1.Effects of cornel iridoid glycoside on inflammatory reaction in the brain of traumatic brain injury rat model

AIM:To observe the effects of cornus iridoid glycosides(CIG)on inflammatory reaction especially the inflammatory cytokines in the brain after traumatic brain injury,and to explore the possible mechanisms of its neuroprotective effect.METHODS:SD rats were intragastrically administered with different doses of CIG(30,60 and 120 mg?kg-1?d-1)for 7 d.The traumatic brain injury rat model was induced by improved Feeney's fall weight method,and the brains were taken out 24 h and 72 h after brain injury,respectively.The morphological changes were observed by HE staining in the cerebral cortex.The expressions of inflammatory cytokine tumor necrosis factor-?(TNF-?)and interleukin-1?(IL-1?)were detected by immunohistochemical method.The image processing and statistical analysis were used to measure the number and the area of immunoreactive cells.RESULTS:HE staining showed the pathological changes were serious in the cerebral cortex of model group,and compared with the model group,the pathological changes were obviously reduced in CIG group.The positive immunoreactive cells of TNF-? and IL-1? were mainly distributed around the foci of contusion,the expressions of TNF-? and IL-1? in the model group were significantly higher than those in sham operated group,and the high expressions were sustained from 24 h to 72 h after brain injury.Compared with the model group,the levels of TNF-? and IL-1? in the brain of CIG treatment groups were obviously decreased in a dose-dependent manner and the inhibitory effects of TNF-? and IL-1? were more significant at 72 h after brain injury.CONCLUSION:CIG may have neuroprotective effect on traumatic brain injury through inhibiting the expression of inflammatory cytokines and reducing the inflammatory reaction.

The rupture of vulnerable carotid atherosclemtic plaques and distal embolization of atherornatous debris are the most important pathogenesis of atherosclerotic stroke.The serolog- ic(and plasma)markers associated with vulnerable plaques will change significantly in this process,and show their different meanings.The studies in this field are in its infancy and still need to be further validated,This article reviews the advances in research on the correlation of these biomakers with vulnerable plaque and ischemic cerebrovascular disease,and briefly intro- duces the clinical application prospect of some of the biomakers.
4.Application of sub-Tenon's anesthesia for compound trabeculectomy with high intraocular pressure

AIM:To evaluate the efficacy and safety of Sub-tenon's anesthesia for compound trabeculectomy with high intraocular pressure.
METHODS: Forty - six eyes ( 46 cases ) of primary glaucoma received compound trabeculectomy under Sub-tenon's anesthesia, whose preoperative intraocular pressure were higher than normal after 24 to 48h of combined medication. Both efficacy and complication of the anesthesia were studied.
RESULTS: One minute after anesthetic injection, all cases were able to achieve the effect of analgesia and eye brake. During the operation, 0 level of anesthesia effect included 35 eyes ( 76%) , 1 level of anesthesia effect included 10 eyes ( 22%) , 2 level of anesthesia effect included 1 eye(2%). Only 1 case of these patients needed to add the surface anesthetic once, and other cases were successfully operatedunder Sub-tenon's anesthesia. The total effective rate was 98%. No anesthesia complications occurred in all cases.
CONCLUSION: Sub - Tenon's anesthesia is safe, effective, simple and quick for compound trabeculectomy with high intraocular pressure.
5.Effect of compound mylabris capsules combined with neoadjuvant on efficacy and expression of Ki67,ER,PR in patients with HER2-negative breast carcinoma

Objective To investigate effect of compound mylabris capsules combined with neoadjuvant on efficacy and expression of Ki67,ER,PR in patients with HER2-negative breast carcinoma.Methods Eighty-one HER2 negative-breast cancer patients from the hospital were randomly divided into treatment group(41cases) and control group(40 cases) by random number table method.Control group was treated with three weeks of neoadjuvant chemotherapy,firstly given with cyclophosphamide(CTX)600 mg/m2 +adriamycin amycin(Ad)60 mg/m2,and then next course with docetaxel(DOC) 100 mg/m2, intravenous drip for 1 h,one period per three weeks,one times per week,and for 6 periods.Patients in treatment group were additionally given with compound mylabris capsules from one day before chemotherapy,three particles per time,two times per day,seven times per period,and for another one week after chemotherapy.Score of traditional Chinese medicine(TCM) symptoms,clinical efficacy,and levels of hemorheology indexs were compared.Serum levels of Ki67,ER,and PR were detected between both groups.Results Total efficacy of treatment group was recently 56.10%,which was superior to control group 30.00%(P<0.05).AT the ending of chemotherapy and 1 weeks after chemotherapy,TCM score, plasma viscosity,and high, middle and low shear of whole blood were obviously lower than control group(P<0.05).Serum level of Ki67 in treatment group was lower, while ER and PR were higher than control group at the ending of chemotherapy and 1 weeks after chemotherapy with statistically significant difference ( P<0.05).Conclusion Compound mylabris capsules combined with neoadjuvant chemotherapy in treating breast carcinoma with HER2 negative can decrease TCM symptoms, improve blood viscosity and clinical efficacy,and inhibit expression of Ki67 and up-regualte ER and PR levels.

