4.Novel nerve cell scaffold materials for repair of spinal cord injury Current appfications and future prospects

At present, the research regarding repair of spinal cord mainly focuses on tissue engineering. Neural tissue engineering materials provide three-dimensional template for tissue regeneration and also environment for synthesis of extracellular matrix. This paper summarizes the types of nerve transplant materials and the research progress in application for treatment of spinal cord injury, so as to provide theoretical evidence for repair of spinal cord injury. But some problems exist in application of nerve cell scaffold materials for repair of spinal cord injury: poor mechanical properties lead to slow degradation speed, causing difficulties in tissue reconstruction with respect to velocity and in subsequent reconstruction of porous three-dimensional scaffold. In recent years, novel biomaterials with specific repair function have been made by the engineering method through combining the biological molecule with specific signal identification function and available materials, which is an advanced projeot in the current field of biomaterials.
5.Effect of mild hypothermia on the expression of hypoxia-inducible factor-1a and glucose transporter-1 in a rat model of chronic cerebral ischemia-reperfusion

Objective To investigate the effect of mild hypothermia on the expression of hypoxia-inducible factor-1α (HIF-1α) and glucose transporter-1 (GLUT-1) in a rat model of chronic cerebral ischemia-reperfusion.Methods Thirty-six female SD rats weighing 170-210 g were randomly divided into 3 groups (n = 12 each):sham operation group (group S), normal body temperature group (group NT ) and mild hypothermia group (group MH). Arterio-venous fistula was established by end-to-end anastomosis between the right common carotid artery and right external jugular vein for 6 weeks followed by reperfusion. In group MH, mild hypothermia was induced at the initiation of reperfusion and the rectal temperature was reduced to 31.5-32.5 ℃. In group S and NT, the rectal temperature was maintained at 37-38 ℃. Six rats in each group were sacrificed at 3 and 48 h of reperfusion. The brains were immediately removed for determination of the expression of HIF-1α, GLUT-1, HIF-1α mRNA and GLUT-1 mRNA and microscopic examination. Results Compared with group S, the expression of HIF-1α and HIF-1α mRNA at 3 and 48 h of reperfusion and GLUT-1 mRNA at 3 h of reperfusion was up-regulated, while the expression of GLUT-1 and GLUT-1 mRNA at 48 h of reperfusion was down-regulated in group NT (P < 0.05).Compared with group NT, the expression of HIF-1α and HIF-1α mRNA at 48 h of reperfusion and HIF-1α mRNA and GLUT-1 mRNA at 3 h of reperfusion was down-regulated, while the expression of GLUT-1 and GLUT-1 mRNA at 48 h of reperfusion was up-regulated in group MH (P < 0.05).Microscopic examination showed that the injury to the ultrastructure of blood-brain barrier was significantly attenuated in group MH compared with group NT. Conclusion Mild hypothermia can attenuate chronic ischemia-reperfusion injury by down-regulating the expression of HIF-1α and up-regulating the expression of GLUT-1.
7.Inhibitory effect of taurine in hypoxia-induced rat pulmonary artery smooth muscle cell proliferation and signal transduction mechanism.

OBJECTIVETo discuss the effect of taurine (Tau) on the proliferation of hypoxia-induced pulmonary artery smooth muscle cells (PASMCs), and study whether the extracellular signal-regulated kinase 1/2 (ERK1/2) signal pathway participated in the Tau-inhibited PASMC proliferation process and the possible molecular mechanism.
METHODThe primary culture was performed for PASMCs in rats. The second to fifth generations were adopted for the experiment. The Tau concentration was 80 mmol x L(-1). The concentration of ERK1/2 blocker (PD98059) was 50 micromol x L(-1). The drug administration time was 24 h. The effect of Tau on the PASMC proliferation was detected by MTT assay, immunofluorescence staining method and western blot under different conditions. The PASMCs were growing were divided into four groups: the normoxia group, the normoxia + Tau group, the hypoxia group and the hypoxia + Tau group. The Western blot was adopted to detect whether the ERK1/2 signal pathway participated in the Tau-inhibited PASMC proliferation process. Subsequently, the PASMCs were divided into five groups: the normoxia group, the hypoxia group, the hypoxia + Tau group, the hypoxia + Tau + PD98059 group and the hypoxia + PD98059 group.
RESULTHypoxia could induce the PASMC proliferation. Under the conditions of normoxia, Tau had no effect on the PASMC proliferation. Under the conditions of normoxia and hypoxia, Tau had no effect on the expression of the tumor necrosis factor-alpha (TNF-alpha) among PASMCs. Tau could reverse the expression up-regulation of hypoxia-induced proliferative cell nuclear antigen (PCNA) (P < 0.01) and Cyclin A (Cyclin A) (P < 0. 05). Under the conditions of normoxia, Tau had no effect on the expression of phosphoryl extracellular signal-regulated kinase 1/2 (p-ERK1/2). Hypoxia could up-regulate the p-ERK1/2 expression (P < 0.01). Tau could reverse the up-regulation of the hypoxia-induced p-ERK1/2 expression(P < 0.01). Both PD98059 and Tau could inhibit the up-regulated expressions of PCNA, Cyclin A and p-ERK1/2. According to the comparison between the single addition of Tau and PD98059 under conditions of hypoxia, the hypoxia + Tau + PD98059 group showed more significant down-regulation in the expressions of PCNA, Cyclin A and p-ERK1/2.
CONCLUSIONTau could inhibit the hypoxia-induced PASMC proliferation, and may regulate it through ERK1/2 pathway.

