2.Isolation and Screening of Endophytic Antifungal Bacteria from Mangroves

The endophytic bacteria of the mangroves were studied in this paper. The results show that there are 1.728 (0. 195 -4.225)?104cfu/g (fw) bacterial endophytes in the variety of mangroves, the most population of the endophytic bacteria was found in Rhizophora stylosa, the figure was4. 225?104cfu/g (fw) , the next was Bruguiera gymnorrhiza, Aegiceras corniculatum, Kandelia candel and Avicennia marina. In parts of the mangroves, the contents of the bacteria in stem was the most, the figure was 1. 649?10 cfu/g (fw) , then the root and the leaf. Of the bacteria, about 43. 53% strains expressed the antagonism against the growth of the plant pathogens, such as Fusarium oxyspontm f. sp. cubense , Colletotrichum sp. and Rahtonia solanaceance etc. and these bacteria were identified as Bacillus sp. . The results also showed that 9 of the 13 strains (69. 23% ) could promote the growth of the tomato, while 4 strains (30. 77% ) restrained the tomato's growth.
3.Bond strengths of customized titanium brackets manufactured by selective laser melting.

OBJECTIVETo investigate the bond strengths of customized titanium bracket manufactured by selective laser melting.
METHODSEighty human premolars which had been extracted for orthodontic purpose were collected and divided randomly (by random table) into two groups (customized bracket group and 3M bracket group, 40 molars in each group). The 35% phosphoric acid was used for etching and the brackets were bonded with 3M Unitek bonding adhesive. All bonded specimens were placed in saline for 24 hours at room temperature and were tested on DWD3050 electronic testing machine to determine the shear bond strength and tensile bond strength. After debonding, the adhesive remnant indexes (ARI) were recorded.
RESULTSThe shear bond strengths of customized brackets was 6.80 (6.20, 8.32) MPa, which was significantly lower than that of the 3M brackets [10.46 (9.72, 11.48) MPa] (Z = -3.463, P < 0.05). And the tensile bond strengths of customized brackets was (6.93 ± 1.21) MPa, which was significantly higher than that of the 3M brackets [(5.88 ± 1.23) MPa] (t = 2.81, P < 0.05). No significant difference was found in the ARI between two different kinds of the brackets.
CONCLUSIONSThe shear bond strength and tensile bond strength of both kinds of brackets were enough for clinic application.

4.Application of integrated digital maxillodental model in computer aided design of individualized lingual brackets.

OBJECTIVETo create an integrated digital maxillodental model and to apply it in computer aided design (CAD) of individualized lingual brackets in order to align both crowns and roots without fenestration and dehiscence.
METHODSCone-beam computerized tomography (CBCT)-based maxillodental model and laser-scanned dental model were integrated by auto registration in 10 patients with malocclusions. The registration error was calculated automatically. Three observers tested the method independently. The inter-observer difference was investigated. An integrated model was selected randomly and the setup was created with roots and jaws in good relationship without fenestration and dehiscence. The individualized lingual brackets were designed by CAD on the setup.
RESULTSNo significant difference was found in inter observers (P > 0.05). The registration errors of maxilla and mandible were (0.144 ± 0.020) mm and (0.141 ± 0.022) mm, respectively. The digital individualized lingual brackets based on the virtual treatment in integrated digital maxillodental model were produced.
CONCLUSIONSAn integrated digital maxillodental model was created in good accuracy. By applying the integrated model, individualized lingual brackets were designed.