7.Lentivirus mediated RNA interference inhibits mesothelin expression in ovarian cancer cells and cell proliferation

Objective:To construct a recombinant lentivirus plasmid of RNA interference targeting (MSLN) gene and to observe its effect on MSLN expression in human ovarian cancer cell line OVCAR-3 and its effect on cell proliferation. Methods: According to the Genbank information of MSLN, four RNA interfering sequences and a negative sequence were designed and inserted into plasmid pRNAT-U6.2/Lenti and 5 kinds of plasmids were packaged: LV-MSLN-negative,LV-MSLN-shRNA1, LV-MSLN-shRNA2, LV-MSLN-shRNA3, and LV-MSLN-shRNA4; and they were used to transfect OVCAR-3 cells. Western blotting and indirect immunofluorescence were then used to investigate the interfering efficiency. The plasmid with high interfering efficiency was packaged. The cell proliferation test and clone-forming test was used to assess the changes in cell proliferation. Results: DNA sequencing showed that the sequences of 5 recombinant lentivirus plasmids were correct. Lentivirus packaging was successfully done. Western blotting analysis confirmed that LV-MSLN-shRNA4 had the highest interfering efficiency (90%). MSLN specifically bound to cytomembrane of OVCAR-3 cells. Expression of MSLN in the interfered cells (OVC-shRNA) was weaker than that in the control cells (OVC-neg,OVC). BACKGROUND: There are small amount of hematopoietic stem cells, which are easy to divide in vitro and show the difficulty in applying to transplantation.OBJECTIVE: This paper has focused on the role and means of the Wnt, Notch, Bmi_1, Shh, HOXB4 signaling molecule in the maintenance of hematopoietic stem cell self-renewal and regulation. METHODS: With the key words of "HSC, Wnt, Notch, Bmi_1, Shh, HOXB4" for the search, we searched PubMed database (2002-01/2008-12) in English. Literatures closely related to the hematopoietic stem cell self-renewal related signaling molecules were included. Repetitive research and Meta analysis were excluded.RESULTS AND CONCLUSION: The computer initially retrieved 216 documents, of which 30 documents for research. Hematopoietic stem cells are self-renewing, have strong differentiation and growth and regeneration capacities, can produce various types of blood cells ancestor cells, are widely used to treat blood diseases, but the hematopoietic stem cell differentiation in vitro demonstrated that the difficulties used in transplantation. How to make hematopoietic stem cells in vitro amplification and processing, while maintaining hematopoietic stem cell self-renewal characteristics is of a key issue. In recent years, different signaling pathways to enhance the ability of hematopoietic stem cell self-renewal signaling molecule have been a research hotspot. The article focused on the role and means of the Wnt, Notch, Bmi_1, Shh, HOXB4 in the maintenance of hematopoietic stem cell self-renewal and regulation and found that both the above-mentioned five signaling molecules can enhance hematopoietic stem cell self-renewal function. There are also a number of factors playing an important role in the maintenance of hematopoietic stem cell self-renewal process, such as endogenous factors, a series of transcription factors Oct-4, Ehox, Nanog, SCL, Runx1 and so on, to explore how their regulatory networks formed by the interaction control self-renewal of hematopoietic stem cells will become a key point in the research of self-renewal of hematopoietic stem cells.
9.Analysis of ischemic stroke subtypes

Objective To determine the symptomatological characterictics of isehemie stroke in Chinese people on the basis of Oxfordshire Community Stroke Project (OCSP) criteria. Methods 900 patients with ischemic stroke were reviewed. Ischemic stroke was classified into four subtypes. Results Patients were classified as lacunar infarcts(19.3%) ,total anterior circulation infarcts(17.2%) ,partial anterior circulation infarcts (45.3%), and posterior circulation infarcts(18.2%). The statistical comparison between male and female of each subtype is not significant . Conclusion The difference between the studies is caused by race difference and some other reasons.
10.Effect of basic fibroblast growth factor on the formation of blast-colony-forming cell derived from embryonic stem cells

BACKGROUND:Some studies show that basic fibroblast growth factor(bFGF)strongly expresses dudng the process of embryonic stem cells differentiation into hematopoietic stem cell.yolk sac blooding.and fetal liver hematopoiesis.OBJECTIVE:To study the regulation of bFGF on the blast-colony-forming cell(BL-CFC)by adding bFGF in the medium of embryoid body generation.METHODS:The third to fifth generations of the pdmaw mouse embryonic fibroblasts were recovered,and then incubated with the DMEM medium containing mitomvcin C for 2.5 hours in order to lose the proliferative capacity.Then cells were suspended into single cell by trypsinization and inoculated in the gelatin-coated bottle at the density of 10×104/cm2.After culturing for 24 hours,mouse embryonic stem cells(mESC)of D3 were recovered and placed on the feeder layer cells.According to the composition of medium in embryoid body generation.mESCs were divided into two groups:group A:standard medium+VEGF+SCF;group B:standard medium+VEGF+SCF+bFGF.Each group was cultured for 3 days and 6 days respectively,and the cloning number of BL-CFC was quantified,as well as Flk-1+ expression was observed by immunofluorescence staining.Positive number and average absorbance were analyzed using IMAGE-PRO PLUS imaging analysis system.RESULTS AND CONCLUSION:Adding bFGF in the course of embryoid body growth could significantly increase the number of BL-CFC(P<0.01).and the positive results of Flk-1 and the average absorbance were also increased significantly(P<0.01).bFGF effectively promoted embryoid body amplification and proliferation of BL-CFC.