8.Study on Evaluation of the Scientific Research Performance of Provincial Key Disciplines in a Medical University

Objective This article was aimed to take exploratory evaluation on the scientific research performance of provincial key disciplines in a medical university,and to provide a scientific basis for the discipline evaluation related topics.Methods This paper took 29 provincial key disciplines in a medical university as the evaluation system.We calculated the scientific research performance of key disciplines.Data envelopment analysis (DEA) method was used to evaluate the scientific level of performance,while the use of TOPSIS and gray correlation analysis method to rank the efficiency of its research,and the results obtained by the three methods was used for statistical correlation analysis.Results According to the results calculated by DEA Method,there were 18 DEA efficient units and 11 non-DEA efficient units.While in this 11 non-DEA efficient units,8 of them needed to reduce re source inputs and the other 3 units needed to increase research outputs.Meanwhile there were significant positive correlations between evaluation results of TOPSIS Method andGrey Correlation Analysis Method with Data Envelopment Analysis Method,respectively [the correlation coefficient:rs 1 =0.797(P =0.000); rs 2=0.583(P =0.007); rs 3-0.536(P =0.003)].Conclusions Compared to TOPSIS Method and Grey Correlation Analysis Method,DEA Method not only could evaluate the ef fectiveness of discipline,but also provide evidence to improve the non-DEA efficient units.
9.Investigation and Development of Membrane Oxygenator

Based on the advantage of artificial lung, the background, principle and materials of membrane oxygenator are descried briefly. Besides, the purpose of ECC, ECMO, IVOX and TAL, advantage and disadvantage, newest re-search progress of the oxygenator are introduced. The aspects of IVOX and TAL can be modified, and the significance of TAL as to be the real artificial lung is pointed out. By anticipating the development trend of the oxygenator, the feature of ideal artificial lung and the study in the future can be summarized.
10. Potential mechanism of Huanglian Jiedu Decoction for treatment of hypertension based on network pharmacology

Objective To study the mechanism of Huanglian Jiedu Decoction (HJD) in the treatment of hypertension by using network pharmacology. Methods Chemical components and targets related to the four herbs of Chinese meteria medica in HJD were searched through the traditional Chinese medicine systems pharmacology database and analysis platform (TCMSP) to construct the interaction network diagram of the target point of the compounds. The hypertension related targets were screened through OMIM, TTD, and PharmGkb databases; The interactive target of HJD and hypertension was constructed in PPI database; GO analysis of screened core targets between drug target and disease target was carried out using Davidv 6.8 database, and the related pathways of core targets were enriched using KEGG database. Results Using the oral bioavailability (OB) ≥ 30% and drug likeness (DL) ≥ 0.18 as screening condition for the compounds, combined with supplement literatures searched, 61 active components and 154 corresponding protein targets of HJD were screened out. A total of 118 enrichment results were obtained from 228 core targets of HJD in hypertension, including 59 biological processes, 31 molecular functions, and 28 cell compositions. The selected targets were enriched by KEGG database and 45 pathways playing an important role in hypertension were screen out. Conclusion The synergetic effect of HJD with multi-components and multi-pathway was confirmed by network pharmacology, and the main possible mechanism of HJD in treating hypertension was predicted, which lay a foundation for the identification of effective components, the mechanism of action, and clinical application.
