilongjiang province.Methods Polymerase chain reaction (PCR) was used to detect the 5S-23S rRNA intergenic spacer of B.b.s.1 and ompA of SFGR in ticks,dynamically collected from tourist areas of Heilongjiang province in 2010.Amplification products from positive ticks were sequenced,and phylogenetic analysis was conducted by Mega 5.0 software package.Results 849 ticks were collected from two tourist points,with the dominant ticks in Tiger Mountain and Jingpo Lake were Ixodes persulcatus and Haemaphysalis concinna.Regarding the Ixodes persulcatus from Tiger Mountain,the infection rates of B.b.s.1 and SFGR were 26.15% and 10.05%.The infection rate of SFGR was 13.33% in Haemaphysalis concinna and the B.b.s.1 was tndiscovered in the same ticks from Jingpo Lake.However,the co-infection could only be detected in Ixodes persulcatus of both tourist areas.Surveillance data showed that the major ticks were more likely to be appeared in July at Tiger Mountain and in June at Jingpo Lake.Data from the sequence analysis on B.b.s.1 showed that the B.b.s.1 in tourist areas could be classified into three different genotypes,other than B.garinii and B.afzelii.We first detected B.valaisiana-like group genotype in northeast of China.Results from the sequence analysis of SFGR positive products showed that the two DNA sequences of newly detected agents were completely the same as Rickettsia sp.HL-93 which was detected in Hulin and Rickettsia sp.H820 found in northeast,China.Conclusion The co-infection of B.b.s.1 and SFGR was detected in ticks from the tourist areas of Heilongjiang province,and data from the sequencing of specific fragment showed that various kinds of genotypes existed in this area.However; the rates of co-infectionitis-different according to environment,time and population that contributed to the kinds of and the index of ticks existed in the surveys points,also the infection rate of the ticks was studied.