5.Diagnosis and treatment for complications of renal caliceal diverticulum

Objective To analyze the diagnosis and treatment for complications of renal caliceal diverticulum with calculi or infection. Methods A retrospective investigation was performed on 29 cases with renal caliceal diverticulum. The 29 cases included 11 males and 18 females aged 18 to 61 years. Among the study group, 3 cases were simple renal caliceal diverticulum, 12 cases were diagnosed as diverticular calculi and 14 cases presented recurrent urinary tract infections including 3 cases with urinary fistula after unroofing and decompression as renal simple cyst from another hospital. Ten cases underwent an open operation that unroofed and decompressed the cyst, and sutured the diverticular neck. Eight cases underwent laparoscopic operation similar to the open operation, including lithotomy in caliceal diverticulum in 2 cases. Eleven cases diagnosed with caliceal diverticular calculi were taken one-stage percutaneous nephrolithotomy including dilating the diverticular neck, remaining the nephrostomy catheter and Double-J ureteral stents, and 1 case was transferred to open operation.Results The open and laparoscopic operations were performed successfully. One case was cured by Double-J ureteral stenting after postoperative urinary leakage. One case was transferred to open operation for the failure of percutaneous puncturation. X-ray examination revealed that there were no remaining stones after the operation. All the patients were followed up for 6 to 24 months without calculi and infection recurrence. Conclusions Stones and infection are common that complications of renal caliceal diverticulum. Percutaneous nephrolithotomy, laparoscopy and other operations were effective and feasible treatment options for cases with complications of renal caliceal diverticulum. Exact diagnosis was very important for treatment of renal caliceal diverticulum before operation.
6.Fruit-specific RNAi-mediated Lcy gene enhances content of lycopene in tomatoes silencing.

Tomatoes ( Lycopersicon esculentum Mill.) are the principal dietary source of Lycopene which is one of carotenoid and is highly beneficial in preventing some diseases such as the cancer and the heart disease. Suppressing the expression of Lcy gene, the main gene regulating the transformation of the lycopene, is a convenient and effective way to enhance the content of lycopene. The primers were designed according to the gene sequence(U46919)and (X86452) in GenBank. The fruit-specific promoter--phytoene desaturase gene(Pds) promoter and the DNA segment of the Lcy gene were isolated from the genome DNA of tomatoes. The 3'end of Lcy DNA segment was connected together by an intron to inform the RNA interferential segment then which was inserted in the expression vector with the Pds promoter to inform the fruit-specific expression vector. The vector was transformed into the tomatoes through the Agrobacterium tumefaciens. Five transformants were obtained. And the PCR proved that the extra-gene was integrated into the tomato genome. The lycopene in the transgenic tomatoes fruit was increased significantly through analysing the contents of lycopene. These results show that regutating biosynthetic enzyme in carotenoid pathway by RNAi can improve the lycopene content of plant-derived products.

7.Influence of nickel sulphate on lens in SD rat

Background Researches showed that the incidence rate of cataract is high in the nickel mining area. Nickel sulphate can apparently inhibit the metabolism and proliferation of human lens epithelium cells. But the study on the injury mechanism of nickel on lens is still seldom. Objective Present study was to investigate the effect of nickel sulphate on the lens of SD rats. Methods Forty-five SPF SD rats aged from 7 to 14 days were grouped randomly into subcutaneous injection group, intraperitoneal injection group and blank group. Nickel sulphate of 2 g/L ( 10 mg/kg) was subcutaneously or intraperitonealy injected for 45 days. The opacity of rat lens was examined under the slit lamp at two-week interval and scored based on the criteria of LOCS II and LOCS III. The rats were sacrificed in 45 days after experiment and the lens were obtained for the pathological examination. Result The mean score of the anterior subcapsule opacity of rat lens was obviously higher in subcutaneous injection group compared with blank control group with a significant difference between them (t= 14. 311, P < 0. 05 ) , but no significant difference in the anterior subcapsule opacity between intraperitoneal injection group and blank control group (t = 4. 355 , P>0. 05 ). The score of posterior subcapsule opacity of lens were evidently higher in both subcutaneous injection group and intraperitoneal injection group than the blank control group (t = 9. 316,P = 0. 004;t = 7. 464, P = 0. 009) ,so was the mean score of the anterior +posterior subcapsule opacities(t = 23. 387,P=0. 000;t= 10. 533,P = 0. 002) and the total score of rat lens opacity ( t = 12. 358 , P = 0. 001; t = 10. 188 , P = 0. 003 ) . No significant differences were found in cortex opacity score and nuclear opacity score among three groups ( P > 0.05 ). Histopathology examination revealed that the degeneration of lens collagen protein was more serious in subcutaneous injection group and intraperitoneal injection group than the blank control group,and the injury degree of lens collagen protein was more dominant in subcutaneous injection group. Conclusion System administration of nickel sulphate induced the injury of anterior and posterior subcapsule of lens in SD rat.
8.Effects of sodium fluoride on the morphologic characteristics of primarily cultured thyroid cells of rats

Objective To investigate the effects of different concentrations of sodium fluoride on the morphologic characteristics of primarily cultured thyroid cells of SD rats and in order to obtain important proof for approaehing the mechani8m of thyroid gland damage caused by fluoride.Methods Thyroid cells of SD rat were primarily culture for 96 hours,and cell density was adjusted to 5.0×108/L Cell suspension with 5 ml Wills seeded into 6 weII plates,after 12 hours,0(contr01),10.100,1000 μmol/L of sodium fluoride was added into the well, witll each well representing different level of treatment group.Finally the cultured thyroid cells were collected for morph010gic study.Results Under microscope,the transparency of the control thyroid cells Was good,and cells gathered in cluster and adhered to wall.But a lot of cells treated with fluoride suspended,and lost their transparency-under scaning delectron microscope,the control calls showed integrated membrane and tightness to each other,as well as clear boundary between cells normal proliferation.While the thyroid cells treated with 10,100 μmol/L sodium fluoride 0bviouslv shrinked and deformed,and the cells treated with 1000 μmol/L of sodium fluoride were broken-Conclusions nuoride can affect the growth and development of thyroid cell and damage the structure and morphology.Sodium fluoride affects the morphologie characteristics of thyroid cells in a dose-response manner.
9.Emergency response of infectious snails detected after interruption of schistosomiasis transmission in Hannan District, Wuhan

Abstract: Objective To analyze the emergency response and long-term intervention effects after the detection of infectious snails epidemic by loop-mediated isothermal amplification (LAMP) assays in Hannan District, Wuhan City, and to explore the application of LAMP in early surveillance and early-warning of schistosomiasis transmission. Methods Snails picked up by the risk monitoring system in Hannan District were examined by anatomical microscopy and LAMP technology to identify the schistosomiasis infection. Emergency response and intensive intervention were initiated in the environment where positive snails appeared, and the long-term effects were evaluated. Results In May 2018, the infectious snails were detected by LAMP technology in Hannan District, and the positive snails were located in Zhujiacha, Dongzhuang Village, Obstacles and weeds were removed and buried by machine in Zhujiacha. 12 700 m2 of snails were killed by drugs, and the mortality rate of snails was more than 80%; no new seropositive persons were found in the emergency examination within 500 m of the positive snail sites. 506 people were examined in Dong Zhuang Village at the end of the year, and 30 positive IHA cases were detected with a blood positive rate of 5.93%, no positive fecal test was found, and all positive blood test patients took preventive medication. The monitoring results of sentinel rats and wild feces were all negative. Health education was carried out, 7 warning signs were deployed and refreshed, and 500 publicity brochures were distributed. After nearly three years of intensified intervention and monitoring in the villages where the positive environment is located, and the density of snails on the stubborn snail has dropped from 0.094/frame to 0.027/frame, and the positive rate of blood test in Dongzhuang Village has steadily dropped from 5.93% to 3.74%. Conclusions The infected snails missed by microscopy were detected by LAMP in Hannan District, which created conditions for the rapid emergency treatment of environment and elimination of positive snail and improved the sensitivity of the surveillance and early warning system in transmission-interrupted areas.
10.Determination of chemical constituents of essential oil from the fruit of Eucalyptus globulus by GC-MS.

OBJECTIVEThe chemical components of essential oil from fruit of Eucalyptus globulus were analyzed by GC-MS.
METHODThe essential oil were extracted by steam distillation, then separated by capillary gas chromatography. The amount of the component from essential oil were determined by normalization methods. Chromatographic conditions were: capillary column DB-WAX (0.32 mm x 30 m, 0.25 microm) was used, column temperature: initial temperature at 40 degrees C for 3 min,ramping 5 C x min(-1) to 250 degrees C (holding for 10 min) the detector MS.
RESULT31 component from the fruit of E. globulus were identified, which accounted for over 93.7% of total volatile oil.
CONCLUSIONThe methods is reliable, stabilize and can be applied to identify the volatile oil from the fruit of E. globulus.
